The purpose of this study was to isolate DNA probe which could identify Fusobacterium nucleatum strains in Southern blot analysis. This study procedure includes ⑴ shole-genomic DNA extraction of Fusobacterium nucleatum (F . nucleatum) ATCC 25586, F. nucleatum ATCC 237626, F. nucleatum ATTC 10953, F. nucleatum ATTC 49256, F. nucleatum ATCC 51190 (2) construction of the genomic DNA library. ⑶ screening of restriction fragment of genomic DNA, ⑷ identification of species-specific DNA probe by Southern blot hyridization. About 150 restriction fragements of several F. nucleatum genomic DNA digested with the Hind Ⅲ were obtained. One of them, the probe Fu9, restriction fragements of F. nucleatum Fusobacterium nucleatum ATCC 25586, was shown to be hybridized with all of the F. nucleatum strains used in this study but the length of the bands are different among the strains. From that result, the probe Fu9 may be used in identification of F. nucleatum stains.
