        Anti-adipogenic effect of Lactobacillus fermentum MG431 and MG4244 through AMPK pathway in 3T3-L1 preadipocytes

        This study evaluated the anti-adipogenic effectsand mechanisms underlying the action of Lactobacillusfermentum MG4231 and MG4244 strains on adipogenesisand lipid accumulation in 3T3-L1 preadipocytes. Treatmentwith cell-free extracts (CFEs) from the two strainsreduced lipid accumulation and intracellular triglycerideproduction in 3T3-L1 adipocytes by more than 50%. Theinhibitory effects of L. fermentum on lipid accumulationwere mediated by the downregulation of FAS and aP2resulting from the inhibition of PPARc and C/EBPa geneexpression. Moreover, AMPK and HSL phosphorylationwas upregulated by CFE treatment. These results indicatedthat the anti-adipogenic and lipolysis activities of L. fermentumstrains were caused by increased AMPK and HSLphosphorylation. Both strains displayed high leucine arylamidaseand b-galactosidase enzymatic activity, withexcellent adhesion to epithelial cells. Therefore, we identifiedL. fermentum as potential new probiotics for theprevention of obesity.
