The microorganisms producing cellulase were isolated from environmental soil of Duryu Park in Daegu, Korea. One strain producing cellulase was selected by forming an active zone on a trypan blue LB agar plate containing 0.5% carboxymethyl cellulose (CMC) and 0.02% trypan blue. In the analysis of 16S rRNA gene sequence from the strain, the isolated strain showed 99.43% of identity with Lactobacillus murinus so that the isolated strain was named Lactobacillus murinus HP-01. And then, isolated L. murinus HP-01 was cultivated on TSB-G medium including 0.5% CMC as a substrate. As a result, the strain showed the highest activity of cellulase after 2 days. In the effect of pH and temperature, cellulase from L. murinus HP-01 showed the highest activity at pH 5.0 and 50°C, respectively. On the optimal condition, the cellulase was found to be stable at pH ranging from 4.0 to 9.0, and it exhibited thermal stability with a half-life of 1.24 hours at 50 °C. In the investigation of substrate specificity for cellulosic substrates, the specific activity of the cellulase from L. murinus HP-01 was highest for CMC, followed by sigmacell cellulose, alpha cellulose, swollen avicel cellulose.
