      Advanced anticoagulant suction drainage was prepared by anticoagulant coating with the hydrophobic multicomponent heparin conjugates, and named as MCK-Hepafilm (15K). The coating surface was analyzed by the staining test of azure-A and scanning electron microscope (SEM), which shows excellent coating effect on silicone as well as PVC. The coating condition of silicone tube was optimized to 3.0 wt% concentration of coating solution and the solvent casting temperature to 35 oC. In vitro anticoagulant effect is confirmed by observation of platelet adhesion, blood coagulation and adhesion of bacteria. Platelet adhesion on coating surface of PVC and silicone drainage tube was remarkably reduced, and completely prevented from whole blood clots of rabbit. The adhered number of Escherichia coil (E. coil) bacteria on coated surface of PVC drainage tube was reduced with 7.2?.0 CFU/cm2 as compared to the non-coated surface of 10.1?.2 CFU/cm2. Rabbit experiment has been carried out for the MCK-Hepafilm (15K) coated suction drainage device. The efficacy of the MCK-Hepafilm coat was examined by measuring the debris weight forming clogging in the tube (MCK-Hepafilm coated & uncoated) and analyzing the change in blood flow pressure through it, over a period of time, after insertion in the abdomen of the rabbit. In the coating tube, the internal resistance of PVC was reduced, and also the debris weight of silicone was reduced to 315.5?.4 mg as compared with non-coated 555.7?0.6 mg. The obtained results indicate the effectiveness of this MCK-Hepafilm (15K) coating technology for blood contacting medical devices including suction drainage devices in improving their hemocompatibility.
