      • A genetic system for RNase E variant-controlled overproduction of ColE1-type plasmid DNA

        Escherichia coli endoribonuclease RNase E (Rne) regulates replication of ColE1-type plasmids by cleaving RNA I transcripts, which are synthesized from the plasmid and regulate the plasmid replication as antisense RNA. Here, we report the development of a genetic system that efficiently overproduces ColE1-type plasmid DNA when an RNase E variant that confers a hyperactive phenotype in RNA I cleavage is conditionally expressed from chromosome. This genetic system offers a method for isolation of large quantities of pure ColE1-type plasmid DNA, which have been most commonly used as molecular biology and biotechnology tools for research and industrial purposes.

        Expression of toll-like receptor 2 and 4 is increased in the respiratory epithelial cells of chronic idiopathic interstitial pneumonia patients

        Background: Idiopathic interstitial pneumonia (IIP) is characterized by chronic interstitial inflammation and fibrosis. Although mounting evidence has suggested that toll-like receptor (TLR) 2 and TLR4 are involved in the pathogenesis of non-infectious lung injury in vitro and in mouse models, their roles in human IIP remain unknown. Methods: To address this issue, we investigated the expression patterns of TLR2 and TLR4 by immunohistochemistry in resected lung tissues from patients with usual interstitial pneumonia (UIP) or nonspecific interstitial pneumonia (NSIP). Results: Type II pneumocytes, bronchial epithelial cells (BECs), and alveolar macrophages accounted for the majority of TLR2- and TLR4-expressing cells in both UIP and NSIP. The numbers of TLR2 and TLR4-positive respiratory epithelial (RE) cells, including type II pneumocytes and BECs, were significantly greater in UIP and NSIP than in the control. In particular, the numbers of TLR2-positive RE cells were much greater in UIP than in NSIP. The intensities of TLR2 and TLR4 expression in type II pneumocytes were also significantly stronger in UIP and NSIP than in the control. A comparison of the TLR expression patterns between the fibroblastic and fibrotic areas in UIP indicated that the numbers TLR2 and TLR4-positive RE cells were similar in fibroblastic areas, whereas the TLR2-positive RE cells outnumbered the TLR4-positive RE cells in the fibrotic areas. Conclusions: This study demonstrates that RE cells over-express TLR2 and TLR4 in the lungs of IIP patients. These findings suggest that high expression of TLRs may contribute to the pathogenesis of human IIP.

        Scytalidium parasiticum sp. nov., a New Species Parasitizing on Ganoderma boninense Isolated from Oil Palm in Peninsular Malaysia

        A mycoparasite, Scytalidium parasiticum sp. nov., isolated from the basidiomata of Ganoderma boninense causing basal stem rot of oil palm in Johor, Malaysia, is described and illustrated. It is distinct from other Scytalidium species in having smaller asci and ascospores (teleomorphic stage), longer arthroconidia (anamorphic stage), hyaline to yellowish chlamydospores, and producing a fluorescent pigment. The phylogenetic position of S. parasiticum was determined by sequence analyses of the internal transcribed spacers and the small-subunit ribosomal RNA gene regions. A key to identify Scytalidium species with teleomorphic stage is provided.

        Fish Purchasing Behavior and Implications for Promoting Sustainable Consumption: A Malaysian Case

        Purpose: Motivated by the fact that Malaysians fish consumption patterns and preferences have raised sustainability concerns, this study aims to understand the key entry points to promote sustainable consumption and increased uptake of sustainably sourced fish among Malaysian consumers. This is achieved by exploring the individual determinants of Malaysian consumers’ fish purchasing behaviors and their subjective and objective knowledge of fish. Research design, data, and methodology: A survey was conducted using interviewled questionnaire on 250 consumers in selected Malaysian populations recruited via street-intercept random sampling. Results: Findings confirmed the homogeneous societal culture of fish consumption amongst Malaysians. Females and consumers with advanced degree are found to be potentially effective primary targets of sustainable consumption interventions. Future interventions should consider the solutions to the low health and sustainability literacies among Malaysian consumers, and limited availability, accessibility, and affordability of sustainable options in Malaysia. Conclusion: The study findings provide new insights for the multisectoral stakeholders in the region working on promoting sustainable fish consumption and sustainable fisheries in general.

        A Meta-Analysis on the Impact of Induction Chemotherapy on Survival Outcomes for Sinonasal Squamous Cell Carcinoma

        Background and Objectives: Malignant neoplasms of the nasal cavity and paranasal sinuses are an aggressive form of tumour that tends to be diagnosed at a locoregionally advanced stage. Among its various histological subtypes, squamous cell carcinomas (SCC) are the most common form of sinonasal cancer, with approximately 50%–60% of sinonasal malignancies being SCC. This review aims to investigate the impact of induction chemotherapy on survival outcomes for patients undergoing cancer care for sinonasal squamous cell carcinomas (SNSCC).Methods: Two reviewers independently assessed 2,118 studies pooled from four bibliographic databases, namely MEDLINE, Embase, Cochrane Central Register of Controlled Trials and Web of Science and hand-searched grey literature. This systematic review and meta-analysis was performed in accordance with the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines. As this was a time-to-event analysis, hazard ratios (HRs) and standard errors were derived from individual studies and pooled together for analysis. If these values were not available directly, statistical methods were used to indirectly extract the required data via Kaplan-Meier curves or via the p-value and the numbers of events.Results: Results suggest a notable improvement in overall survival (HR=0.56, 95% confidence interval [CI]=[0.36, 0.86], p<0.009) and a promising improvement in disease-free survival (HR=0.82, 95% CI=[0.62, 1.08], p=0.16) for patients who underwent induction chemotherapy in addition to definitive treatment for SNSCC.Conclusion: Our findings add to existing literature by providing a precise pooled estimate of the beneficial effects, revealing that induction chemotherapy is not just a viable add-on to traditional treatment regimens but also improves survival outcomes when compared to definitive local therapy as a standalone treatment. Clinicians should consider incorporating induction chemotherapy as a treatment option for locoregionally advanced SNSCC.

        Another Diachronic Perspective on English Tough-Constructions

Ever since toughconstructions in English began to attract the attention of early transformational grammarians, there has been considerable controversy about the analysis of the syntactic and/or semantic relationship between the tough-subject and the gap in the infinitival phrase/clause. Although the type of approach that assumes no syntactic connectivity has long been favored by current syntactic frameworks, it is not clear whether this type of approach is a truly well-motivated position supported by sufficient empirical evidence. In particular, it has been noted that there is some substantia! evidence both for and against it. This paper attempts to provide a new perspective on the analysis of English tough-constructions by considering the insights from the diachrony of prepositional stranding constructions in the history of English.

        흙-토목섬유(土木纖維) Interface 특성(特性)에 관한 연구(硏究)

        The objective of this paper is to show that the soil-geotextile interaction needs to be addressed in addition to the usual tensile and modulus properties when the geotextile is being designed for a specific application. The soil-geotextile interaction can be directly assessed by standard direct shear test. The data presented here show that the shear strength paramaters describing the soil-geotextile interface can he greatly influenced by the type of the geotextile. In this investigation, we examined nine different geotextiles of varying construction and surface textures with two standard soil, under five loading conditions, and compared the shear strength and the frictional resistance with the corresponding values of soil itself. The following conclusions were drawned from this study. 1. The shear stress-strain curve shows that there are the residual shear stresses at the soil-geotextile interface. Because of the hydraulic gradient between the soil and the geotextile, the excessive pore water can migrate into the geotextile and among the filaments and dissipate through the soil-geotextile interface. 2. The shear strength of the soil-geotextile interface is affected by the moisture content of the soil. At moisture content lower than the optimum water content of the Proctor compaction test, the shear strength of the soil-geotextile interface is greater. 3. The type and surface roughness of the geotextile have the greatest influence on the interface friction angle between the soil and the geotextile. 4. The maximum internal friction angle mobilized between the soil and the geotextile is on the average 82% of that of the soil itself. 5. The maximum adhesion mobilized between the soil and the geotextile is on the average 67% of that of the soil itself,
