This study was conducted to investigate the antimicrobial effect, antioxidative activity and cytotoxicity of Sanguisorbae Radix extract and fermented Sanguisorbae Radix extract.
Sanguisorbae Radix containing saponin embryo sac, pomolic acid, vitamin A, triterpenoides, tannin, and phenolic acid have several different functions, such as high blood pressure restraint, gout protection, antioxidative effect and anticancer.
In this respect, the following showed the results about antibacterial activity, antioxidant activity and cytotoxicity of fermented Sanguisorbae Radix extract.
1. Antibacterial activity of Sanguisorbae Radix extract was different according to declared bacteria, and looked on against B. subtilis, B. cereus, V. parahaemolyticus, S. aureus. antibacterial activity of fermented Sanguisorbae Radix extract appeared relatively highly against B. cereus and S. aureus and didn't show any difference.
2. The experiment results to find out the minimum inhibition concentration showed that if not fermented, there existed antibacterial activity in 5 percents concentration, and if fermented, 10 percents concentration.
3. Sanguisorbae Radix extract was safe from heat. It showed high antibacterial activity within the range of pH 3 to pH 7 but had been reduced within the range of pH 9 to pH 11.
4. The result of antibacterial activity with fractions of Sanguisorbae Radix extract looked on highly against B. cereus, B. subtilis, S. aureus, V. parahaemolyticus in water, n-BuOH, and EtOAc fractions and antibacterial activity with fraction concentration of extract had in over 5 percents.
5. Antifungal activity of Sanguisorbae Radix extract was lower than that of antibacterial activity, and antifungal activity was discovered against F. solani, R. solani, A. panax, C. gloeosporioides, P. expansum. The result of antifungal activity with various concentration had in even 10 percents concentration against F. solani, R. solani, and C. gloeosporioides.
6. Total flavonoid content of Sanguisorbae Radix extract showed over 65㎎/g regardless of fermentation, and it had been decreased after fermentation. Total polyphenol content expressed over about 20㎎/g, and it had been increased after fermentation.
7. Electron donating ability in comparison to Vitamin C showed over 90 percents' activity at about the same time, and antioxidative activity of Sanguisorbae Radix extract expressed highly. Superoxide dismutase-like activity showed 15 percents in fermentation before and after. So both electron donating ability and superoxide dismutase-like activity didn't show a special difference.
8. Nitrite scavenging ability expressed high activity, over 70 percents in 2.5 pH of an acid condition which was similar to the stomach of the body and showed higher activity than that of BHT. Also it will be able to be used for making inflammatory body's disease get better and be treated because it had the effect which prevents nitric oxide generating.
9. Sanguisorbae Radix extract had function that prevents generating lipid peroxidation in not only fibrolast cell but also HepG2 cell. Restraining lipid peroxidation immediately reduced MDA form and protected liver cell. With this reasons, it will be worth studying further and applying in the future.
10. Fermented Sanguisorbae Radix extract in 10㎍/㎎ concentration showed that there had the effect of Raw 264.7 cell activity, but in over 50㎍/㎎ concentration, there had no the effect of Raw 264.7 cell activity. In the effect of increasing fibrolast cell, made fibroblast cell growth rate higher than that of a contrast group in all of concentration. Therefore, it was summarized that the cytotoxicity of Sanguisorbae Radix extract appeared lowly. The cytotoxicity of HepG2 cell expressed the highest in from 10 to 50㎍/㎎ concentration. So with the reason why fibroblast cell as well as HepG2 cell showed a high cell viability over 80 percents in even a lot higher density, it should be apply to various fields as biological activities.
