        Physiological Characteristics of Lactobacillus casei Strains and Their Alleviation Effects against Inflammatory Bowel Disease

        Lactobacillus casei, one of the most widely used probiotics, has been reported to alleviate multiple diseases. However, the effects of this species on intestinal diseases are strain-specific. Here, we aimed to screen L. casei strains with inflammatory bowel disease (IBD)-alleviating effects based on in vitro physiological characteristics. Therefore, the physiological characteristics of 29 L. casei strains were determined, including gastrointestinal transit tolerance, oligosaccharide fermentation, HT-29 cell adhesion, generation time, exopolysaccharide production, acetic acid production, and conjugated linoleic acid synthesis. The effects of five candidate strains on mice with induced colitis were also evaluated. The results showed that among all tested L. casei strains, only Lactobacillus casei M2S01 effectively relieved colitis. This strain recovered body weight, restored disease activity index score, and promoted anti-inflammatory cytokine expression. Gut microbiota sequencing showed that L. casei M2S01 restored a healthy gut microbiome composition. The western blotting showed that the alleviating effects of L. casei M2S01 on IBD were related to the inhibition of the NF-κB pathway. A good gastrointestinal tolerance ability may be one of the prerequisites for the IBD-alleviating effects of L. casei. Our results verified the efficacy of L. casei in alleviating IBD and lay the foundation for the rapid screening of L. casei strain with IBD-alleviating effects.

        미세먼지 처리에 따른 전나무, 중국단풍, 소나무, 굴참나무의 생리⋅생화학적 반응 및 흡착 특성

        In recent years, the frequency of warnings about particulate matter (PM) has gradually increased in Korea, along with an increase in its intensity. Because of their vast surface area, reactivity to external particles, and characteristics of their leaves, urban trees can act as biofilters, reducing PM pollution. However, the air pollutant PM can cause various types of damage not only to human health but also to vegetation. Studies performed to date on the responses of trees to PM are still insufficient. Here, we analyzed the correlation between PM adsorption and physiological and biochemical responses of four major street tree species, namely, Abies holophylla, Acer buergerianum, Pinus densiflora, and Quercus variabilis, under conditions of approximately 300 µg m-3 of fly ash emissions using a phytotron. The results showed that the physiological and biochemical responses and PM adsorption differed depending on the tree species. In correlation analysis, it was confirmed that there were positive correlations between physiological factors, and PM adsorption on adaxial leaf surfaces negatively impacted the physiological characteristics. This study provides fundamental information for selecting tree species to reduce PM pollution and develop sustainable urban forests. 도시와 자연에 부정적인 영향을 끼치는 미세먼지에 대한 저감 연구가 점점 심화되고 있다. 이에따른 미세먼지 저감 대책 중 하나로써 도시 수목의 미세먼지 저감 연구가 활발히 진행되고 있다. 하지만 이와 관련하여 미세먼지에 의한 도시 수목의 피해 반응 연구는 부족한 실정이다. 본 연구는 도시 수목 중 전나무(Abies holophylla), 중국단풍(Acer buergerianum), 소나무(Pinus densiflora), 굴참나무(Quercus variabilis)에 대하여 고농도 미세먼지 처리로 나타나는 피해반응을 조사하기 위한 목적으로 수행되었다. 위 네 수종을 대상으로 식물생장조절챔버(phytotron)에서 시간 당 300 μg m-3 농도의 미세먼지를 처리하여 각 수종의 생리적, 생화학적 변화 반응과 잎의 흡착면적을 측정하였다. 그리고 순광합성량, 기공전도도, 증산률, 엽록소 함량, ROS, MDA, 잎의 흡착면적에 대해서 상관 분석을 진행하여 상호 간의 관계를 살펴보았다. 연구 결과, 수종마다 생리·생화학적 반응과 흡착면적이 종 특이적으로 나타났다. 순광합성량의 경우 전나무에서 가장 큰감소세를 보였으며 소나무와 굴참나무는 큰 변화를 나타내지 않았다. 굴참나무는 ROS의 변화 또한 미미하였다. 모든 수종에서 MDA 함량이 공통적으로 증가하였다. 4주 처리 이후 흡착면적에서 전나무는 잎의 앞면, 소나무는 잎의 뒷면에 높은면적량을 나타냈다. 상관성 분석 결과 생리적 반응 요인들 간에 양의 상관관계가 있음을 확인하였고, 잎의 앞면에 높은 흡착면적을 보일수록 생리적 반응에 부정적인 영향이 있음을 확인하였다. 본 연구는 미세먼지 저감과 지속 가능한 도시 숲을효율적으로 조성하기 위한 수종 선택의 기초 데이터를 제공한다.

        새만금간척지에 식재한 포플러 클론의 생리적 특성

        본 연구에서 일반 토양에 비해 무기양분 함량이 적고 배수성이 좋지 않은 새만금간척지 토양에 식재 된 포플러수종의 생리적 특성을 조사하였다. 7월과 8월에, 이태리포플러 클론인 Venziano, L.Avanzo와 미루나무 교잡종 클론인97-18, 97-19은 생리적인 특성의 차이를 보였다. 이태리포플러 클론이 미루나무 교잡종 클론에 비해 생리적 특성이 우수하였다. 특히 Venziano 클론의 순광합성률은 각각 평균적으로 18.62 μmol CO<sub>2</sub> m<sup>-2</sup> · s<sup>-1</sup>, 17.11μmol CO<sub>2</sub> m<sup>-2</sup> · s<sup>-1</sup>로 다른 클론들과 비교해서 높은 수치를 보였다. 반면, 미루나무 교잡종인 97-18클론의 7월 광색소 함량은 2.37 mg · g<sup>-1</sup>FW으로 가장 높은 수치를 보였으나, 8월에는 이태리포플러 클론보다 낮은 함량을 보였다. 미루나무 교잡종은 8월에 순광합성률, 광색소함량 등 전반적인 생리적 활성이 떨어졌다. 결론적으로, 새만금간척지에 식재된 포플러 묘목의 생리적인 특성은 이태리포플러 종인 Venziano 클론이 우수하였다. This research aims to study the physiological characteristics of poplar clones in Saemanguem reclaimed land where has lower minerals and bad drainage than those in paddy field. In July and August, different poplar clones including Populus euramericana clone (Venziano and L.Avanzo) and deltoides hybrid clone (97-18 and 97-19) showed different physiological characteristics. Physiological characteristics of P. euramericana clones were better than those of P. deltoides hybrid clones. Especially, the average net photosynthetic rate (A<sub>net</sub>) of clone Venziano were 18.62 μmol CO<sub>2</sub> m<sup>-2</sup> · s<sup>-1</sup> and 17.11μmol CO<sub>2</sub> m<sup>-2</sup> · s<sup>-1</sup>, respectively. Those were the highest numerical values among other poplar clones. On the other, the total chlorophyll and carotenoid contents of P. deltoides hybrid clones were higher than those of P. euramericana clones in July. Particularly, the total chlorophyll content of clone 97-18 was 2.37 mg · g<sup>-1</sup>FW. In August, those of P. deltoides hybrid clones decreased than in July. In term of the physiological activity such as net photosynthetic rate and stomatal conductance of P. deltoides hybrid clones were much lower in August than in July. As a result, physiological characteristics of P. euramericana Venziano clone was superior than the other clones in Short rotation coppice (SRC) in Saemanguem area.

        Physiological Characteristics and Production of Folic Acid of Lactobacillus plantarum JA71 Isolated from Jeotgal, a Traditional Korean Fermented Seafood

        Folic acid, one of the B group of vitamins, is an essential substance for maintaining the functions of the nervous system, and is also known to decrease the level of homocysteine in plasma. Homocysteine influences the lowering of the cognitive function in humans, and especially in elderly people. In order to determine the strains with a strong capacity to produce folic acid, 190 bacteria were isolated from various kinds of jeotgal and chungkuk-jang. In our test experiment, JA71 was found to contain $9.03{\mu}g/mL$ of folic acid after 24 h of incubation in an MRS broth. This showed that JA71 has the highest folic acid production ability compared to the other lactic acid bacteria that were isolated. JA71 was identified as Lactobacillus plantarum by the result of API carbohydrate fermentation pattern and 16s rDNA sequence. JA71 was investigated for its physiological characteristics. The optimum growth temperature of JA71 was $37^{\circ}C$, and the cultures took 12 h to reach pH 4.4. JA71 proved more sensitive to bacitracin when compared with fifteen different antibiotics, and showed most resistance to neomycin and vancomycin. Moreover, it was comparatively tolerant of bile juice and acid, and displayed resistance to Escherichia coli, Salmonella Typhimurium, and Staphylococcus aureus with restraint rates of 60.4%, 96.7%, and 76.2%, respectively. These results demonstrate that JA71 could be an excellent strain for application to functional products.

        Physiological Characteristics and GABA Production of Lactobacillus plantarum K255 Isolated from Kimchi

        As a major inhibitory neurotransmitter of the central nervous system in animals, ${\gamma}$-aminobutyric acid (GABA) has several physiological functions, such as anti-hypertensive, diuretic, tranquilizer and anti-stress effects in human. In order to determine strains with high GABA producing ability and glutamate decarboxylase (GAD) activity, 273 bacteria were isolated from various types of Kimchi. Strain K255 contained $386.37{\mu}g/mL$ of GABA in MRS broth containing 1% MSG, $600.63{\mu}g/mL$ of GABA in MRS broth containing 2% MSG and $821.24{\mu}g/mL$ of GABA in MRS broth containing 3% MSG. It showed that K255 had the highest GABA production ability compared to other commercial lactic acid bacteria. K255 was identified as Lactobacillus plantarum based on its API carbohydrate fermentation pattern and 16S rDNA sequence. K255 was investigated for its physiological characteristics. The optimum growth temperature of K255 was $37^{\circ}C$and cultures took 13 h to reach the pH 4.4. K255 showed more sensitive to bacitracin in a comparison of fifteen different antibiotics, and showed most resistance to kanamycin and vancomycin. Moreover, it was comparatively tolerant to bile juice and acid and displayed resistance to Escherichia coli, Salmonella Typhimurium, Staphylococcus aureus, with rates of 30.8%, 29.7%, and 23.4% respectively. These results demonstrate that K255 could be an excellent strain for the production of functional products.

        Physiological Characteristics and Anti-obesity Effect of Lactobacillus plantarum Q180 Isolated from Feces

        Obesity is strongly associated with several metabolic and chronic diseases and has become a major public health problem of worldwide concern. This study aimed to investigate the physiological characteristics and anti-obesity effects of Lactobacillus plantarum Q180. Lactobacillus plantarum Q180 was isolated from the faces of healthy adults and found to have a lipase inhibitory activity of $83.61{\pm}2.32%$ and inhibited adipocyte differentiation of 3T3-L1 cells ($14.63{\pm}1.37%$) at a concentration of $100{\mu}g/mL$. The strain was investigated for its physiological characteristics. The optimum growth temperature of L. plantarum Q180 was $37^{\circ}C$. Lactobacillus plantarum Q180 showed higher sensitivity to novobiocin in a comparison of fifteen different antibiotics and showed the highest resistance to rifampicin, polymyxin B and vancomycin. The strain showed higher ${\beta}$-galactosidase and N-acetyl-${\beta}$-glucosaminidase activities. It also did not produce carcinogenic enzymes such as ${\beta}$-glucuronidase. The survival rate of L. plantarum Q180 in MRS broth containing 0.3% bile was 97.8%. Moreover, the strain showed a 97.2% survival rate after incubation for 3 h in pH 2.0. Lactobacillus plantarum Q180 was displayed resistance to Escherichia coli, Salmonella Typhimurium and Staphylococcus aureus with rates of 55.6%, 38.0% and 47.6%, respectively. These results demonstrate that L. plantarum Q180 has potential as a probiotic with anti-obesity effects.
