The limitation of previous silk mat was that the size of silk mat was determined by the cocoon size. However, flatwise-spun silk mat (FS) has no limitation in production size. The purpose of this study was to compare FS to conventional silk mat (TDI) in terms of its physical and biological properties. Biodegradability assay, tensile strength, scanning electronic microscopic analysis, and FT-IR analysis were done. Bone regeneration was evaluated in the critical sized bone defect of rat model. FS group showed lower tensile strength and similar tensile strain to those of TDI group. When compared to TDI, FS group showed similar spectrum, but higher random coil and helix structures because of higher sericin contents. Biodegradability assay demonstrated similar behavior between FS group and TDI group. In animal model, FS group showed higher bone regeneration than TDI group in both micro-computerized tomogram and histomorphometric analysis. In conclusion, FS would have wider clinical application than TDI because of no size limitation.
