      Biological efficiency (BE), the ratio of fresh mushrooms harvested per dry substrate weight and expressed as percentage of Lentinula edodes, also known as the shiitake was identified using the Sanjo 701 strain stored in the Department of Mushroom of the Korea National College of Agriculture and Fisheries. The strains were grown in glass columns with varying level of moisture content and different length of mixing periods of 0.5,1,2, and 3 hours. The substrate was sterilized to avoid contamination using the pressure steam Autoclave Sterilizer between normal and high pressure sterilization. Results show that mycelial growth (126mm/15 days) is optimized at 55% moisture level content. Two (2) hours mixing time yields the best mycelial growth of 117mm/15 days. On the other hand, normal pressure sterilization yielded better result with mycelial growth of 96mm/15 days at 100°C sterilization as compare to 88mm /15 days sterilization at 121°C. Mycelial density is higher, 3(+++) at normal pressure sterilization as compare to 2(++) at high pressure sterilization. Further, the sawdust with mixture of 5% woodchips also increased the porosity of the substrate and yielded higher mycelial growth. Thus, at this point we have shown the importance on how to achieve optimum harvest or potential increased yield of shiitake considering its moisture content, mixing time period and substrate porosity.
