        Anti-Wrinkle Activity of Ziyuglycoside I Isolated from a <i>Sanguisorba officinalis</i> Root Extract and Its Application as a Cosmeceutical Ingredient

        <P>In order to investigate the potential of a <I>Sanguisorba officinalis</I> root extract as an active ingredient for wrinkle-care cosmetics, we measured its free radical scavenging activity, elastase inhibitory activity, expression of MMP-1 (matrix metalloprotease-1) <I>in vitro</I>, and type I collagen synthesis in normal human fibroblast cells. To isolate the main components from the <I>S. officinalis</I> root extract, we purified the extract by solvent fractionation, column chromatography, and recrystallization. The active component was identified as ziyuglycoside I by a spectroscopic analysis. Ziyuglycoside I increased the expression of type I collagen in a dose-dependent manner (by up to 71.3% at 50 μ<SMALL>M</SMALL>). A clinical study of a formulation containing ziyuglycoside I, which involved visual evaluation and image analysis, showed a significantly different effect (<I>p</I><0.05) of the test formulation from that of the placebo. This result suggests that ziyuglycoside I isolated from <I>S. officinalis</I> root extract could be used as an active ingredient for cosmetics.</P>

        Phytotherapeutic Activities of <i>Sanguisorba officinalis</i> and its Chemical Constituents: A Review

        <P><I>Sanguisorba officinalis</I> Linne (<I>S. officinalis</I>, Rosaceae) has been used as a medicinal plant for the treatment of burns, hematemesis, melena, intestinal infections, and dermatitis for a long time in China, Korea, and Japan. The therapeutic efficacy of this herb is intimately associated with its anti-oxidant, anti-inflammatory, antiviral, antifungal, hemostatic, and anticancer activities. Its root contains triterpenoid saponins (zigyuglycoside I: C<TEX>$ _{41}$</TEX>H<TEX>$ _{66}$</TEX>O<TEX>$ _{13}$</TEX> and ziyuglycoside II: C<TEX>$ _{35}$</TEX>H<TEX>$ _{56}$</TEX>O<SUB>8</SUB>) and tannins (sanguiin H-6: C<TEX>$ _{82}$</TEX>H<TEX>$ _{54}$</TEX>O<TEX>$ _{52}$</TEX>). It has been recently revealed that these active constituents of <I>S. officinalis</I> possess antiwrinkle properties without cytotoxicity. They also have anticancer effects by inducing apoptosis and cell cycle arrest. Moreover, they can inhibit proliferative tumorigenesis. The underlying mechanism involved in the pharmacological actions of these active constituents is mainly related to p38 MAPK signaling. Although various studies have reported its therapeutic activities and major chemical components, review articles that extensively organize various properties of <I>S. officinalis</I> and its major constituents are still scarce. Taken together, the objective of this paper is to provide overall pharmacological and phytochemical profiles of <I>S. officinalis</I> and its constituents (including ziyuglycoside I, ziyuglycoside II, and sanguiin H-6), and their potential roles in clinical applications for the treatment of inflammatory diseases, bleeding disorders, and cancer.</P>

        Chemical constituents from Sanguisorba officinalis L. and their inhibitory effects on LPS-stimulated pro-inflammatory cytokine production in bone marrow-derived dendritic cells

        Sanguisorba officinalis L. (Rosaceae) is a perennial herbaceous plant and its roots have been used as an important astringent medicine in Eastern Asian countries over many thousand years. In this phytochemical research, 23 compounds (1-23) were isolated from the roots of S. officinalis. Their chemical structures were identified by extensive spectroscopic methods, including 1D and 2D NMR experiments. The anti-inflammatory effects of extracts and isolated compounds were investigated by measuring the production of pro-inflammatory cytokine IL-12 p40, IL-6 and <TEX>$TNF-{\alpha}$</TEX> in LPS-stimulated bone marrow-derived dendritic cell. Compounds 1, 3, 7-8, 11-14 revealed promising anti-inflammatory effects. These results suggested that some phenolic compounds and monoterpenoids from S. officinalis could be potential candidates for anti-inflammatory treatments.

        Skin Whitening Effects of Sanguisorba officinalis and Stichopus japonicus

        This study is aimed to evaluate skin whitening effects of Sanguisorba officinalis and Stichopus japonicus for cosmeceutical ingredients. The extract of Stichopus japonicus showed 61.78% inhibition of tyrosinase activity, and the mixture of Sanguisorba officinalis extract and Stichopus japonicus extract showed 59.14% inhibition of tyrosinase activity. On the clone M-3 cell meloanocyte, the mixture of Sanguisorba officinalis extract and Stichopus japonicus extract showed remarkable inhibition of melanogenesis. Also, those extracts were not irritable in ocular irritation test. It is concluded from these results that Sanguisorba officinalis and Stichopus japonicus have skin whitening effect. It could be used as natural depigmental material in cosmeceutical ingredients.

        지유 에탄올추출물의 생체외 항산화 활성

        The objective of this study was to investigate the antioxidative capacity of ethanol extracts from Sanguisorbae officinalis L. root (Sanguisorbae radix) in vitro. The concentration of Sanguisorbae radix extract at which DPPH radical scavenging activity was inhibited by 50% was 0.33 mg/mL, which was similar to IC50 of α-tocopherol (0.40 mg/mL), as compared to 100% by pyrogallol as a reference. Total antioxidant status was examined by total antioxidant capacity against ABTS radical reactions. Total antioxidant capacities of Sanguisorbae radix extract were significantly (p<0.05) higher than those of α-tocopherol. Superoxide scavenging activities of Sanguisorbae radix extract were significantly (p<0.05) higher than those of catechin. Oxygen radical absorbance capacities of Sanguisorbae radix extract were significantly (p<0.05)higher than those of ascorbic acid. Cupric reducing antioxidant capacities of Sanguisorbae radix extract were significantly (p<0.05) higher than those of α-tocopherol. Sanguisorbae radix extract prevented supercoiled DNA strand breakage induced by hydroxyl radical and peroxyl radical. Total phenolic contents of Sanguisorbae radix extract at concentrations of 0.5 and 5 mg/mL were 0.50 and 3.33 mM gallic acid equivalents, respectively. Sanguisorbae radix extract at concentration of 0.01, 0.1 and 0.5 mg/mL inhibited 0.2 mM tert-butyl hydroperoxide-induced cytotoxicity by 33.8, 79.1 and 96.9%,respectively, in HepG2 cell culture system. Thus, strong antioxidant and cytotoxicity-ihnibiting effects of Sanguisorbae radix extract seem to be due to, at least in part, the prevention from free radicals-induced oxidation as well as high levels in total phenolic contents. 본 연구에서는 지유 에탄올추출물의 항산화 효과를 조사하였다. Pyrogallol의 억제율을 100%로 기준하였을 때,DPPH 라디칼을 50% 억제시키는데 필요한 지유 추출물의농도는 0.33 mg/mL으로 α-tocopherol의 IC50(0.40mg/mL)과 유사하게 나타났다. 지유 추출물의 총항산화능은 α-tocopherol에 비해 높게 나타났다. 지유 추출물의superoxide 소거활성은 catechin에 비해 높게 나타났다. 지유 추출물의 peroxyl 라디칼 소거활성은 ascorbic acid에 비해 높게 나타났다. 지유 추출물의 구리이온 환원력은α-tocopherol에 비해 높게 나타났다. 지유 추출물은hydroxyl 라디칼 및 peroxyl 라디칼로 유발된 supercoiledDNA strand의 절단을 억제시켰다. 지유 추출물 0.5 및 5mg/mL의 총페놀 함량은 각각 0.50 및 3.33 mM gallicacid와 동등한 수준이었다. 또한, HepG2 세포주를 이용한세포배양에서 지유 추출물 0.01, 0.1 및 0.5 mg/mL 농도의 첨가는 0.2 mM t-BHP로 유도된 세포독성을 각각33.8, 79.1 및 96.9% 감소시켰다. 따라서, 본 연구 결과들은 지유 추출물의 강력한 항산화 효과와 세포독성 억제효과를 나타내며, 이러한 효능은 적어도 자유라디칼의 산화억제와 높은 총페놀 함량에 기인하는 것으로 사료된다.

        To develop a new anti-skin aging cosmetics or functional foods by using antioxidative activity and collagenase inhibitor, a potent collagenase or elastase inhibitor was screened from various extracts of medicinal plants and its optimal extraction condition was investigated. And antioxidative activity, antimicrobial activity and inhibitory of effect against collagenase activity were investigated. In the these results, we selected the Sanguisorba (Sanguisorba officinalis L.) that presents a potential biological activities. Sanguisorba which is very rich in triterpenoid saponin and tannins was recently reported its anti-oxidant activities and phytoestogenic activities in vivo test and many clinical studies. The experiments were carried out in vitro to determine anti-oxidant activities of Sanguisorba extracts on DPPH radical scavenging activity assay, Superoxidase scavenging activity assay, Elastase and collagenase activity assay. It show that the Sanguisorba extracts have the most significant anti-oxidant on free radical scavenging activity assay, and also inhibited significantly activities of elastase, collagenase. Further, Sanguisorba extracts are activated Type I collagen protein expression in CCD-986sk cells. These result suggest that the Sanguisorba extracts on DPPH radical scavenging activity assay, Superoxidase scavenging activity assay, elastase and collagenase activity assay, Type I collagen protein expression in CCD-986sk cells effected could be developed cosmetic ingredients for anti-aging.

        Antibacterial Activity of Sanguisorba officinalis against Helicobacter pylori

        In this study, a medicinal herbal plant, Sanguisorba officinalis, was examined and screened for anti-Helicobacter pylori (H. pylori) activity. Seventy percent ethanol was used for herbal extraction. For anti-H. pylori activity screening, inhibitory zone tests as an in vitro assay and in vivo study using a Mongolian gerbil (Meriones unguiculatus) model were performed. Also, the safety of herbal compounds was evaluated by animal study. As a result of inhibitory zone test, Sanguisorba officinalis extract demonstrated strong anti-H. pylori activities. Also, as results of in vivo animal studies, Sanguisorba officinalis extract demonstrated strong therapeutic effects against H. pylori infection according to the criteria of histological examination and rapid urease test. As results of the safety study, after 28 days treatment of the Sanguisorba officinalis extract, the animals were not detected any grossly and histological changes. These results demonstrate that it can be successfully cured against H. pylori infection and protected from H. pylori-induced pathology with Sanguisorba officinalis extract. It could be a promising candidate herb treatment for patients with gastric complaints including gastric ulcer caused by H. pylori.

        Antioxidant Activities of Sanguisorba officinalis and Stichopus japonicus

        This study is designed to apply for cosmeceutical ingredients with Sanguisorba officinalis and Stichopus japonicus distributed all over the country. The antioxidizing effect of the ethanol extract of Stichopus japonicus displayed stronger inhibition than the Sanguisorba officinalis ethanol extract and mixture of Sanguisorba officinalis and Stichopus japonicus extract. They used in the experiment as 63.28% with the density of 50 ㎍/mL, and compared with the ascorbic acid used as positive counterpart. Since the physiological activating ingredient generating the antioxidizing effect was confirmed with the Sanguisorba officinalis and Stichopus japonicus with the experiment results, the possibility for natural antioxidants was implied.

        당뇨를 유도한 동물모델에서 지유 추출물의 항산화 활성

        Background: Sanguisorba officinalis has been used in traditional Asian medicine owing to its beneficial effects on various diseases. The purpose of this study was to evaluate the effect of S. officinalis on the antioxidant system of Streptozotocin (STZ) and Alloxan (ALL) induced diabetic rats. Methods and Results: Triglyceride and Low-Density Lipoprotein (LDL)-cholesterol levels decreased in the STZ-induced diabetic groups treated with S. officinalis extract (SOE) compared to the corresponding levels in the control groups. Moreover, in the ALLinduced diabetic groups, SOE reduced triglyceride, LDL-cholesterol, and High-Density Lipoprotein (HDL)-cholesterol levels. Malondialdehyde (MDA) levels decreased significantly in the STZ and ALL-induced groups treated with SOE compared to the corresponding levels in the control group. Further, Glutathione (GSH) levels increased but did not reach statistical significance. The levels of Superoxide Dismutase (SOD) and Glutathione-S-Transferase (GST) showed a tendency to recover with SOE treatment in the STZ and ALL-induced diabetic groups. In addition, Catalase (CAT) levels in the SOE treatment group decreased significantly compared to those in the control group. Conclusions: These results suggest that SOE might be an effective agent in attenuating oxidative stress in diabetic patients by improving blood lipid profiles and inducing the anti-oxidative enzyme systems.

        지유 분획의 Candida albicans에 대한 항균효과

        We have studied the antifungal effect of 19 medicinal plants with paper disc diffusion method against candida albicans. As a result, Sanguisorba officinalis L., Cinnamomum cassia, Rheum coreanum, Perilla frutescens and Eugenia caryophyllata have been found to be effective against C. albicans. Among these, Sanguisorba officinalis L. was most effective at 24 hours and 48 hours. Its clear zone diameter was 17 mm for 24 hours and 16 mm for 48 hours. The antifungal activity of the solvent fraction of Sanguisorba officinalis L. by n-hexane, chloroform, ethyl acetate and n-butanol was the best for the chloroform fraction of 28 mm for 24 hours and 18 mm for 48 hours. The MIC concentration of the chloroform fraction was 80 mg/50 ml at 24 hours and 240 mg/50 ml at 48 hours.
