        Desmin Binding Property of Nebulin Isoforms

        Nebulin is a giant ($600{\sim}900$ kDa), modular sarcomeric protein proposed to regulate the assembly, and to specify the precise lengths of actin filamints in vertebrate skeletal muscles. Recently, There is an evidence that the nebulin also expressed in non muscle tissue, brain and liver. We identified a new isoform of nebulin from adult brain library by PCR screening. It contains two simple-repeats exon 165, 166 and linker-repeats exon $154{\sim}161$ except exon 159. The nebulin modules M160 to M170 (exon 150 to exon 161) has been shown to bind desmin. In mature striated muscle, desmin intermediate filaments surround Z-discs and link individual myofibrils laterally at their Z-discs and to other intracellular structures, including the costameres and the intercalated discs of the sarcolemma, sarcoplasmic reticulum, mitochondria, T-tubules, and nuclei. Therefore, it is an interesting possibility that the differential splice pathways within the linker region of nebulin modify the affinity of nebulin's interaction with desmin. The specific interactions of nebulin and desmin were confirmed in vivo by yeast two hybrid experiments. To verify in the cellular level the interaction between nebulin isoform and desmin, we transfected COS-7 cell with EGFP-tagged nebulin and DsRed-tagged desmin. Based on evidence showing that despite exon 159 was deleted, the new isoform of nebulin was interact with desmin. This suggest that nebulin in brain may interact with another intermediate filament. The conservation of these ligand-binding capacity in brain and skeletal nebulins suggest that nebulins may have conserved roles in brain and skeletal muscle.

        Background : The aim of this report is to show that treadmill running exercise under well-controlled conditions is to improve of regeneration in rat gastrocnemius muscles after sciatic nerve crushing injury. Material & Method : Male Sprague-Dawley rats (1 month old) were submitted to bouts of exercise on a treadmill up a 10 degrees decline and speed is 24m/min for 60 min per day and gastrocnemius muscles were analysed at different exercise periods (5, 10, 15, and 20 days) by immunohistochemistry in comparison with injured non-exercised muscles. Rats were sacrificed at 9th (5 days exercise), 16th (10 days exercise), 23rd (15 days exercise), 30th day (20 days exercise) after sciatic nerve crushing injury. The regenerative processes were analysed by detection of routine immunoreactivity for the two intermediate filaments, desmin and vimentin. Desmin and vimentin are specific components of the cytoskeleton of striated muscle fibers and of mononuclear cells of mesenchymal origin including myoblasts, respectively. Results : Desmin was revealed in processes of degeneration and regeneration and vimentin was revealed in regeneration process. Non-exercised rats had more desmin- and vimentin-positive myofibers than that of treadmill exercised rats at 9th, 16th, 23rd, and 30th day after sciatic nerve crushing injury. At 30th day, non-exercised rats had several desminand vimentin-positive myofibers, but exercised rats had numerous normal myofibers. Conclusion : It is concluded that treadmill running exercise is able to improve regeneration processes in gastrocnemius muscles after sciatic nerve crushing injury.

        Primary leiomyoma of the ovaries in a dog

        We would like to report a case of leiomyoma of the ovaries in a dog. Leiomyoma is commonly seen in the vagina in dogs. However, it is a very rare neoplasm in the ovaries. As there have only been a few reported cases, this report provides valuable information on veterinary medicine and pathology. Masses found in the ovaries need to be differentiated from other ovarian tumors. Therefore, we describe the gross, histopathological, and immunohistochemical features of a case of ovarian leiomyoma in a 10-year-old female Yorkshire Terrier dog. The mass on the right of the uterus was found accidentally by pelvic ultrasonography. Laparatomy revealed a large multi-nodulated ovarian mass. Grossly, cut surfaces of the mass showed multiple firm whitish nodules in the cortex and bloody loose connective tissue in the medulla. Histopathologically, the cortex of the mass was composed of spindle cells forming interlacing fascicles. The cells had elongated, blunt-ended nuclei and eosinophilic cytoplasm as detected by hematoxylin and eosin staining. Immunohistochemical stained sections were immunoreactive for α-smooth muscle actin and desmin but negative for vimentin and S-100. Therefore, differential diagnosis confirmed leiomyoma based on morphology and positive staining for α-smooth muscle actin and desmin.

        Chronic pyometra due to bilateral ovarian tumors in a South American sea lion

        A 14-year-old female South American sea lion (Otaria byronia) with persistent vaginal secretion and chronic hemorrhagic diarrhea was encountered. During postmortem examination, the uterus was found to resemble a balloon with mucosal congestion and was filled with grayish milky material. The ovaries also had abnormal features, including necrotic surface lesions and multiple whitish foci in the cut section. Hemorrhages and ulcerated changes due to toxemia were observed in other organs, including the liver, spleen, lung, intestines, and lymph nodes. Microscopically, the left ovary contained interlacing fascicles of fibroblast-like cells with blunt-end nuclei showing cytoplasmic positive immunoreactivity against alpha-smooth muscle actin and desmin. The right ovary contained cells with round to cigar-shaped nuclei showing cytoplasmic positive immunoreactivity against vimentin. In conclusion, based on classification of bilateral ovarian tumors as a leiomyoma in the left region and a fibroma in the right region, this sea lion was diagnosed with chronic closed pyometra.

      • Desmin expression profile in reactive astrocytes in the 3-nitropropionic acid–lesioned striatum of rat: Characterization and comparison with glial fibrillary acidic protein and nestin

        <P><B>Abstract</B></P> <P>Desmin, a muscle-specific, type-III intermediate-filament protein, is reportedly expressed in astrocytes in the central nervous system. These cells become reactive astrocytes in response to brain injuries. To elucidate whether desmin is involved in this process, we examined the spatiotemporal expression profiles of desmin and their relationship with two astroglial intermediate filaments, glial fibrillary acidic protein (GFAP) and nestin, in the striatum of rats treated with the mitochondrial toxin 3-nitropropionic acid (3-NP). Weak, constitutive immunoreactivity for desmin was observed in astrocytes generally, and in reactive astrocytes in the peri-lesional area, its expression increased in parallel with that of GFAP over 3 d post-lesion and was maintained until at least day 28. Desmin, GFAP, and nestin showed characteristic time-dependent expression patterns in reactive astrocytes forming the astroglial scar; delayed and long-lasting induction of desmin and GFAP, and rapid but transient induction of nestin. In the lesion core, desmin was expressed in two categories of perivascular cells: nestin-negative and nestin-positive. These findings show that desmin, together with GFAP and nestin, is a dynamic component of intermediate filaments in activated astroglia, which may account for the dynamic structural changes seen in these cells in response to brain injuries.</P> <P><B>Highlights</B></P> <P> <UL> <LI> We show temporal regulation of desmin, GFAP, and nestin in the lesioned striatum. </LI> <LI> Desmin expression is astrocyte-specific and coincides with GFAP. </LI> <LI> The astroglial scar shows long-lasting desmin and GFAP expression. </LI> <LI> Nestin is expressed early but transiently in astroglial scar formation. </LI> <LI> Desmin is a dynamic component of intermediate filaments in activated astroglia. </LI> </UL> </P>

        Proteomic Analysis of Differentially Expressed Proteins in Bovine Endometrium with Endometritis

        Endometritis is one of the primary reasons for reproductive failure. In order to investigate endometritis-associated marker proteins, proteomic analysis was performed on bovine endometrium with endometritis. In bovine endometritis, desmin, α-actin-2, heat-shock protein (HSP) 27, peroxiredoxin-6, luteinizing hormone receptor isoform 1, collectin-43 precursor, deoxyribonuclease-I (DNase-I), and MHC class I heavy chain (MHC-Ih) were up-regulated. In contrast, transferrin, interleukin-2 precursor, hemoglobin β subunit, and potassium channel tetramerisation domain- containing 11 (KCTD11) were down-regulated in comparison to normal endometrium. The proteomic results were validated by semiquantitative-PCR and immunoblot analysis. The mRNA levels of desmin, transferrin, α-actin-2, HSP27, KCTD11, and MHC-Ih were up-regulated by over 1.5-fold, and showed a pattern similar to their proteomic profiles. Desmin and α-actin-2 protein showed positive correlations between proteomic analysis and immunoblot analysis. These results suggest that desmin and α-actin-2 may play important roles in endometritis-related function, and could be useful markers for the diagnosis of bovine endometritis.

      • Immunohistochemical Study of Myogenic Sarcomas

        Using immunohistochemical staining with monoclonal antibodies to muscle cell components, histogenetical grouping and differential diagnosis of myogenic sarcoma could be possible. However immunohistochemical staining properties of myogenic sarcomas are not well defined in the formalin-fixed, paraffin-embedded tissue. We attempted to differentiate leiomyosarcoma from rhabdomyosarcoma. Twenty-nine formlin fixed myogenic sarcomas were investigated by means of immunohistochemical technique with the use of monoclonal antibodies panel against vimentin, desmin and myoglobin. In addition, the degree of differentiation of these sarcomas was investigated in connection with the immunoreactivity and intensity of immunohistochemical stains. The results were as follows; 1. Vimentin was detected in nine (60%) out of fifteen cases of leiomyosarcoma and in seven (50%) out of fourteen cases of rhabdomyosarcoma. 2. Desmin was detected in seven (47%) out of fifteen cases of leiomyosarcoma and in eight (57%) out of fourteen cases of rhabdomyosarcoma. 3. Myoglobin was detected in eleven (79%) out of fourteen cases of rhabdomyosarcoma. In all leiornyosarcomas, myoglobin was not detected. 4. The immunoreactivity and intensity of these sarcomas with anti-desmin and anti-myoglobin were related to the degree of tumor cell differentiation, but vimentin was more frequently detected in the poorly differentiated tumor cells. The above results suggest that demonstration of vimentin, desmin and myoglobin in the formalin-fixed, parnffin-embedded tissue may be a helpful adjunct tool in the diagiosis of myogenic sarcoma.
