The purpose of this study was to investigate the pathogenesis of experimental lipid nephrosis in albino rats.
In the investigation, histopathological findings, changes in number of mesangial cells and quantitative changes of proteinuria induced by single and multiple injections of aminonucleoside and a single injecton of diethylene glycol were observed.
1) In the group with a single injection of diethylene glycol, marked pathological changes were seen in the proximal convoluted tubules during the first and second day following the injection. On the fifth day the above mentioned changes resumed to normal. During that time the number of mesangial cells did not increased.
2) In the group with a single injection of a large dose of aminonucleoside, increased proteinuria was seen on the first day after the injection and its peak was reached on the 15th to the 21st day. In the group with repeated injections of six divided small doses, an increased proteinuria was observed on the 8th day after the injection and its peak was reached on the 14th to the 22nd day.3) In the group with a single injection of a large dose of aminonucleoside, the histopathological changes of the renal tubules were observed on the fifth to the 31st day and the most remarkable changes occurred on the 15th to the 21st day after the injection. In the group of six repeated smallinjections, the changes were seen on the 9th to the 25th day, and its peak was on the 15th day.
4) In comparing the number of mesangial cells in the nephrotic animal with that of normal albino rats (av. 52.6/10 glomeruli), it was observed that a single large injection of aminonucleoside resulted in the increase of the cells to 60 on the fifth day, 64 on the 15th day, 63 on the 21st day, and 57 on the 32nd day. In tile group with six repeated injections the number of cells was 61 on the 9th day, 63 on the 15th day, and 61 on the 25th day.
5) In conclusion the number of mesangial cells had a tendency to increase in the animal which received aminonucleoside. This increase was greatest in the period when the histological change was at its peak.
