The earthworm has been used as a fibrinolytic agent in East Asia for several thousand years. Fibrinolytic activities by the earthworm have been tested since 1983 and proteolytic enzyme, named Lumbrokinase, has been extracted and purified from Lumbricus rubellus. Lumbrokinase shows an ability of direct degradation of fibrin to minimize side effect, and it shows greater stability compared with other fibrinolytic agents. Recently, it is immobilized in a Korean type total artificial heart polyurethane valve, and it lead to decreased thrombus formation and better biocompatibility. We have investigated the three dimensional structure of this enzyme by the X-ray crystallography method. The microcrystals of Lumbrokinase Ⅲ-2 has been obtained in the condition of 18% PEG8000, 0.1M Na cacodylate buffer(pH6.5), and 0.2M magnesium acetate. The preliminary X-ray diffraction experiment shows that this crystal has a space group of P21 and three molecules in asymmetric unit. For the further structural analysis, the homology model of Lumbrokinase has been built from the structure of porcine beta trypsin. It is expected that the crystal structure of Lumbrokinase can be obtained in a short period by using molecular-replacement method and the high resolution three-dimensional stucture of Lumbrokinase will provide an crucial information about the protein engineering and new fibrinolytic agent development.
