Identifications of the regulatory mechanism for Corpus Lutea have been founded by using expressed sequence tags ( ESTs ) that characterized for the subtracted cDNA library on 5 days of estrus cycle. The present study was conducted to obtain the whole sequence of the ESTs gene and characterize the expressing its mRNA in bovine corpus lutea.
1. The analysis result of the tissue from corpus lutea start to degradation had a low cytosol and aggregation of the nuclear. During the estrus cycle, the cytosol was magnified and differentiation of the nuclear was clearly manifested. At this time, we could found the cell death and formation the blood vessel.
2. By the result of BLAST analysis for the EST genes in 5-Day of corpus lutea. One of the genes was checked the Bovine Microtubule gene.
3. Bovine coupus lutea at various stages was used in the Hematoxylin & Eosin, SQ-PCR, Real Time- PCR, Southern Blot Hybridization. And, we also analyzed the expression pattern of relationship between Microtubule and VEGF ( growth factor ), TNFR1 ( cell death ). It was the same expressing pattern between Microtubule and VEGF. Thus, we thought that Microtubule' appearance could be effect to the expressing pattern of VEGF.
4. The expressing result of TNFR1 was very low during the activation of Microtubule and VEGF. However, it' expression gradually increased in the degradation of CL2~CL1.
Taking together, we summarized that Microtubule affect the cell and then relationship of the VEGF and TNFR1 play a pivotal role for the cell nutrition and activation depending on the stage of corpus lutea in bovine ovary.
