돼지 바이러스성 병원체는 감염된 돼지에 의한 직접적인 전파양식 또는 다양한 매개체들을 통한 간접적인 전파 양식을 통해 전파되어진다. 야생 설치류와 고양이는 porcine circovirus type 2 (PCV2), encephalomyocarditis virus (EMCV), rotavirus (RV), pseudorabies virus (PRV)와 같은 돼지 바이러스성 병원체의 보균자나 전파자의 위치를 차지한다. PCV2는 설치류감염과 바이러스증식성이 실험적으로 증명되었고, 임신된 마우스에 감염되었을 때 태아로 수직감염이 가능하며, 돼지에서 PMWS의 병변과 유사하게 마우스에서 병변을 일으키는 역할을 하는 것으로 알려져 있다. EMCV는 일반적으로 설치류의 바이러스로서 간주되어지고 있으며, 설치류가 질병 전파의 매개체로 보고되어 있고, 설치류의 사체나 분변에서 바이러스가 분리되었다. 또한 RV는 설치류에 의해, PRV는 고양이나 너구리에 의한 전파의 가능성이 있다. 이러한 전파 양식에 대한 국내의 상황을 파악하고 증명하고자 연구를 수행하였다. 본 연구의 목적은 국내에서 발생하고 있는 주요한 돼지 바이러스성 병원체를 야생 설치류와 고양이에서 검출 확인을 통해 최종적으로 전파의 가능성을 확인하고자 함이다. 연구를 수행하기 위해 12개 양돈장을 선정하였으며, 이들 농장에서 랫드 98마리, 마우스 4마리, 고양이 24마리를 포획하였다. 부검을 실시하여 뇌, 편도, 폐, 심장, 간, 비장, 신장, 소장, 대장, 장간막림프절의 장기 시료를 채취하였다. 조직시료에서 돼지질병을 유발하는 대표적인 바이러스인 PCV2, PEDV, TGEV, RV, EMCV, PRRSV, PPV, PRV, JEV 및 CSFV를 PCR과 RT-PCR법을 이용하여 검출하였고, 랫드에서 분리된 PCV2 염기서열 분석을 통해 기존의 분리주와 비교분석하였다. 랫드에서 PCV2는 13두/98두, EMCV는 1두/98두로 나타났고, 마우스에서 PCV2는 2두/4두, EMCV는 3두/4두로 나타났으며, 고양이에서 PEDV는 1두/24두로 나타났다. 야생 설치류와 고양이에서의 총 검출은 PCV2 15두/126두, PEDV 1두/126두, EMCV 4두/126두로 나타났다. PCV2의 염기서열에 의한 상동성 분석 결과는 한국분리주, 캐나다분리주, 중국분리주, 프랑스분리주와 98.7~99.5%의 상동성을 보였으며, 아미노산서열에 의한 상동성 분석 결과 한국분리주, 캐나다분리주, 중국분리주, 프랑스분리주와 96.0~98.4%의 상동성을 보였다. 본 연구를 통해 돼지질병의 원인체인 PCV2와 EMCV의 설치류에 의한 전파 가능성을 확인하였으며, PCV2의 염기서열 분석을 통해 돼지에서 분리되어진 바이러스와 매우 유사한 바이러스임을 확인하였다.

Transmission of swine viral pathogens is achieved by direct contact with infected pigs or indirect contact through the various pathogen mediators including disease carriers. The wild rodents and cats have been suspected to be the reservoir or carrier of some of swine viral pathogens such as porcine circovirus type 2 (PCV2), encephalomyocarditis virus (EMCV), rotavirus (RV) pseudorabies virus (PRV). EMCV in rats and mice are believed to be the principal reservoir of the virus. In addition, PCV2 that is an important pathogen implicated in the pathogenesis of postweaning multisystemic wasting syndrome (PMWS) had been shown to be capable of viral replication in mice and give rise to fetal infection. Thus, the objective of this study is to determine the swine viral pathogens which are infective to wild rats, mice and cats and finally to evaluate the possibility of the wild animal-borne transmission of the important swine pathogens in Korea. For the study, we selected 12 pig farms, under consideration such as size of farm, recent history records, accessibility, and cooperation. Ninty-eight wild rats, 4 wild mice and 24 cats living around the swine farms were captured and then evaluated the infection of specific swine pathogens selected, ten viral pathogens (PCV2, PEDV, TGEV, RV, EMCV, PRRSV, PPV, PRV, JEV and CSFV) using appropriate verified methods such as PCR and RT-PCR. As results, among the swine viral pathogens examined, PCV2 (13/98) and EMCV (1/98) infection had been evidenced in the wild rats, PCV2 (2/4) and EMCV (3/4) in the wild mice, and PEDV (1/24) in the wild cats, which was limited to specific swine farms. Totally, PCV2 (15/126), PEDV (1/126) and EMCV (4/126) were detected in the wild rodents and cats. Sequence analyzed PCV2 isolated from rat. PCV2 nucleotide sequence homology range from 98.7% to 99.5% with Korean strains, Canadian strains, Chinese strains and France strains. PCV2 deduced amino acid sequence homology range from 96.0% to 98.4% with Korean strains, Canadian strains, Chinese strains and France strains. Based on our results, wild rats, mice could be reservoirs of PCV2, EMCV.
