      • Analysis of Blue Honeysuckle Binding Force During Harvest Period

        The purpose of this study is to understand the characteristics of proper binding force to harvest blue honeysuckle. After tracking such force over 16 days for four breeds of blue honeysuckle during the harvest period, we find that the binding force is mainly distributed in the range of 0.1 ~ 1.7N (P<0.05). This distribution range varies slightly between breeds: the picking forces of L.caerulea subsp.altaicai and Berel are greater than Bluebird and wild L.caerulea var.edulis of Changbai Mountain, and the binding forces of all breeds decrease over time with different slopes. Moreover, we also find that the binding force in the afternoon are significant larger than in the morning for the same day (P<0.05), and no significant difference is found between sunny and rainy days. We categorize factors affecting the binding force, such as testing day, morning and afternoon, relative humidity, temperature, weather conditions into environmental factors and time factor.

      • Tamoxifen과 blue honeysuckle 복합제제의 7일간 반복 투여가 tamoxifen의 약물동력학에 미치는 영향 연구

        Objectives : This study aims to evaluate the effects of co-administration of blue honeysuckle (BH) and tamoxifen on pharmacokinetics of tamoxifen after 7 days repeated administration, as a process of the comprehensive and integrative medicine. Materials and methods : After 7 day-repeated oral treatment of 4 mg/kg of tamoxifen or tamoxifen 0.4 mg/㎖ with BH 40, 20 and 10 mg/㎖ concentration mixed formulas, the plasma was collected at 30 min before administration, 30 min, 1, 2, 3.5, 4, 6, 8 and 24 hrs, respectively. Plasma concentrations of tamoxifen were analyzed using LC-MS/MS methods. PK parameters of tamoxifen (Tmax, Cmax, AUC, t<sub>1/2 </sub>and MRT<sub>inf</sub>) were analysis as compared witht amoxifen administered rats using noncompartmental PK data analyzer programs. Results : Although no significant changes on the tamoxifen plasma concentrations and PK parameters were demonstrated after end of 7 day-repeated oral administration of mixed formulas consisted of tamoxifen and BH 40 or 10 mg/㎖ as compared to tamoxifen 0.4 mg/㎖ treated rats, seven day-repeated oral administration of tamoxifen and BH 20 mg/㎖ mixed formula induced significant increase of plasma tamoxifen concentrations from 30 min to 3.5 hrs after end of administration with significant increases of Cmax as compared with tamoxifen 0.4 mg/㎖ single formula treated rats, not in AUC<sub>0-t</sub>, and AUC<sub>0-inf</sub>, respectively. Conclusion : Based on the results of the present study, 7 day-repeated oral administration of tamoxifen and BH 40, 20 or 10 mg/㎖ mixed formulas did not significantly influenced on the overall the oral bioavailability of tamoxifen, somewhat differed from the results of our previous single oral administration study, in which single oral administration of tamoxifen 0.4 mg/㎖ and BH 10 mg/㎖ mixed formulation induced decreases of the oral bioavailability of tamoxifen, but not in tamoxifen 0.4 mg/㎖ and BH 40 or 20 mg/㎖ mixed formulas.

        In this study, we demonstrated the anti-inflammatory activities of two kinds of blue honeysuckle (Lonicera caerulea var. edulis Turcz, Lonicera caerulea var. longibracteata f. ovata Hara) extracts in the LPS-stimulated macrophages. These two extracts showed non-cytotoxic activity, even at the high concentration. These two extracts of honeysuckle reduced the production of nitric oxide, cyclooxigenase-2 and pro-inflammatory cytokines. The inhibitory activities of two extracts of honeysuckle were found to be caused by blockage of NF-κB activation.

        Analgesic Effect of Blue Honeysuckle on the Rat Menopausal Pain, Primary Dysmenorrhea

        Objectives : We observed the possibilities that blue honeysuckle has favorable analgesic or refinement effects on the Primary dysmenorrhea (PD) in rats. Methods : Estradiol benzoate and oxytocin were used to induce the PD rat model. And Blue honeysuckle concentration lyophilized powders (BH) 500, 250 and 125 ㎎/㎏ and 500 ㎎/㎏ of Lonicerae Flos aqueous extract lyophilized powders (LF) were orally administered, once a day for 10 days at 30 min after each estradiol benzoate treatment. Then the changes on the body weights and gains during experimental periods, abdominal writhing response for analgesic activities, uterine weights, uterus lipid peroxidation, antioxidant defense system – glutathione contents, superoxide dismutase and catalase activities, NF-κB and COX-2 mRNA expressions were monitored with uterus histopathology including immunohistochemistry for tumor necrosis factor (TNF)-α and inducible nitric oxide synthase (iNOS).. Results : Inflammatory and oxidative stress mediated PD signs were favorably and dose-dependently inhibited by 10 days continuous oral administration of three different dosages of BH – 500, 250 and 125 ㎎/㎏ as comparable to those of indomethacin(IND) 5 ㎎/㎏ treated rats in BH 500 ㎎/㎏ administered PD rats, and similar to those of LF 500 ㎎/㎏ in BH 125 ㎎/㎏, at least in a condition of the present PD rat model. Conclusions : The results suggest that BH has favorable analgesic and refinement activities on the estradiol benzoate and oxytocin treatment-induced PD signs through anti-inflammatory and antioxidative potentials.

        Hepatoprotective Effect of Blue Honeysuckle on Rat Hypothyroidism

        This study was to determine whether blue honeysuckle lyophilized concentrated powder (BH) has positive effects on hypothyroidism and related liver damage in propylthiouracil-induced Hypothyroidism. Levothyroxine (LT4)-treated group was administrated by intraperitoneal injection with LT4, while BH or Flos Lonicerae lyophilized aqueous extract (LF)-treated groups were orally administrated for 42 days which two weeks pri or to PTU injection. The changes in organ weight, serum AST & ALT, serum lipid level, liver defense system were measured and the histopathology of the liver was observed. The oral administrations of 125, 250, and 500 mg/kg of BH showed favorable effects compared to LF on hypothyroidism-related liver damages through the enhance antioxidant defense system.

        Effects of the mixed formulation of sorafenib and blue honeysuckle on the pharmacokinetics profiles of sorafenib

        Objective : This study investigated the effects of concentrated and lyophilized blue honeysuckle powders (BH) on the pharmacokinetics (PK) of sorafenib were observed. Method : The blood was collected at 0.5 hr before single oral treatment of sorafenib (40 mg/kg) or sorafenib with BH (400, 200 and 100 mg/kg) mixed formulas administration, and 0.5, 1, 2, 3, 4, 6, 8 and 24 hrs after the end of single or mixed formula administration. Plasma concentrations of sorafenib were analyzed using LC-MS/MS methods. Tmax, Cmax, AUC, t1/2 and MRTinf of sorafenib were analysis as compared with sorafenib single treatment. Results : Single oral administration of mixed formulas induced significant increases of plasma sorafenib concentrations from 0.5 hr after end of administration throughout all blood collected time points, as compared with sorafenib single formula treated rats, and significant decreases of sorafenib Tmax with increases of Cmax, AUC0-t and AUC0-inf were detected in sorafenib and BH 400 mg/kg mixed formulation treated rats as compared with sorafenib single formula treated rats, respectively. Inaddition, sorafenib and BH 200 or 100 mg/kg mixed formula treated rats also showed significant increases of sorafenib Cmax, AUC0-t and AUC0-inf, respectively. Conclusions : According to these results, mixed formulation of BH with sorafenib increased the bioavailability of sorafenib through the increment of the absorptions.

        Protective Effects of Blue Honeysuckle on Rat Hypothyroidism Induced by Propylthiouracil

        The objective of the present study is to determine whether blue honeysuckle lyophilized concentrated powder(BH) has favorable effects on hypothyroidism and related reproductive organ damage. Hypothyroidism was inducedby 9 subcutaneous administration of propylthiouracil (PTU) for 28 days. Levothyroxine (LT4)-treated group wasintraperitoneally injected with LT4 for the same period, while for BH (125, 250, and 500 mg/kg) or Flos Loniceraelyophilized aqueous extract (LF, 250 mg/kg)-treated groups, the test materials were orally administrated for 42 days:two weeks before PTU injection and during PTU administration. The changes in serum thyroid hormone levels, serummale sex hormone levels, and testis antioxidant defense system were observed by histopathology of the thyroid gland,epididymis, prostate, and testis. The oral administrations of 125, 250, and 500 mg/kg of BH showed favorable effectscompared to LF on hypothyroidism and related damages of reproductive organs through augmentation of the antioxidantdefense system in the testis. In conclusion, BH is a promising new potent thyroid gland protecting agent.

        Effects of mixed formulation of tamoxifen and blue honeysuckle on the pharmacokinetics profiles of tamoxifen after single oral administration

        Objectives: Here, we investigated the effects of concentrated and lyophilized powders Blue honeysuckle (BH) on the PK of tamoxifen, to establish the pharmacokinetics (PK) profiles as one of essential process in new drug development. Methods: After single oral treatment of 0.4 mg/ml of tamoxifen or tamoxifen 0.4 with BH 40, 20 and 10 mg/ml, the plasma were collected at 0.5 hr before administration, 0.5, 1, 2, 3, 4, 6, 8 and 24 hr after end of single or mixed formula treatment. Plasma concentrations of tamoxifen were analyzed using LC-MS/MS methods. Tmax, Cmax, AUC, t1/2 and MRTinf were analyzed using noncompartmental PK data analyzer programs. Results: Tamoxifen and BH 40 mg/ml did not induce any significant change on the plasma tamoxifen concentrations, while significant decreases were observed in tamoxifen and BH 10 mg/ml from 2 to 8 hr as compared with tamoxifen only, respectively. Furthermore, significant increases of Tmax in tamoxifen and BH 40 mg/ml, significant decreases of Cmax in tamoxifen and BH 20 mg/ml, significant decreases of AUC0-t, AUC0-inf and MRTinf in tamoxifen and BH 10 mg/ml were demonstrated as compared with tamoxifen only. Conclusion: Taken together, tamoxifen and BH 10 mg/ml induced significant decrease of the oral bioavailability of tamoxifen, while tamoxifen and BH 40 or 20 mg/ml did not critically influenced, suggesting formulated BH concentration-independencies. It, therefore, seems to be needed that pharmacokinetic study after repeated administration should be tested to conclude the effects of BH on the pharmacokinetics of tamoxifen.

      • Anti-obesity and fatty liver-preventing activities of <i>Lonicera caerulea </i> in high-fat diet-fed mice

        <P>Blue honeysuckle (BH, <I>Lonicera caerulea</I>) is used as a traditional medicine in Russia, Japan and China, but is not commonly considered as an edible berry in Europe, USA or Korea. BH has been revealed to decrease serum cholesterol and triacylglycerol (triglyceride or TG) levels through the activation of AMP-activated protein kinase (AMPK), thus it is expected to be a health functional food and pharmaceutical agent for the prevention of non-alcoholic liver damage, in addition to effects as a suppressor of hyperlipidemia and as an anti-obesity agent. In the present study, the pharmacological activity of BH extract (BHe) was observed in high-fat diet (HFD)-fed mice. Significant increases in fat pad weight, body weight, fat accumulation (body and abdominal fat density, and thickness of the periovarian and abdominal wall) and serum biochemical levels (aspartate transaminase, alanine amino-transferase, alkaline phosphatase, lactate dehydrogenase, γ-glutamyltransferase, total cholesterol, low-density lipoprotein and TG, with the exception of high-density lipoprotein) were observed in HFD-fed mice. In addition, increases in adipocyte hypertrophy, the area of steatohepatitis and hepatocyte hypertrophy were observed, whereas decreased zymogen content was identified upon histopathological observation. Increased deterioration of the endogenous antioxidant defense system (liver catalase, glutathione and superoxide dismutase) and hepatic lipid peroxidation was observed. In addition, there were decreases in hepatic glucokinase activity, AMPKα1 and AMPKα2 mRNA expression, adipose tissue uncoupling protein 2 expression, and adiponectin mRNA expression, increases in phosphoenolpyruvate carboxykinase and glucose-6-phosphatase activity, hepatic acetyl-CoA carboxylase 1 mRNA expression, and the expression of leptin, CCAAT/enhancer-binding protein (C/EBP) α, C/EBPβ and sterol-regulatory-element-binding protein 1c mRNA in the periovarian tissue. Furthermore, non-alcoholic fatty liver disease (NAFLD) and obesity were significantly inhibited by the continuous administration of BHe for 84 days. These results revealed that BHe may be a promising novel drug or functional food candidate for the treatment of obesity and NAFLD.</P>
