DAN content analysis of formalin fixed paraffin-embedded tissue permits determination of the influence of DNA content on the prognosis in cancer patients. Of key importance in such analysis is the accuracy of DNA content determination. Digestion with enzymes playsand important role in obtaining good results in cellular DNA content measurements from paraffin-embedded tissue using flow cytometer. We have compared three groups of DNA histograms obtained from fresh tumor tissue, paraffin embedded same tumor tissue digestedpepsin alone(Hedley method) and proteinase K digestion with each other. Concerning the coefficient of variation(CV), the fresh tissues produced the lowest CV and that of proteinase K method was lower than that of the pepsin digestion alone method. However,the proteinase K digestion method had important drawbacks such as loosing aneuploid fraction due to overdigestion. The mean CV of the pepsin digestion alone method was 4.92. Although the CV s of the pepsin digestion method using paraffin embedded tissue were a little higher than that of fresh tissue, the paraffin blocks could be the best samples to analyse DNA content of tumors, because we can use the tumor tissue with minimum contamination of normal tissue.
