      Changes in the activity of serum and liver G-6-Pase were studied in acutely and chronically carbontetrach oride poisoned rabbits, and evidences were presented indicating that acute and chronic form of liver damage can be easily detected by the determination of serum G-6-Pase activity.
      1. The G-6-Pase was most abundantly found in the liver, and in liver localized exclusively at the cytoplasmic fraction. In normal serum only negligible activity was detected.
      2. The enzyme activity in the serum increased within a few hours after the injection of carbon tetrach oride, the highest activity was reached after 24 hours, and thereafter the enzyme level decreased sharply, although the decreasing rate was not so great as found in serum activity of glutamic-pyrubic transaminase.
      3. A considerable amount of G-6-Pase activity in acutely CCl₄ poisoned rabbits (at 50 hr. after CCl₄ poisoning) was still found to remain in liver. However, in chronically CCl₄ poisoned rabbits (CCl₄, given for one month every other day), only negligible activity was found both in serum and in liver.
      4. The medium G-6-Pase activity (liver slice shaked in oxygen saturated Ringer's solution for 60 minutes at 37℃) were found to rapid appearance. The highest level of medium G-6-Pase activity was reached after 15 minutes, and thereafter decreased gradually. By contrast, the highest level of medium GPT activity was reached after 30 minutes, and thereafter maintained with equally.
      5. Fresh unfixed Cryostat's frozen sections were incubated in Wachstein-Meisel's medium. In normalrabbits, the G-6-Pase activity in the cytoplasm of hepatic cells was noted considerably, the cells of peripheral area showed more pronounced activity than those of the central area. In acute CCl₄ poisoned rabbits, the G-6-Pase activity in the cytoplasm of hepatic cells was noted diffusely. In chronic CCl₄ poisoned rabbits, negligible activity of G-6-Pase was found diffusely in central and peripheral area.
      6. From these results, it was inferred that Increased G-6-Pase activity was noted in the serum of acute CCl₄ poisoned rabbits and decreased G-6-Pase activity were noted in the serum and tissue of chronic CCl₄ poisoned rabbits.
