Using SRB assay, cytotoxicity test was performed on human lung carcinoma, A549 cell line for screening the soil microorganisms producing anticancer agent. Among 4,265 microorganisms, one microorganism was selected for its effect on the expression of c-myc gene. The culture extract of selected microorganism, named No.2303, inhibited the expression of c-myc gene in human lung carcinoma, A549 cells. The optimal culture condition for the production of anticancer material was determined as follows ; 3.0% lactose, 2.0% yeast extract, 2.0% polypeptone, 1.0% KH_2PO_4, 3.0% Na_2HPO_4, 12H_2O, and 7 day cultivation at 30℃. The effect of extract on the morphology and apoptosis was also studied. The extract induced the morphological changes and apoptosis in A549 cells within 24 hours.
