        Expression of FKBP prolyl isomerase 5 gene in tissues of muscovy duck at different growth stages and its association with muscovy duck weight

        Objective: FKBP prolyl isomerase 5 (FKBP5) has been shown to play an important role in metabolically active tissues such as skeletal muscle. However, the expression of FKBP5 in Muscovy duck tissues and its association with body weight are still unclear. Methods: In this study, real-time quantitative polymerase chain reaction was used to detect the expression of FKBP5 in different tissues of Muscovy duck at different growth stages. Further, single nucleotide polymorphisms (SNPs) were detected in the exon region of FKBP5 and were combined analyzed with the body weight of 334 Muscovy ducks. Results: FKBP5 was highly expressed in various tissues of Muscovy duck at days 17, 19, 21, 24, and 27 of embryonic development. In addition, the expression of FKBP5 in the tissues of female adult Muscovy ducks was higher than that of male Muscovy ducks. Besides, an association analysis indicated that 3 SNPs were related to body weight trait. At the g.4819252 A>G, the body weight of AG genotype was significantly higher than that of the AA and the GG genotype. At the g.4821390 G>A, the genotype GA was extremely significantly related to body weight. At the g.4830622 T>G, the body weight of TT was significantly higher than GG and TG. Conclusion: These findings indicate the possible effects of expression levels in various tissues and the SNPs of FKBP5 on Muscovy duck body weight trait. FKBP5 could be used as molecular marker for muscle development trait using early marker-assisted selection of Muscovy ducks.         토종오리의 교배 조합이 오리고기의 부분육 생산수율, 육질 및 관능검사에 미치는 영향

        본 시험은 토종오리와 육용오리의 교배 조합이 부분육 비율과 육질에 미치는 영향을 연구하기 위해 수행하였다. 토종오리와 육용오리의 교배 조합으로 발생된 4처리구의 오리 360수를 평사에서 사육하고, 일정 주령(A, B, C 8주령, D 6주령)에 도달하였을 때 각 처리구에서 암수 각각 9수씩 선별하였다. 교배 조합 방법은 토종오리(♀)×토종오리(♂), 육용오리(♀)×토종오리(♂), 토종오리(♀)×육용오리(♂), 육용오리(♀)×육용오리(♂)의 4처리구를 각각 A, B, C, D로 하였다. 선별된 개체들은 도압(屠鴨)하여 부분육 비율을 측정하고, 오리고기의 이화학적 성상과 관능검사를 실시하였다. 날개와 목 부위의 비율은 D 처리구가 가장 낮았으며(P<0.05), 가슴 부위는 B 처리구에서 가장 높았다(P<0.05). 다리 부위는 처리구간 차이가 없었으며, 부분육을 제외한 부위는 A, B, D 처리구에서 C 처리구에 비해 유의적으로 높았다(P<0.05). 오리고기의 pH는 처리구간 차이가 없었으나(P>0.05), 지방, 단백질 함량은 B 처리구가 가장 높았으며(P<0.05), 콜라겐 함량은 D 처리구가 가장 높았다(P<0.05). 오리고기의 육색과 물리학적 성상(전단력, 가열 감량, 보수력)은 처리구간 차이가 없었다(P>0.05). 오리고기의 관능검사는 다즙성에서 D 처리구가 가장 높았으나(P<0.05), B와 D 처리구 사이에는 유의적 차이가 없었다. 따라서 토종오리와 육용오리의 교배 조합은 이화학적 성상이나 관능검사의 개선 효과가 뚜렷하지 않지만 가슴육 수율 향상에 영향을 주는 것으로 사료된다. This work was carried out to investigate on the retail cut yield and the meat quality of crossbred ducks. A total of 360 pullets that were produced from 4 mating methods used in this work. Four mating method were A) native ducks (♀) × native ducks (♂), B) meat-type ducks (♀) × native ducks (♂), C) native ducks (♀) × meat-type ducks (♂), and D) meat-type ducks (♀) × meat-type ducks (♂). Ducks were bred at the flat house, and selected nine ducks with similar weights from each treatment at the certain weeks (A, B, C 8 weeks; D 6 weeks). Selected ducks were slaughtered, calculated the retail cut yield(wing, back, neck, breast, leg), analyzed the physico-chemical compositions, and tested the sensory evaluation. Wing and neck meat ratios of D treatment were lowest (P<0.05), and breast meat ratio of B treatment was high (P<0.05) compared to other treatments. pH of duck meat was no difference among treatments, fat and protein contents of B treatment was highest, and collagen contents of D treatment was highest among all treatments. Meat color and physical compositions was no difference among treatments. Juiciness of D treatment meats was highest, and there was no difference between B and D treatments. Finally, mating method of native duck and meat-type duck affected on the retail cut yield, but did not improve on the physico-chemical compositions and sensory evaluation.

        Use of Duckweed as a Protein Supplement for Breeding Ducks

        An experiment was conducted at the experimental duck farm of Cantho University to determine the effects of feeding duckweed (Lemna minor) that replaced commercial protein supplements in diets for local and Cherry Valley breeding ducks. The experiment included a total of 180 ducks, with five treatments and three replicates and six breeding ducks (one male plus five females together) per replicate, for both local and exotic Cherry Valley ducks. The five diets were fed ad libitum and were based on rice byproducts supplemented with roasted soya bean meal plus dried fishmeal at levels of 100% (SF100, control), 75 (SF75), 50 (SF50), 25 (SF25) or zero (SF0) % of the protein in the control diet, corresponding to 18, 15, 13, 10 and 8% CP in the diets for both breeds, respectively. Fresh duckweed was supplied ad libitum for all treatments. These diets were fed to local breeding ducks from 7 to 12 months of age, and to exotic breeding ducks (Cherry Valley) from 8 to 13 months of age. Total mean daily dry matter (DM) intakes were 183, 178, 176, 177 and 174 g (p<0.05) for the local ducks, and 221, 208, 215, 219 and 210 g (p<0.01) for the exotic ducks for the SF100 (control), SF75, SF50, SF25 and SF0 diets, respectively. Laying rates of the local ducks were 66.5, 65.2, 62.9, 63.1 and 62.3%, and of the Cherry Valley ducks 61.9, 58.4, 58.9, 59.1 and 53.5% (p<0.001) for the control (SF100), SF75, SF50, SF25 and SF0 treatments, respectively. Fertile egg rates were 95.6, 95.6, 97.8, 97.8 and 92.2%, and hatchabilities 89.4, 80.6, 87.2, 88.6 and 77.8% (p<0.05) for the local breed, and 97.8, 97.8, 91.1, 92.2 and 90.0% (p<0.05) and 72.8, 74.7, 75.0, 74.3 and 76.7% for the Cherry Valley ducks for diets SF100, SF75, SF50, SF25 and SF0, respectively. Corresponding feed conversion ratios (dry matter basis) were 3.83, 3.82, 3.89, 4.01 and 3.96 kg feed per kg egg mass for the local ducks and 4.52, 4.56, 4.58, 4.73 and 5.02 kg feed per kg egg mass for the Cherry Valley ducks for the SF100, SF75, SF50, SF25 and SF0 treatments, respectively. Replacement of 100% of the protein supplement by fresh duckweed in the diets of the local laying ducks decreased the feed costs by 25% compared to the control diet.

        Comparison of Meat Characteristics between Korean Native Duck and Imported Commercial Duck Raised under Identical Rearing and Feeding Condition

        This research was conducted to compare the meat characteristics of Korean native duck and imported commercial duck. The Korean native ducks and imported commercial ducks (broiler duck: Grimaud) were raised under identical rearing and feeding conditions for 8 wk and 6 wk, respectively. At the end of the rearing period, ten ducks from each group were slaughtered, and breast and leg meat were obtained 24 h after slaughtering for analyses of meat characteristics. The results showed that the breast of Korean native ducks contained lower moisture and fat, and higher protein and water holding capacity (WHC) than those of imported commercial ducks (p<0.05). The breast of Korean native ducks also had higher CIE $a^*$ and lower CIE $L^*$ (p<0.05). After cooking, the breast meat of Korean native ducks had higher shear-force, sensory attributes of texture, taste and overall likeness (p<0.05). Also, the breast meat of Korean native ducks contained a higher percentage of palmitic acid ($C_{16:0}$) and arachidonic acid ($C_{20:4}$) (p<0.05) than those of imported commercial ducks. Furthermore, the leg meat of Korean native ducks contained higher percentages of total unsaturated fatty acid and lower percentages of total saturated fatty acid (p<0.05). It is concluded that the meat from Korean native ducks, especially breast meat, had better quality parameters and contained higher amounts of unsaturated fatty acids.

        COMPARATIVE STUDIES ON THE UTILIZATION OF CALCIUM BETWEEN LAYING TSAIYA DUCK AND LEGHORN HEN

        Studies were conducted to compare the utilization of dietary calcium between brown laying Tsaiya duck and Leghorn hen. Birds were fed corn-soybean diets containing 1.0, 2.0, 3.0, 4.0 and 5.0% calcium, respectively, with five birds per treatment. The metabolizability of calcium was determined by the indicator method. Experiments were conducted three times each at the age of 27, 31 and 36 weeks. Results appeared that duck eggs were heavier with better shell quality in comparison with hen eggs. Ducks fed 1% calcium diet resulted in severely depressed egg production (19%), but not for hens which still had 56% egg production. The metabolizability of calcium for hens was significantly higher than that for ducks (P<0.05) when both were fed diets containing 1 or 2% calcium. As the dietary calcium level was increased to 3 to 5%, there was no significant difference in calcium metabolizability between ducks and hens. When the dietary calcium was between 2 to 5%, the ducks retained more calcium than did the hens. Plasma calcium content for both hens and ducks fed 1% calcium diet was about the same. When the amount of the dietary calcium was increased to 2-5%, the plasma calcium level of ducks was approximately 7-10 mg/dl higher than that of hens. The calcium content in the egg shell of duck was significantly higher than that of hens, too. As the dietary calcium level was increased, there was a decreased magnesium content in the eggshell of hens, but not for ducks. The magnesium level in the eggshell was higher in hens than that in ducks. It is concluded that ducks could retain significantly more calcium and maintain higher plasma calcium level which might be the reason for larger eggs with better shell quality by ducks.

        Effects of age and diet forms on growth-development patterns, serum metabolism indicators, and parameters of body fat deposition in Cherry Valley ducks

        Objective: This study was conducted to investigate the effects of age and diet forms on growth-development patterns, serum metabolism indicators, and parameters of body fat deposition in Cherry Valley ducks. Methods: According to the hatching age and initial weight, a total of 150 1-day-old male SM3 Cherry Valley ducks were randomly assigned to two diet forms (pellet vs powder form). Each treatment had with 5 replicates per treatment and 15 meat ducks per replicate. The study lasted 42 d, which was divided into two periods (1 to 21 vs 22 to 42 d). Results: Our results showed that compared with powder group, ducks in pellet group had greater growth performance during different period (p<0.05). The inflection point was 24 d and was not numerically affected by diet forms. Increasing age (42 vs 21 d) significantly increased the weight of body fat and hepatic fat metabolism related enzyme activities in ducks (p<0.05), meanwhile, increasing age (42 vs 21 d) improved serum metabolism indicators and decreased mRNA expression levels of fat metabolism-related genes in liver (p<0.05). Ducks fed different diets (pellet vs powder form) increased growth performance as well as the weight of body fat and improved serum metabolism indicators (p<0.05). In addition, interactions were found between age and diet forms on the levels of serum metabolism indicators in ducks (p<0.05). Conclusion: In conclusion, powder feed reduced growth performance of ducks, and the day of inflection point was 24 days old. Ducks with higher age or fed with pellet diet showed higher fat deposition.         Plasma metabolites associated with physiological and biochemical indexes indicate the effect of caging stress on mallard ducks (Anas platyrhynchos)

        Objective: Cage rearing has critical implications for the laying duck industry because it is convenient for feeding and management. However, caging stress is a type of chronic stress that induces maladaptation. Environmental stress responses have been extensively studied, but no detailed information is available about the comprehensive changes in plasma metabolites at different stages of caging stress in ducks. We designed this experiment to analyze the effects of caging stress on performance parameters and oxidative stress indexes in ducks. Methods: Liquid chromatography tandem mass spectrometry (LC/MS-MS) was used to determine the changes in metabolites in duck plasma at 5 (CR5), 10 (CR10), and 15 (CR15) days after cage rearing and traditional breeding (TB). The associated pathways of differentially altered metabolites were analyzed using Kyoto encyclopedia of genes and genomes (KEGG) database. Results: The results of this study indicate that caging stress decreased performance parameters, and the plasma total superoxide dismutase levels were increased in the CR10 group compared with the other groups. In addition, 1,431 metabolites were detected. Compared with the TB group, 134, 381, and 190 differentially produced metabolites were identified in the CR5, CR10, and CR15 groups, respectively. The results of principal component analysis (PCA) show that the selected components sufficiently distinguish the TB group and CR10 group. KEGG analysis results revealed that the differentially altered metabolites in duck plasma from the CR5 and TB groups were mainly associated with ovarian steroidogenesis, biosynthesis of unsaturated fatty acids, and phenylalanine metabolism. Conclusion: In this study, the production performance, blood indexes, number of metabolites and PCA were compared to determine effect of the caging stress stage on ducks. We inferred from the experimental results that caging-stressed ducks were in the sensitive phase in the first 5 days after caging, caging for approximately 10 days was an important transition phase, and then the duck continually adapted.         MicroRNA analysis reveals the role of miR-214 in duck adipocyte differentiation

        Objective: Fat deposition in poultry is an important factor in production performance and meat quality research. miRNAs also play important roles in regulating adipocyte differentiation process. This study was to investigate the expression patterns of miRNAs in duck adipocytes after differentiation and explore the role of miR-214 in regulating carnitine palmitoyltransferases 2 (CPT2) gene expression during duck adipocyte differentiation. Methods: Successful systems for the isolation, culture, and induction of duck primary fat cells was developed in the experiment. Using Illumina next-generation sequencing, the miRNAs libraries of duck adipocytes were established. miRanda was used to predict differentially expressed (DE) miRNAs and their target genes. The expression patterns of miR-214 and CPT2 during the differentiation were verified by quantitative real-time polymerase chain reaction and western blot. Luciferase reporter assays were used to explore the specific regions of CPT2 targeted by miR-214. We used a miR-214 over-expression strategy in vitro to further investigate its effect on differentiation process and CPT2 gene transcription. Results: There were 481 miRNAs identified in duck adipocytes, included 57 DE miRNA candidates. And the 1,046 targets genes of DE miRNAs were mainly involved in p53 signaling, FoxO signaling, and fatty acid metabolism pathways. miR-214 and CPT2 showed contrasting expression patterns before and after differentiation, and they were selected for further research. The expression of miR-214 was decreased during the first 3 days of duck adipocytes differentiation, and then increased, while the expression of CPT2 increased both in the transcriptional and protein level. The luciferase assay suggested that miR-214 targets the 3’untranslated region of CPT2. Overexpression of miR-214 not only promoted the formation of lipid droplets but also decreased the protein abundance of CPT2. Conclusion: Current study reports the expression profile of miRNAs in duck adipocytes differentiated for 4 days. And miR-214 has been proved to have the regulator potential for fat deposition in duck. This study was to investigate the expression patterns of miRNAs in duck adipocytes after differentiation and explore the role of miR-214 in regulating carnitine palmitoyltransferases 2 (CPT2) gene expression during duck adipocyte differentiation.Methods: Successful systems for the isolation, culture, and induction of duck primary fat cells was developed in the experiment. Using Illumina next-generation sequencing, the miRNAs libraries of duck adipocytes were established. miRanda was used to predict differentially expressed (DE) miRNAs and their target genes. The expression patterns of miR-214 and CPT2 during the differentiation were verified by quantitative real-time polymerase chain reaction and western blot. Luciferase reporter assays were used to explore the specific regions of CPT2 targeted by miR-214. We used a miR-214 over-expression strategy in vitro to further investigate its effect on differentiation process and CPT2 gene transcription.Results: There were 481 miRNAs identified in duck adipocytes, included 57 DE miRNA candidates. And the 1,046 targets genes of DE miRNAs were mainly involved in p53 signaling, FoxO signaling, and fatty acid metabolism pathways. miR-214 and CPT2 showed contrasting expression patterns before and after differentiation, and they were selected for further research. The expression of miR-214 was decreased during the first 3 days of duck adipocytes differentiation, and then increased, while the expression of CPT2 increased both in the transcriptional and protein level. The luciferase assay suggested that miR-214 targets the 3’untranslated region of CPT2. Overexpression of miR-214 not only promoted the formation of lipid droplets but also decreased the protein abundance of CPT2.Conclusion: Current study reports the expression profile of miRNAs in duck adipocytes differentiated for 4 days. And miR-214 has been proved to have the regulator potential for fat deposition in duck.

        신에게 갈 수 없는 생선 - 물천구

        거칠고 험한 바다에서의 조업을 통해 생계를 이어가는 어민들은 자연환경의 변화에 민감하게 반응하며 자연에 대한 경외감을 가지고 다양한 종교활동과 의례활동을 행한다. 의례과정에서 어민들은 신의 은총에 대한 감사의 표현으로 수산물 등을 제물로 바친다. 그런데 모든 수산물이 제물로 사용되는 것은 아니다. 그들은 그 자체의 속성 또는 관습적 이유 때문에 일부 수산물을 제물로 사용하지 않는다. 베트남 섬선시 어민들도 자신이 잡은 수산물 중 물천구라는 생선을 신에게 바치지 않는다. 현재 물천구는 겨울철 어민들의 주요한 수입원이며 식생활에서도 중요한 위치를 점하고 있지만 의례과정에서는 금기시되고 있다. 본 논문에서는 의례과정에서 나타나는 물천구에 대한 금기와 그 현상에 대한 어민의 설명을 구성하는 사회경제적 맥락에 대한 분석을 통해 의례행위에 대한 설명이 사회적으로 구성되는 과정을 명확히 하고자 한다. 베트남 섬선시 어민들은 물천구를 신에게 바치지 않는 이유에 대하여 세 가지 방식으로 설명한다; “우리들이 전부 먹어버렸기 때문에 신에게 바칠 것이 없다.” “못생겼기 때문에 바치지 않는다.” “바친 이후에 먹으면 식어서 맛이 없기 때문에 바치지 않는다.” 섬선시 어민들이 물천구를 제물로 사용하지 않는 행위가 사회적으로 구성된 것과 마찬가지로 금기에 대한 그들의 설명도 사회적으로 구성된 것이다. 어민들은 물천구를 먹기 전부터 크기와 관계없이 신에게 바치지 않았다. 물천구는 못생겼고 불결하다고 여겨졌기 때문에 신성한 의례과정에 들어갈 수 없었다. 그런데 1990년대 이후 물천구에 대한 수요가 증가하기 시작했으며, 현재 그것은 어민들의 식생활과 경제생활에서 중요한 위치를 점하고 있다. 어민들은 이러한 현재적 상황에 비추어 의례과정에서 물천구를 제물로 사용하지 않는 것에 대하여 설명한다. 못생겼다고 하는 상징적 요인에 의해 그것에 대한 금기가 만들어졌지만, 현재는 많은 사람들이 먹는다는 사실과 식으면 맛이 없다고 하는 경험적 사실에 의해 그러한 금기 행위가 재생산되고 있다. 1990년대 이후 물천구는 경제적으로 높은 지위를 점하게 되었지만, 종교적으로는 여전히 낮은 위치에 있다. Fishermen, who earn their livelihood through fishing at rough sea, are sensitive to changes in the natural environment and do religious ceremonies. In the process of ceremonies fishermen offer something to God to express thanksgiving. Because of conventional reasons or its characteristics, people do not offer food to God. Vietnamese fishermen also do not offer food to God. In Samson city, Vietnam, bombay duck is an important source of income of fishermen and holds a important position in the fisherman’s diet. Although bombay duck has a high economic value, it is treated as having no value in ceremonies. Why do fishermen not offer Bombay duck to God? How do fishermen explain that? In this paper, I delineate the relationship of God, fishermen, and Bombay duck, and discuss socio-economical characteristics of this relationship. Fishermen give three explanations for why they do not offer Bombay duck to God: “because we eat all, we do not have any bombay duck to offer to God”, “because that fish is ugly, we do not offer it”, and “after ceremonies, bombay duck gets cold and is tasteless, so we do not offer it.” These explanations reflect the socio-economical changes that surround bombay duck. Now, bombay duck holds an important position in the fisherman’s diet and economic life, and the fishermen explain their ceremonial activities in the light of these conditions. Meanwhile, the economic value and religious value change differently over time. The economic value of bombay duck changes faster than the religious value. Fishermen did not eat bombay duck, they did not offer it to God, and bombay duck did not have economic value until the 1990s. After that, a large number of fishermen started eating bombay duck and it got a high economic value. Nevertheless, bombay duck is still regarded as bad-looking and fishermen do not offer it to God.
