백내장 수정체의 상피세포에 있어 TGF-βisoform mRNA 발현의 정량분석 = mRNA expression of the TGF-βisoforms in cataract lens epithelial cells

국문 초록 (Abstract)

목적: Transforming growth factor(TGF)-β는 백내장의 발생과 관련이 있는 것으로 알려져 있다. 백내장 수정체에서 TGF-β isoform mRNA의 발현을 정량분석하여 각각의 TGF-β isoform과 백내장 발생의 연관성을 알아보고자 하였다.
방법: 전체 30명, 30안에서 추출한 수정체상피세포조직을 이용하여 real-time SYBR Green polymerase chain reaction으로 TGF-β1, β2와 β3 mRNA의 발현을 정량하여 백내장의 종류(노인성 25안, 초로성 5안)와 당뇨병의 유무(당뇨 17안, 비당뇨 13안)에 따라서 비교 분석하였다.
결과: TGF-β3 mRNA는 30안 모두에서, TGF-β1 mRNA는 당뇨 노인성 백내장 1 안을 제외한 29안(96.7%)에서, TGF-β mRNA는 9안(당뇨 노인성 3/14안, 21.4%; 비당뇨 노인성 2/11안, 18.2%; 당뇨 초로성 2/3안, 66.7%; 비당뇨 초로성 2/2안, 100%)에서 발현되었다. TGF-β1 mRNA의 발현양은 당뇨 노인성 백내장에서 비당뇨 노인성 백내장과 비교하여 유의하게 적었다(P=O.056).
결론: TGF-β는 백내장의 발생과 연관이 있으며, TGF-β2는 초로성보다는 노인성 백내장에서, TGF-β1은 노인성 백내장에 있어 당뇨가 있는 경우에 더욱 중요한 의미를 갖는 듯 하다

다국어 초록 (Multilingual Abstract)

Objectives: Transforming growth factor (TGF)-0 has been accepted to be associated with cataractogenesis. I quantitated the mRNA expression of TGF-0 isoforms in cataractous crystalline lens to determine the effect of the isoforms on cataractogenesis.
Methods: With lens epithelial cells of total thirty eyes in thirty patients, mRNA expression of TGF-D1, D2 and )33 were quantitated by real-time SYBR Green polymerase chain reaction and the results were compared according to types of cataract (senile, 25 eyes vs. presenile, 5 eyes) and presence of diabetus mellitus (diabetic, 17 eyes vs. non-diabetic, 13 eyes).
Results: Each isoforms mRNA of TGF-0 were expressed; TGF-03 in all thirty eyes, TGF-01 in 29 eyes (96.7%) except one diabetic senile cataract and TGF-02 in 9 eyes (3/14 eyes of diabetic senile, 21.4%; 2/11 eyes of non-diabetic senile, 18.2%; 2/3 eyes of dabetic presenile, 66.7%; 2/2 eyes of non-diabetic presenile, 100%). The amount of TGF-Dl mRNA expression was significantly low the diabetic senile cataract compared to the non-diabetic senile cataract (P = 0.056). Conclusions: TGF-D would be associated with cataractogenesis. TGF-02 might be more meaningful in the senile cataract than in the presenile and TGF-01 in the diabetic condition of the senile cataract than in the non-diabetic.
