This study was carried out to investigate the effects of plant growth regulators, optimal development stage of anthers on microspore derived callus induction and plantlet regeneration of Digitalis purpurea L. From the tetrad to late microspore stage of anthers of Digitalis purpurea L. were cultured on Murashige and Skoog``s medium supplemented with various combination of auxins and cytokinin. Although high frequency of embryogenic callus were induced on MS medium with 1.0 mg·L-1 2,4-D and 1.0 mg·L-1 BA (95.9%) or 0.5 mg·L-1 NAA and 1.0 mg·L-1 BA (45.8%), these callus could not produce normal plant. The combination of 3.0 mg·L-1 IBA and 3.0 mg·L-1 BA was formed low quantitative callus (34.1%), however, 65.9% of induced callus did pollen embryogenesis and plant regeneration. The most of suitable stage of anther culture for the callus induction was 20 days before anthesis at the tetrad or early microspore stage before first pollen mitosis. Three distinct pathways could be recognized in the androgenesis, one involving mainly the vegetative cell, the second starting with the vegetative and the generative cell, respectively, and the third accompanying with two equal vegetative cells in the pollen grains.
