본 연구에서는 천연물 유래의 항당뇨합병증 활성소재를 개발하기 위한 기초자료 확보를 위하여 실험을 수행하였으며, 미나리 70% ethyl alcohol 추출물의 ethyl acetate 분획물에서 우수한 최종당화산물 생성 저해활성(IC50=52.2±3.5μg/mL)을 확인하였다. 활성성분의 동정을 위하여 Toyopearl-HW40 및 ODS gel을 충진제로 활용한 칼럼크로마토그래피를 수행하여 4종의 flavonoid 유도체 화합물을 분리하였다. 분리한 화합물의 구조는 NMR, MS 스펙트럼 등의 기기분석 데이터 해석과 표준품과의 HPLC 직접 비교를 통하여 hyperoside, isorhamnetin 3-O-galactoside, isorhamnetin 및 persicarin으로 구조 결정하였다. 이들 화합물에 대하여 최종당화산물 생성 저해활성을 평가한 결과galactose가 결합되어 있는 배당체 화합물인 hyperoside 및 isorhamnetin 3-O-galactoside의 IC50 값이 각각 28.7±1.3 및 41.5±2.5 μM로 가장 우수한 생성 저해활성을 나타내었다. 또한 이들 물질의 hydroxyl 라디칼 소거 활성은 hyperoside의 IC50 값이 14.3±0.9 μM로 가장 우수한 소거활성이 나타났으며, methoxyl기가 치환된 isorhamnetin 3-O-galactoside는 15.0±1.6 μM로 유사한 IC50 값을 확인하였다. 비배당체인 isorhamnetin 및 persicarin의 활성은 위에서 언급한 배당체에 비하여 상대적으로 낮은 활성임을 확인하였다. 향후 우수한 활성을 나타낸 배당체 화합물에 대하여 활성 기작 및 추가적인 연구가 필요하다고 생각된다.

Bioactivity-guided isolation was conducted on a ethyl acetate (EtOAc)-soluble portion of an aqueous ethanolic extract from Oenanthe javanica using an advanced glycation end products (AGEs) formation inhibition assay. The experiments led to isolation of four previously characterized flavonoids, hyperoside (1), isorhamnetin 3-O-galactoside (2), isorhamnetin (3), and persicarin (4). The structures were established on the basis of NMR and MS spectroscopic data interpretations. Among these isolated compounds, hyperoside (1) and isorhamnetin 3-O-galactoside (2) showed significant inhibitory effects against AGEs formation, with IC50 values of 28.7±1.3 and 41.5±2.5 μM, respectively. Furthermore, all tested compounds were evaluated for radical scavenging activity using hydroxyl radicals, and compounds 1 and 2 exhibited the most potent capacities. These results suggest that the flavonoids isolated from O. javanica might be beneficial for the prevention and treatment of diabetic complications and related diseases.
