In this study, in order to promote the use of galic by-products, the anti-oxidant, anti-inflammatory, whitening effects and antimicrobial activity of water extract, 70% ethanol extract of Allium sativum L. Stems (ASSW, ASSE) was investigated. and developed transparent sunscreen cosmetic which has waterproofing property and no white turbidity when applied to skin. Total phenolics were more present in ASSE(44.7±1.32) than ASSW(37.08±1.51). Electron donationg ability (EDA) and ABTS+ radical scavenging activity were evaluated ASSW(44.02%), ASSE(50.84%) and ASSW(96.86%), ASSE(97.79%) at 1,000 μg/mL, respectively. To examine the potential anti-inflammatory effects of ASSW, ASSE levels of nitric oxide (NO) and pro-inflammatory cytokines, such as interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), prostaglandin-E2 (PGE2) and cell proliferation were measured. The ASSW, ASSE at 100 μg/mL concentration showed inhibitory effect on NO production by 17.75% and 22.02%, respectively. ASSW and ASSE showed inhibitory effects on the expression of the PGE2 by 54.33%, 59.87% when treated at 100 μg/mL. Production of interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β) were decreased by approximately over 15% upon ASSW, ASSE treatment (100 μg/mL). Allium sativum L. Stems extracts were tested for protein and mRNA of iNOS, COX-2 levels. These results suggest that ASSE has a suppressed production of many inflammatory mediators then ASSW by LPS. Allium sativum L. Stems extracts doesn’t inhibits pERK, pJNK, pp38. In this experiment, we know that ASSE is more effective than ASSW. so the solvent extracts(Hexan, Butanol, Ethyl acetate, Water) of Allium sativum L. Stems were investigated again for the activities of anti-oxidant and anti-inflammation. Ethyl acetate(ASSE-EA) showed the best effect at anti-oxidant among others. and then we have investigated decrease effects of ASSE-EA on protein levels of iNOS, COX-2. As a result iNOS, COX-2 indicated dose-dependently decrease 68.41%, 30.45% at 25 μg/mL, respectively. To confirm the skin-whitening effects of Allium sativum L. Stems, we did experiment such as tyrosinase, protein levels of tyrosinase, MITF, TRP-1 and TRP-2 on B16F10 melanoma cells. But, there was no suppression. antimicrobial activity also couldn’t show striking effects.
Measured the refractive index of water phase and oil phase to develop transparent sunscreen cosmetic. The transparent sunscreen is prepared by exploiting refractive index difference between oil-phase and water-phase of water-in-oil(W/O) emulsion. To make the same refractive index, we did control of polyol to raise refractive index in water phase and used the silicon oil for drop the refractive index of the oil-phase. SPF and PFA values of transparent sunscreen were indicated 30.99±1.65 and 3.01±0.30.
