Induction of erythroleukaemia by F - MuLV and Purification of Erythropoietin

Friend Murine Leukemia Virus (F-MuLV) is affective against 2-day old new borns in NFS/n Swiss mice. Infected babies suffer from splenomegaly and anemia at the age of 4-5 weeks beyond which the condition worsens and is fatal. However, the spleen size can be reduced by hypertransfusion for 8 weeks. If left alone after this treatment, the spleen enlarges and becomes palpable again. This time the spleen cells are transplantable and inflict the same disease if injected to normal adults. While these leukemic cells were considered to be hormone-independent, addition of erythropoietin (EPO) was found to be essential for maximum growth in culture. Influence of EPO on the in vivo growth of leukemic cells was indicated by our findings that administration of the hormone caused a more severe leukemia and rapid death while, transfusion of red blood cells, which lowers endogenous EPO, led to increased survival of leukemic mice. It is suggested that hormone-associated therapy may be efficacious in the treatment of this and perhaps, other leukemias. Several erythroleukemic cell lines released factors which stimulated erythropoiesis in vivo and in vitro. A simple five step scheme has been devised which allows purification of this erythropoietic activity to apparent homogeneity. The methods employed included lectin affinity chromatography (wheat germ agglutinin), gel filteration (ultro gel ACA 44), ion exchange, hydroxylapatite, and high performance liquid chromatography, Following polyacrylamide gel electrophoresis, biological activity was recovered in an area corresponding to a molecular weight of 35,000 daltons. Silver staining of a polyacrylamide gel after electrophoresis of our most purified preparation revealed a single band of 35,000 daltons.

Since the velocity is dependent on the fiber orientation in anisotropic composites, the application of traditional acoustic emission (AE) source location techniques based on the constant velocity to composite structures has been practically impossible. The anisotropy makes the source location procedure complicated and deteriorates the accuracy of the location. In this study, we have divided the region of interest(ROI) into a set of finite elements, taken each element as a virtual source, and calculated the arrival time differences between sensors by using the velocities at every degree from 0 to 90. The calculated and the experimentally measured values of the arrival time difference are then compared to minimize the location error. The results from two different materials, namely AA6061-T6 and CFRP(uni-directional, UD, [0]_32) laminate confirmed the practical usefulness of the proposed method.

The purpose of this study is to analyze the Beaux-Arts model as modern educational system focused on Paul P. Cret's architecture. The study process is to investigate his writings on American Beaux-Arts system and his architecture indicating Beaux-Arts composition related to American classicism. It is Paul Cret who influenced the establish-ment of academic system a professional practice. The artistic and academic nature of architectural practice is the essence of Beaux-Arts movement to redefine a modern profession. By stressing the importance of the his achieved through a rigorous training in Beaux-Arts system, the formulation of a rational method and vocabulary of american modern made possible practice and architectural education through design methodology focusing on planar form.
