Construction of a New Transfer Vector for Baculovirus Carrying E. coli Galactokinase Gene = 대장균의 galactokinase 유전자를 가지고 있는 Baculovirus를 위한 새로운 이동벡터의 개발

Autographa californica californica nuclear polyhedrosis virus의 다각체 유전자 지역에 대장균이 galactokinase 유전자를 가지고 있는 새로운 이동벡터가 만들어졌다. Plasmid pAcI은 AcNPV의 EcoRI-I 단편을 pUC18의 EcoRI 자리에 크로닝한 후 두 개의 BamHI 자리를 제거하여 만들어졌다. 대장균의 glaK 유전자를 가지고 있는 Plasmid pKO100는 제한효소로 절단된 후 1% agarose gel에서 분리 되었고 gaIK 유전자를 포함하고 있던 DNA 단편이 gel 조작으로부터 전기적으로 회수되었다. pAcGKI은 gaIK 유전자를 포함하고 있는 DNA 단편을 pAcI의 polyhednn promoter 지역에 크로닝함으로써 만들어졌다. galk 유전자는 polyhednn promoter의 조절기작을 밝히기 위한 보고 유전자로써 유용할 것이다.

A new transfer vector with E. coli galactokinase gene (galK) inserted within Autographa californica, nuclear polyhedrosis virus (AcNPV) polyhedrin coding region has been constructed. Plasmid pAcI was generated by cloning EcoRI-I fragment of AcNPV into the EcoRI site of pUC18 and deleting two BamHI sites. Plasmid pKOl00 carrying E. coli galK gene was cut with restriction endonuclease, separated on 1% agarose gel, and DNA fragment containing galK gene was electroeluted from the gel piece. pAcGK1 was constructed by inserting DNA fragment containing galK gene into the polyhedrin coding region of pAcI. The galK gene would be useful as a reporter for the elucidation of the regulation mechanism of polyhedrin promoter.
