The purpose of the present study was investigate the role of MyD88 on the expression level of NF-κB and cytokines
in hindlimb muscles of female mice with a single bout of acute exercise.
Eight balb/c(wild type) female mice was randomly assigned to experimental group or control group. And eight MyD88 knockout female mice was also randomly assigned to experimental group or control group.
The exercise protocol consists of a single bout of high-intensity treadmill exercise. (inclination 10˚, speed 17cm/sec 10min, 33cm/sec 10min, 50cm/sec). Mice was killed 1-2 hours after the exhaustion protocol.
The level of mRNA expression in RelB, adjusting a role of MyD88 on plantaris muscle in MyD88 KO mice with a single bout of aerobic exercise, was significantly increased, but it is also unable to detect any changes in soleus muscle. Additionally, the level of expressions of TNF-α mRNA was significantly suppressed in WT soleus muscle compared with control. It also reveals that there is a significant difference in TNF-α mRNA basal level between WT control and MyD88 KO control.
However there is no significant effect of a single bout of treadmill exercise on the expression level of NF-κB subunit, P50, P52, P65, RelB mRNA in both soleus and plantaris muscle, except RelB in MyD88 KO mice. And there is no significant change of a single bout of treadmill exercise on the expression level of IL-6 in both soleus and plantaris.
In conclusion, NF-κB and cytokines gene expression are differently requested by exercise based on type, intensity
and duration.
