      In the current study, we examined the effects of Glechoma hederacea var. longituba extracts (GH) on ceramide production and anti-inflammatory efficacy in keratinocytes. HaCaT cells were stimulated with M5 (mixture of IL- 17A, IL-22, IL-1α, TNF-α, and oncostatin M) in order to establish a psoriatic keratinocyte model in vitro. Our results showed that treatment with GH resulted in a reduction of S1P (sphigosine 1-phosphate) levels by inhibition of ceramidase in HaCaT cells. In M5-treated HaCaT cells, release of proinflammatory cytokines and chemokines including TNF-α, IL-1β, CCL-1, and CCL-2 was significantly inhibited by GH. In addition, phosphorylation of MAPKs was also inhibited by GH. Phosphorylation of NF-κB and STAT3 was also inhibited by GH in psoriatic keratinocytes. These findings suggest that GH can inhibit production of ceramide and suppress expression of inflammatory proteins. Therefore, GH may be useful as a natural cosmetic material.
