우리나라 대형선망어업에서 어획된 태평양참다랑어의 CPUE 표준화(2003-2016)

Pacific bluefin tuna (Thunnus orientalis) has been mostly caught by the Korean offshore large purse seine fishery in Korean waters. The annual catch of Pacific bluefin tuna caught by the offshore large purse seine fishery in Korean waters showed less than 1,000 mt until the 1990s except for 1997. The catch sharply increased to 2,401 mt in 2000 and recorded the highest of 2,601 mt in 2003, but the catch has generally decreased with a fluctuation thereafter. The main fishing ground of Pacific bluefin tuna of this fishery is formed around Jeju Island. However, it expanded to the Yellow Sea, the coastal of Busan, and the East Sea, which depends on the migration patterns of Pacific bluefin tuna by season. The CPUE standardization of Pacific bluefin tuna was conducted using Generalized Linear Model (GLM) to assess the proxy of the abundance index. The data used for the GLM were catch (weight), effort (number of hauls), catch ratio of Pacific bluefin tuna, moon phase by year, quarter and area. The standardized CPUE from 2004 to 2011, except for 2003 and 2010, showed a steady trend, and then increased until 2014. The CPUE in 2015 decreased, and in 2016 was higher than that in 2015. The result of GLM suggests that the effect of the catch ratio of Pacific bluefin tuna is the largest factor affecting the nominal CPUE.

The feeding habits of the Pacific bluefin tuna, Thunnus orientalis were examined based on the stomach contents of 330 specimens caught by offshore large purse seine fishery in the southern sea of Korea, 2011. The size of Pacific bluefin tuna used in this study ranged from 34.1 to 67.3 cm in fork length (FL). The percentage of empty stomachs was 41.8%. The main prey items were Pisces and Cephalopoda based on percentage IRI (index of relative importance). The main fish preys were Trachurus japonicus, Bregmaceros japonicus, Engraulis japonicus, and Cephalopoda were Todarodes pacificus, Loligo edulis, Watasenia scintillans. T. orientalis showed ontogenetic change in diet composition. Although Pisces dominated the diet of all length classes the portion of Cephalopoda was relatively higher in size between 40 and 50 cm other than length classes. In terms of seasonal variation in feeding habits, Pisces was the main prey group in all seasons, but Cephalopoda was also frequently consumed during spring and autumn based on %IRI.

T. orientalis showed ontogenetic changes in prey item that Pisces was dominated in size class of 30-89 cm (FL), Cephalopoda in 90-179 cm (FL), and Pisces above 180 cm. As for seasonal changes in prey item, Cephalopoda was dominant in spring and Pisces was dominant in summer, autumn and winter. %F, %N, %W and %IRI cluster analysis divided area into three groups: Group A was dominated by Pisces; Group B was dominated by Cephalopoda, and Group C was dominated by Euphausiacea.

참다랑어(Thunnus orientalis)의 배합사료 개발을 위해 전갱이(Trachrus japonicus) 근육 합성 엑기스(SE)를 사용하여 omission test를 통해 섭이촉진물질을 검색하였다. 인공종묘 생산된 참다랑어 치어(평균체중 $9.0{\pm}0.91\;g$: 실험 1, 2, 3; $1.6{\pm}0.23\;g$: 실험 4)를 사용하여 전갱이 근육 100 g 상당 함량의 각각의 섭이촉진 물질을 카제인 기초 사료에 첨가하여 총 4회의 사육실험을 실시하였다. 실험 1에서 인공합성엑기스(SE)와 SE로부터 아미노산 관련 화합물(SE-A), 핵산관련물질(SE-N) 그리고 유기염기화합물(SE-O)을 각각 제외한 시험액을 제조하여 천연 전갱이 근육 엑기스(NE)와 비교한 결과, SE-A와 SE-N구는 SE보다 낮은 섭이활성을 보였으나, SE-O구는 SE와 유사한 섭이활성을 보였다. 실험 2에서는 IMP가 섭이 촉진에 가장 많은 영향을 미치는 것이 인정되었으며, 실험 3에서는 L-alanine, L-glutamic acid, L-histidine, L-lysine, taurine 그리고 IMP의 혼합물이 SE와 섭이활성의 차이를 보이지 않았다. 실험 4에서는 L-histidine, L-glutamine 그리고 IMP 혼합물이 가장 높은 섭이활성을 보였으며, 그 다음으로는 SE, L-lysine, L-alanine and IMP 혼합물, L-histidine, L-glutamic acid, L-lysine and L-alanine과 IMP의 혼합물이 높은 섭이활성을 보였다. 본 연구결과 참다랑어 치어의 유효 섭이촉진물질은 L-histidine, L-glutamic acid 그리고 IMP로 판단되었다. For establishing a basal diet for the Pacific bluefin tuna Thunnus orientalis (PBT), feeding stimulants were initially identified by omission test using the synthetic extract of horse mackerel, Trachurus japonicus. Four feeding trials were conducted using juvenile PBT weighing $9.0{\pm}0.91\;g$ (trial 1, 2 and 3) and $1.6{\pm}0.23\;g$ (trial 4), which were originated from an artificial seedling production. The fish fed the casein diet with each test solution were added at the ratio of 100 g casein diet to 100 g jack mackerel muscle. A complete synthetic extract of jack mackerel containing all 3 fractions, amino acid, nucleotide and organic nitrogenous base, exhibited a comparable feeding stimulant activity compared to that of natural extract. The omission of nucleotide or amino acid fraction showed lower feeding activity, but the omission of other nitrogenous fraction maintained a similar feeding stimulant activity compared to that of the synthetic extract (trial 1). Inosine-5' monophosphate $Na_2$ (IMP) was identified as a major constituent for maintaining feeding activity. The mixture of L-alanine, L-glutamic acid, L-histidine, L-lysine, taurine and IMP induced a similar feeding activity compared to that of the synthetic extract (trial 2 and 3). In trial 4, the highest feeding activity was finally obtained in the mixture of L-histidine, L-glutamine and IMP, followed by the synthetic extract, the mixture of L-lysine, L-alanine and IMP, IMP and the mixture of L-histidine, L-glutamic acid, L-lysine and L-alanine. These results revealed that the mixture of L-histidine, L-glutamic acid and IMP for the proper feeding stimulant of PBT in this study.

The PBT skin collagen was composed of two ${\alpha}$ chains (${\alpha}1$ and ${\alpha}2$) and one ${\beta}$ chain. The denaturation temperature of PBT collagen was low although it was rich in proline and hydroxyproline. The primary structure of PBT skin collagen was almost identical to that of calf and salmon skin collagen; however, it differed with respect to the epitope recognition of the antibody against salmon type I collagen. These results suggest that the primary structure of skin collagen was highly conserved among animal species, although partial sequences that included the epitope structure differed among collagens.

This study was conducted to estimate dietary utilization of enzyme treated fish meal (EFM) on juvenile pacific bluefin tuna (PBT) Thunuus orientalis. In first experiment (EXP 1), the pacific bluefin tuna (initial body weight 2.36g) was randomly disposed two experimental tanks. The fish were fed EFM containing 31.9% of enzyme treated fish meal and sand lance (SL) for 15 days, respectively. Weight gain (WG) was showed lower in EFM than SL. But survival and protein efficiency ratio (PER) were higher in EFM than SL. Feed conversion ratio (FCR) of EFT lower than SL. In second experiment (EXP 2) was conducted to retest growth performance between EFM and SL. The high PER and survival were obtained in EFM than SL, and FCR of EFM was lower than that of SL. EFM are suitable dietary protein source in diet of juvenile PBT considering FCR, PER and survival in feeding trial.
Genome Wide Computational Identification of Tuna (Thunnus orientalis) MicroRNAs and Their Targets

Applying genome-wide computational-based approaches (using the draft genome sequence published in recent years) and following a set of strict filtering criteria, a total of 48 potentially conserved microRNAs belonging to 30 families were identified from economically important fish tuna (Thunnus orientalis). Using BLAST and RNA hybrid program a total of 19 potential miRNA targets were also identified in this study, which includes a number of signaling molecules (serine/threonine-protein kinase, GTPase activating protein etc.) and transcription factors (F-box protein, zinc finger protein etc.). This study provides the basis for gaining a better understanding of miRNA-mediated gene regulatory processes in fishes.
