Differentiation of Penicillium chrysogenum YUFE 1308 during shake and submerged culture was studied.
Practical media of penicillium production and synchronizing culture technique were involved in the experiments.
Changes of the levels in mycelial ribonucleic acid, total proteins, total carbohydrates, residual sugars, nitrate ions, and pH values are determined.
Sporulation of this fungus occurred in submerged culture at 24 hours after replacement when the levels of residual glucose had decreased and nitrogen source had been limited. Optimal range of pH was between 5.4 to 5.8. Synthetic rates of total proteins, total RNAs and total carbohydrates in the mycelia decreased when sporulation is induced.
Branching of the hyphae occurred when levels of residual sugars, nitrate ions and pH values had fallen respectively in shake culture at 48 hours after hyphal growth.
Antibiotic activities were detectable in the extraction of culture broth from presporulation and sporulating stages.
