고장초 메탄올 추출물의 항알레르기 활성을 조사하기 위해, 비만세포주인 RBL-2H3 세포를 이용하였으며, 고장초 추출물을 처리한 후 IgE-항체, A23187 및 PMA를 이용하여 세포를 활성화시켜 분비되는 β-hexosaminidase, TNF-α의 양을 측정하였다. 그 결과 고장초 추출물이 활성화된 비만세포에서 분비되는 β-hexosaminidase, TNF-α를 농도 의존적으로 억제하는 것을 확인할 수 있었다. 또한, 비만세포에 탈과립이 발생할 경우 나타나는 형태학적 변화에 대한 시료의 영향을 검색하기 위해 고장초 추출물을 처리한 세포에 compound 48/80을 처리하였으며, 그 결과 고장초 추출물을 처리한 경우 대조군에 비해 탈과립이 현저히 억제되는 것을 관찰하였다. 따라서 고장초 추출물은 다양한 자극으로 인한 비만세포의 탈과립 저해를 통해 알레르기 반응을 억제시키는 것으로 판단되며, 본 연구결과는 고장초의 항알레르기 기능성 소재로의 이용 가능성을 시사해준다.

The anti-allergic effect of the methanol extract of Zizania latifolia Turcz was investigated. Methanol extract of Z. latifolia Trucz did not display any cytotoxicity activities in the IgE-sensitized RBL-2H3 cells. The extract was found to inhibit compound 48/80-induced degranulation and antigen-induced β-hexosaminidase release as well as PMA plus A23187-induced TNF-α production in RBL-2H3 mast cells in a dose-dependent manner. Release of β-hexosaminidase, a marker for the release of histamine in mast cells, was inhibited with an IC50 value of 73.8 μg/mL. These results suggest that Z. latifolia extracts may be useful for the prevention of type I allergic reaction.
