        Stimulatory Activity of Lignans from <i>Machilus thunbergii</i> on Osteoblast Differentiation

        <P>Phytoestrogens are naturally occurring compounds exerting estrogenic activity, and include isoflavonoids, flavonoids and lignans. In the present study, we evaluated the stimulating activity of six lignans, meso-dihydroguaiaretic acid, nordihydroguaiaretic acid, machilin A, guaiacin, isoguaiacin and isoguaiacin dimethylether, from <I>Machilus thunbergii</I>, on osteoblast differentiation employing primary cultures of mouse osteoblast as an <I>in vitro</I> assay system. Among the six lignans tested, arylnaphthalene type lignans such as guaiacin, isoguaiacin and isoguaiacin dimethylether significantly increased alkaline phosphatase activity, whereas bibenzylbutane type lignans such as meso-dihydroguaiaretic acid, nordihydroguaiaretic acid and machilin A showed little effects. Isoguaiacin and isoguaiacin dimethylether also increased collagen synthesis as well as calcium deposition. In addition, treatment of the mouse osteoblasts with tamoxifen markedly reduced ALP activity increased by isoguaiacin or isoguaiacin dimethylether, suggesting the involvement of estrogen receptor in the action of these lignans on osteoblast differentiation. Taken together, these results suggest that arylnaphthalene type lignans such as guaiacin, isoguaiacin and isoguaiacin dimethylether significantly increase osteoblast differentiation.</P>

        추출 조건에 따른 오미자 열매 추출물의 lignan 성분들의 함량 분석 및 항산화 효과

        Schisandra chinensis (S. chinensis) contains several compounds that exhibit various physiological activities such as anti-oxidation and anti-inflammation. The major compounds of S. chinensis are lignans such as schisandrol A, schisandrol B, schisandrin A, gomisin N and schisandrin C. In this study, the contents of five lignans from S. chinensis was quantitatively analyzed by using high-performance liquid chromatography (HPLC) to find the optimal extraction method. Additionally, the analysis method was validated and a simultaneous analysis method was established. DPPH and ABTS scavenging assays were performed to evaluate the anti-oxidant effect of S. chinensis extracts and fractions. The results showed that the calibration curves of five lignans exhibited high linearity with a correlation coefficient (R²) of 0.9998. The optimal extraction method for S. chinensis was the reflux extraction method with 60% ethanol at 100℃ and the contents of five lignans were 1.73, 0.28, 0.85, 1.28 and 0.41%. The contents of five lignans in n-hexane fraction was 6.1 to 8.5 times compared with those in the extract prepared by optimal extraction method. The results of anti-oxidation of S. chinensis extracts and fractions showed that the anti-oxidant effect was increased dose-dependently depending on the contents of five lignans. Therefore, these results could be used as basic data for the standardization of anti-oxidant health functional foods, cosmetics and medicines including five lignans, which are the active compounds of S. chinensis.

        식이성 식물성 에스트로겐이 건강에 미치는 영향에 대한 관심은 호르몬 대체 요법이 과거에 생각되어 온 것처럼 안전하지도 효과적이지도 않다는 발견과 함께 날로 증대되고 있다. 본 종설에서는 식이성 식물성 에스트로겐의 원료인 isoflavonoids, stilbenes, coumestans 및 lignans에 대하여 요약·정리하였다. 특히 심혈관계에 미치는 영향에 관한 지금 까지 행해진 임상시험 결과를 정리하였다.

        리그난의 화학적 특성, 인체 건강에 미치는 영향 및 리그난의 우유 기능 강화에 관한 연구 고찰

        This review discusses the characteristics of major lignans and related studies and provides a basis for future studies. Lignans are present in various food products consumed daily, such as flaxseed and other seeds, vegetables, fruits, and beverages including coffee, tea, and wine. Lignans are natural phytoestrogens with a structure similar to that of secoisolariciresinol (Seco), mataireinol (Mat), pinoresinol (Pin), medioresinol (Med), lariciresinol (Lari), and syringaresinol, which is then converted to enterodiol (END) and enterolactone (ENL), which are mammalian lignans and the primary biologically active enterolignans, by the intestinal microflora. The associations between lignans and a decreased risk of cardiovascular disease are promising; however, they are not yet well-established, probably owing to low lignan intake in habitual Western diets. Nonetheless, these associations were more prominent at the higher doses in interventional studies. Many studies on humans and animals have reported the benefits of lignan consumption in protecting against CVD and metabolic syndrome by reducing lipid and glucose concentrations. END and ENL reportedly exert protective effects including phytoestrogenic, antioxidant, anti-inflammatory, and anticancer effects through various mechanisms. Moreover, lignans reportedly exert beneficial effects in breast, colon, and prostate cancer and osteoporosis have reported that. However, future studies are required to confirm the association between lignan and disease.

        토성에 따른 삼백초 생육특성과 유효성분 함량

        삼백초(三白草) 재배에 있어서 토성에 따른 생육과 유효성분을 분석하여 안정적인 재배법을 확립하고 고품질 삼백초를 생산하고자 시험을 수행한 결과를 요약하면 다음과 같다. 가. 삼백초의 절수와 엽수, 분지수는 토성 간 차이가 없었으며, 주당 줄기와 잎의 무게는 사양토가 가장 많고, 양토, 식양토, 사토의 순으로 무거웠다. 직경 5mm이하 뿌리는 토성 간차이가 없었으나, 5.1~10.9 mm의 굵기는 사양토, 양토 그리고 식양토에서 437~465 g로 차이가 없었고, 11 mm이상은 식양토가 많았다. 건물율은 19.1~20.8%로 차이가 없었다. 나. 잎의 quercetin-glycoside 함량은 양토와 사양토에서 219.3~222.4 mg/100 g로 가장 높았으며, quercitrin〉rutin〉isoquercitrin〉hyperin의 순으로 높았다. 줄기는 사토와 사양토가 각각 14.8 mg/100 g과 12.4 mg/100 g로 가장 많았으며, quercetin-glycoside 중 rutin이 가장 높았다. 다. 뿌리의 lignans 함량은 식양토, 양토, 사앙토, 사토의 순으로 높은 경향이었으며, lignans 관련 물질 중 manassatin B가 가장 많았다. This study was carried to investigate the effect of soil texture on the growth and the contents of quercetin-glycoside and lignans, and to improve the qualities of Saururus chinensis Baill. Soil texture resulted no significant effects on the number of nodes, the number of leaves, the number of branches and dry matter ratio. However, the shoot dry weight was higher in sandy loam, loam, silt loam and sand soil in that order. Although the weight of rhizomes of below 5 mm in diameter was not significantly different among soil textures, the weight of rhizomes between 5.1 and 10.9 mm and the weight of rhizomes of above 11 mm in diameter ranged 437~465 g and was larger in clay loam than in other soil textures. No significance difference was showed in rhizome dry ratio ranging from 19.1 to 20.8%. The amount of quercetin-glycoside in leaves was higher in loam and sandy loam and ranged from 219.3 to 222.4 mg/100 g of quercetin-glycoside quercitrin, rutin, isoquercitrin and hyperin were higher in that order. On the other hand, quercetin-glycoside contents in stem were 14.8 mg/100 g and 12.4 mg/100 g in sandy and sandy loam, respectively, and were higher than in other soil textures of quercetin-glycoside constituents, the content of rutin was the highest. The content of lignans was increased in clay loam, loam, sandy loam, and sandy in that order of lignans, the manassatin B was the highest.

        Cytotoxic lignans from Viburnum foetidum

        Two new lignans, 3,4,40-trihydroxy-30,9-dimethoxy-9,90-epoxylignan (1), 3,40-dihydroxy-30,4, 9-trimethoxy-9,90-epoxylignan (2), together with one known compound,4,40-dihydroxy-3,30,9-trimethoxy-9,90-epoxylignan(3), were isolated from the 95 % EtOH extract of Viburnumfoetidum. The structures of the two new compounds wereelucidated on the basis of 1D, 2D-NMR, and mass spectralanalysis. All the lignans were in vitro evaluated for theircytotoxic activities against four tumor cell lines (A549, SKOV-3, SKMEL-2 and HCT15).

        <P><B>Abstract</B></P><P>Three new furofuran lignans, (+)‐4,4′‐<I>O</I>‐diangeloylpinoresinol (<B>1</B>), (+)‐4,4′‐<I>O</I>‐diangeloylmedioresinol (<B>2</B>), and (+)‐4,4′‐<I>O</I>‐diangeloylsyringaresinol (<B>3</B>), together with the known compound (+)‐syringaresinol, were isolated from the MeOH extract of <I>Rudbeckia laciniata.</I> The structure elucidation of these compounds were based on 1D‐ and 2D‐NMR, and HR‐ESI‐MS data. The additional structural evidence was obtained from alkaline hydrolysis of the compounds.</P>

        <P>Four new sesqui-lignans, (7R, 7'R, 7 '' S, 8S, 8'S, 8 '' S)-4',5 ''-dihydroxy-3,5,3',4 ''-tetramethoxy-7,9':7',9-diepoxy4,8 ''-oxy-8,8'-sesquineo-lignan-7',9 ''-diol (1), (7R, 7'R, 7 '' S, 8S, 8'S, 8 '' S)-4',3'-dihydroxy-3,5,3',5',4'pentamethoxy-7,9':7',9-diepoxy-4,8'-oxy-8,8'-sesquineo-lignan-7',9'-diol (2), (7R, 7'R, 7'S, 8S, 8'S, 8'S)-3',4'dihydroxy-3,5,4',5 ''-tetramethoxy-7,9':7',9-diepoxy-4,8'-oxy-8,8'-sesquineo-lignan-7',9'-diol (3) and acanthopanax A (7) together with three known compounds (4-6) were isolated from the EtOAc-soluble extract of Acanthopanax senticosus. Their structures were elucidated on the basis of spectroscopic and physicochemical analyses. All the isolates were evaluated for in vitro inhibitory activity against DGAT1 and DGAT2. Among them, compounds 1-6 were found to exhibit selective inhibitory activity on DGAT1 with IC50 values ranging from 61.1 1.3 to 97.7 1.1 111\4 and compound 7 showed selective inhibition of DGAT2 with IC50 value 93.2 1.2.         NMR Assignments of Two Furofuran Lignans from Sesame Seeds

        Two furofuran lignans, sesamolin and sesangolin were isolated from the seeds of Sesamum indicum and S.angolense, respectively. Detailed analysis of the $^1H-and^{13}C-NMR$ spectra of these lignans was carried out by the application of two-dimensional $^1H-^1/H\; COSY\; and^1/H^{13}C$ multiple-bond, multiple-quantum spectroscopic correlation techniques.

        Dietary Intake of Polyphenols and the Risk of Breast Cancer: a Case-Control Study

        Despite mounting evidence that dietary polyphenols might have a protective role against the risk of breast cancer (BC), few studies have assessed the relationship between intake of polyphenol classes and subclasses with BC. Thus, we examined the relationship between dietary polyphenol classes and individual polyphenol subclasses and the risk of BC. Overall, 134 newly diagnosed BC patients and 267 healthy hospitalized controls were studied. Dietary intake was assessed using a validated 168-item food frequency questionnaire (FFQ). To estimate dietary intake of polyphenols, polyphenol content (flavonoids, lignans, stilbenes and phenolic acids) of 80 food items were derived from an updated version of the phenol explorer database containing information on the effects of food processing on polyphenol content. The dietary polyphenol intake was calculated by matching the subjects' food consumption data with our polyphenol content database. Multivariate logistic regression was used to estimate odds ratios (ORs) and 95% confidence intervals (CIs). Controls had higher intake of total polyphenol (marginally significant; p = 0.07), hydroxycinnamic acid (marginally significant; p = 0.05) and lignan (p = 0.01). After adjusting for potential confounders, high consumption of lignans (highest vs. lowest tertile: OR, 0.51; 95% CI, 0.26–0.97; p for trend = 0.04) associated with decreased risk of BC. There was no significant relationship between intake of other polyphenols and risk of BC. Our findings suggest that high lignan intake is associated with a reduced risk of BC.
