표적 세포 전달용 마이크로스캐폴드(microscaffold)는 조직 재생을 위해 개발되었다. 그러나 지금까지 유사한 형태로 개발된 마이크로스캐폴드는 연골과 연골하골을 동시에 치료해야 하는 질환에 적용하는 데에 있어 한계를 가지고 있다. 본 연구에서는 연골 재생을 위한 자기 반응성 마이크로스캐폴드(magneto-responsive microscaffolds for cartilage regeneration, MRM-CR)와 연골하골 재생을 위한 자기 반응성 마이크로스캐폴드(magneto-responsive microscaffolds for subchondral bone regeneration, MRM-SBR)로 명명한 골연골 조직 재생을 위한 연골과 연골하골 맞춤형 자기 반응성 마이크로스캐폴드를 제안한다. 본 연구의 자기 반응성 마이크로스캐폴드는 이중 에멀전(double emulsion) 방식과 미세유체 유동(microfluidic flow) 제어를 이용하여 제작한다. MRM-CR과 MRM-SBR은 크기 차이에 의해 표면에 부착된 자성 나노 입자(magnetic nano partilces, MNPs)의 양에 차이가 나기 때문에 각각의 자화값이 다르다. 생체적합성 측면에서는 두 마이크로스캐폴드 모두 잠재적인 세포 운반체로서 독성 없이 세포를 성장시키는 것으로 나타났다. 마이크로스캐폴드의 자기 작동 테스트에서는, 동일한 자기장 조건에서 상대적으로 큰 MRM-SBR이 MRM-CR보다 더 빠른 것을 확인하였다. 실현 가능성 실험에서 진행한 3D 무릎관절 팬텀(phantom) 타겟팅 실험은 MRM-SBR과 MRM-CR이 순차적으로 타겟으로 이동하는 것을 보여주었다. 따라서, 제안된 자기 반응성 마이크로스캐폴드는 골연골 질환에 대한 비침습적 치료를 제공한다.

Microscaffolds capable of targeted cell delivery have been developed for tissue regeneration. However, the microscaffolds developed so far with similar morphologies have limitations for applications to osteochondral disease, which requires simultaneous treatment of the cartilage and subchondral bone. This study proposes magnetoresponsive microscaffolds tailored to the cartilage and subchondral bone for osteochondral tissue regeneration, named magnetoresponsive microscaffolds for cartilage regeneration (MRM-CR) and for subchondral bone regeneration (MRM-SBR). The morphologies of the magnetoresponsive microscaffolds were controlled using a double emulsion and microfluidic flow. In addition, due to their different sizes, MRM-CR and MRM-SBR have different magnetizations because of the different amounts of magnetic nanoparticles attached to their surfaces. In terms of biocompatibility, both microscaffolds were shown to grow cells without toxicity as potential cell carriers. In magnetic actuation tests of the microscaffolds, the relatively larger MRM-SBR moved faster than the MRM-CR under the same magnetic field strength. In a feasibility test, the magnetic targeting of the microscaffolds in 3D knee cartilage phantoms showed that the MRM-SBR and MRM-CR were sequentially moved to the target sites. Thus, the proposed magnetoresponsive microscaffolds provide noninvasive treatment for osteochondral tissue disease.
