Due to the cyclic structure of the proline side chain, the proly peptides are in equilibrium between cis and trans isomer in solution. In general, proline contained random polypeptide chains exists in aqueous solution as of 10% the cis isomer, The cis-trans equilibrium constants and amounts of the cis(or trans)isomer are not greatly differ in nine different organic solvents tested. The presence of 0.3M LiCl in the solvent increases the amount of cis peptide in equilibrium and the extent of this increment depends of the solvent the largest increases are in TFE and acetone. The effect of LiCl on the cis isomer concentration in equilibrium increased linearly with the increasing LiCl concentration in the range of 0-0.5M. The dffect of LiCl /TFE system varied with the structure of Xaa of Succ-Ala-Xaa-Pro-Phe p-Nitroanilide tested. The order of the amounts of cis isomer concentration is Xaa> Ala> Glu> Lys> Ile> Leu> His> Gly. The LiCl/TFE system sws found to be able to apply usefully in the PPlase assay system.
