가공식품 중 태국칡(Pueraria mirifica) 혼입 판별법 개발

In this study, ribulose bisphosphate carboxylase (rbcL), RNApolymeraseC (rpoC1), intergenic spacer (psbA-trnH), and second internal transcribed spacer (ITS2) as identification markers for discrimination of P. mirifica in foods were selected. To be primer design, we obtained 719 bp, 520 bp, 348 bp, and 507 bp amplicon using universal primers from selected regions of P. mirifica. The regions of rbcL, rpoC1, and psbA-trnH were not proper for design primers because of high homology about P. mirifica, P. lobata, and B. superba. But, we had designed 4 pairs of oligonucleotide primers from ITS2 gene. Predicted amplicon from P. mirifica were obtained 137 bp and 216 bp using finally designed primers SFI12-miri-6F/SFI12-miri-7R and SFI12-miri-6F/SFI12-miri-8R, respectively. The species-specific primers distinguished P. mirifica from related species were able to apply food materials and processed foods. The developed PCR method would be applicable to food safety management for illegally distributed products in markets and internet shopping malls.
Pueraria mirifica 추출물 함유 화장품의 피부 탄력 임상 효능 평가

최근에 천연성분의 화장품 응용에 대한 관심이 높아지고 식물에 함유하고 있는 피토에스트로겐(Phytoestrogen)의 항노화 물질 연구가 중요해지고 있다. 태국 식물 Pueraria mirifica 추출물을 4% 함유한 크림제제(PM 크림)를 추출물 성분이 제외된 플라시보 크림과 비교하여 총 30명 여성을 대상으로 16주간 임상시험을 수행하였다. 피부 모사판을 제작하여 주름 측정을 하였고, 피부 탄력과 보습을 기기 평가하였다. 특히 피부 탄력도는 placebo 크림 사용군 대비 추출물 함유 크림 사용군이 통계적으로 유의하게 탄력도가 증가하였다. 주름이나 보습평가는 특이할 만한 결과를 나타내지는 못했다. Pueraria mirifiaca는 피부 탄력 개선을 주는 항노화 화장품 개발에 응용될 수 있을 것이라 기대된다. Recently, along with the remarkable increase in interest in natural products, the application of phytoestrogens in anti-aging products has become very important. In the focus, we developed Pueraria mirifica (PM), in this in vivo study on skin elasticity and wrinkles, and investigated the anti-aging properties. Thirty healthy women volunteers, between the ages of 30 and 60, applied the cream formula with 4% Pueraria mirifica or placebo, on the face area twice a day for 16 weeks. The evaluation was made by silicon replica analysis, Cutometer SEM 575 and Comeometer followed by statistical treatment using the student's t-test. Results of skin elasticity measurement showed that topical application on the skin increased the total surface of skin elasticity in comparison with placebo group (p<0.05). However, PM cream does not have prominent effect on the depth of wrinkles and skin moisturization. This in vivo study demonstrates that, PM exhibits a significant effect on skin elasticity therefore, is of great interest in anti-aging and firmness skin care products.
태국칡(Pueraria mirifica)으로부터 norsesquiterpene의 분리 및 동정

The roots of Pueraria mirifica were extracted with 70% aqueous ethyl alcohol and partitioned into ethyl acetate (EtOAc), n-butyl alcohol (BuOH), and H2O fractions, successively. From the EtOAc fraction, four norsesquiterpenes were isolated through the repeated silica gel, octadecyl silica gel and Sephadex LH-20 column chromatographies. On the basis of physicochemical and spectroscopic data including nuclear magnetic resonance (NMR), mass spectrometry, and infrared spectroscopy, the chemical structures were identified as megastigm-5-en-3,9-diol (1), linarionoside B (2), 3,5,6,9-tetrahydroxymegastigm-7-ene (3) and 3,4,9-trihydroxymegastigma-5,7-diene (4). Especially, the configuration of the anomer hydroxyl group was determined as a from the coupling constants of the anomer proton (J =8.0 Hz) in the 1H-NMR spectrum. These compounds were isolated for the first time from the roots of P. mirifica in this study.
