이 연구의 목적은 방사선조사를 받은 MC3T3-E1 골모세포의 분화 과정에서 발현되는 BSP와 OC 유전자의 발현에 대한 2-DG와 QCT의 영향을 구명하는 데 있다. Linear accelerator X선조사기를 이용하여 실온에서 1.5 Gy/min의 선량률로 2, 4, 6, 8 Gy를 단회조사 후 배양하여 7, 14, 21일째에 BSP와 OC 유전자의 발현 양상, 그리고 2-DG와 QCT 처리시의 발현 양상을 관찰하여 다음과 같은 결과를 얻었다.
1. 2 Gy의 방사선조사 후 2-DG와 QCT 처리시에는 제 I형 교원질의 발현량은 유의성 있는 변화가 없었으며, 8 Gy의 방사선조사 후 2-DG 처리시에는 발현량이 유의성 있게 감소하였고, QCT 처리시에는 전반적으로 증가하였다.
2. MC3T3-E1 골모세포 분화시, 배양 14일째에 BSP의 발현량이 최대치를 보였고 OC의 발현량은 21일째 최대치를 나타내었다.
3. 골모세포 분화시 2-DG와 QCT을 처리한 경우 모두 BSP는 전반적으로 발현량이 감소하였고, OC는 7일째에 발현량이 증가하였다가 14일과 21일째에는 감소하였다.
4. 2, 4, 8 Gy의 선량에서 BSP와 OC 모두 배양 7일째에는 감소하였으며, 특히 4, 8 Gy에서 크게 감소하였다.
5. 2, 4, 6, 8 Gy 선량에서 배양 7, 14, 21일째 2-DG와 QCT 처리시 BSP와 OC의 발현량은 배양 초기에 크게 증가하였고, 21일째에는 감소하였다.
이상의 결과로부터 2-DG는 저선량의 방사선조사시 배양초기에 BSP와 OC의 발현량을 증가시켰으나 고선량의 방사선조사와 7일 이상의 배양시에는 감소시켰는데, 이러한 경향은 방사선조사에 대한 2-DG의 촉진효과에 의한 일시적 경향이었으며, 장기적으로는 방사선효과를 증가시키는 것으로 볼 수 있다. 반면 QCT는 방사선효과를 억제하거나 대조군과 유사한 수준으로 변화를 억제하여 방사선조사에 의한 골분화를 더욱 억제시킬 수 있을 것으로 사료된다.

Purpose : To investigate the effects of 2-deoxy-D-glucose (2-DG) and quercetin (QCT) on gene expression of bone sialoprotein (BSP) and osteocalcin (OC) during the differentiation in irradiated MC3T3-E1 osteoblastic cells.
Materials and Methods : When MC3T3-E1 osteoblastic cells had reached 70-80% confluence, cultures were transferred to a differentiating medium supplemented with 5 mM 2-DG or 10 μM QCT, and then irradiated with 2, 4, 6, and 8 Gy. At various times after irradiation, the cells were analyzed for the synthesis of type I collagen, and expression of bone sialoprotein and osteocalcin.
Results : The synthesis of type I collagen in cells exposed to 2 Gy of radiation in the presence of 2-DG or QCT showed no significant difference compared with the control group within 15 days post-irradiation. When the cells were irradiated with 8 Gy, 2-DG facilitated the irradiation mediated decrease of type I collagen synthesis, whereas such decrease was inhibited by treating with QCT. During MC3T3-E1 osteoblastic cell differentiation, the mRNA expression of BSP and OC showed the peak value at 14 days and 21 days, respectively. 2-DG or QCT treatment alone decreased the level of BSP mRNA, but increased the OC mRNA level only at early time of differentiation (day 7). In the cells irradiated with 2, 4, 8 Gy, the mRNA expression of BSP and OC decreased at 7 days after the irradiation. The cells were treated with various dose of radiation in the presence of 2-DG or QCT, the mRNA level of both BSP and OC increased although this increase was observed at low dose of radiation (2Gy) and at the early stage of differentiation. However, when the cells were exposed to 4, 6, or 8 Gy, the increase of BSP and OC mRNAs was detected only in cells co-incubated with QCT.
Conclusion : This study demonstrates that 2-DG and QCT affect differently the expression of bone formation related factors, type Ⅰcollagen, BSP, and OC in the irradiated MC3T3-E1 osteoblasic cells, according to the dose of radiation and the times of differentiation. Overall, the present findings suggest that 2-DG and QCT could have the regulatory roles as radiation-sensitizer and -protector, respectively.
