In this study, we considered the development of microphytobenthos pigment analysis method using high performance liquid chromatography (HPLC) and applied it to the spatio-temporal distribution and community composition of microphytobenthos biomass in estuarine tidal flats. The optimal conditions for the microphytobenthos pigment analysis using HPLC were determined for sediment extraction method, HPLC method table, and IS-melting solvent to establish a more efficient measurement method for pigment analysis, and these improved method was applied to the measurement of biomass of microphytobenthos on the tidal flat of the Geum River estuary. For this, seasonal samplings were conducted in November of 2021, and February, May, and August of 2022. Diatoms dominated on the tidal flats of the Geum River estuarine getbol and Seocheon getbol, and various species such as Cryptophytes, Prymnesiophytes, Cyanobacteria, and Chrysophytes appeared. However, the peridinin's marker pigment of dinoflagellates has never been detected in the Geum River estuarine getbol and Seocheon getbol during four seasonal surveys. In addition, the concentration change of Chlorophyll a and Fucoxanthin, the main marker pigments of diatoms, showed similar patterns. Among the sediments, Microphytobenthos biomass showed a high concentration in February, an average of 3.9045 mg m-2 in the range of 0.038 – 7.771 mg m-2 in the Seocheon getbol, and an average of 37.3525 mg m-2 in the range of 0.822 – 73.883 mg m-2 in the Geum River estuarine getbol. The tidal flat at the mouth of the Geum River showed a concentration of about 9.57 times higher than that of the Seocheon getbol. In addition, more diverse pigments were detected in the tidal flat at the mouth of the Geum River estuarine getbol than in the Seocheon getbol.
