To use inulin, a vector was constructed expression the INU1 gene of Kluyeromyces marxinus and inuF gene of Aspergillus niger for exoinulinase, and introduced into several Saccarmyces cerevisiae stains. S. cerevisiae YSH 2.64-2C produced the highest activity among five strains used as a host, when fructose was supplemented as a sole carbon source.
The gemomic DNA encoding exoinulinase A. niger ATCC 6275 was cloned and sequenced. Open reading framse (ORFs) of the exoinulinase gene, inuF, consisted of 1,671 nucleotides encoding 537 amino acids and intron consisted of 60 nucleotides. A. niger enzymes showed 48% identity with that of the Penicillium purpurogenum exoinulinase, but 90% identity with that of the Aspergillus awamori exoinulianse.
