        <i>In Vitro</i> Immunoregulatory Effects of Korean Mistletoe Lectin on Functional Activation of Monocytic and Macrophage-Like Cells

        <P>Korean mistletoe lectin (KML) is one of the major active components in <I>Viscum album</I> var. (<I>coloratum</I>), displaying various biological effects such as anti-tumor and anti-metastatic activities. Even though it has been shown to boost host immune defense mechanisms, the immunomodulatory effects of KML on specific immune responses mediated by macrophages have not been fully elucidated. Therefore, in this study, we aimed to demonstrate KML's regulatory roles on macrophage-mediated immune responses. KML clearly blocked lipopolysaccharide (LPS)-induced events [expression of interleukin (IL)-10, nitric oxide (NO) production and phagocytic uptake], and suppressed the normal expression levels of IL-10 (at 2 ng/ml) and tumor necrosis factor (TNF)-α (at 10 ng/ml). In contrast, (1) the expression of cytokine (TNF-α) and (2) the generation of reactive oxygen species (ROS) induced by LPS were significantly up-regulated with KML co-treatment. In addition, KML itself increased the mRNA levels of IL-3 and IL-23; phagocytic uptake; the surface levels of co-stimulatory molecules (CD80 and CD86), pattern recognition receptors (PRRs) [such as dectin-1 and toll like receptor (TLR)-2] and adhesion molecules [β1-integrins (CD29) and CD43]; and CD29-mediated cell adhesion events. Finally, according to co-treatment of <SMALL>D</SMALL>-galactose with KML under LPS-induced NO production conditions, KML inhibition seems to be mediated by binding to proteins with <SMALL>D</SMALL>-galactose. Therefore, these data suggest that KML may participate in regulating various macrophage-mediated innate and adaptive responses <I>via</I> binding to surface protein with <SMALL>D</SMALL>-galactose and that some of these may deserve in KML's therapeutic activities such as anti-tumor and anti-microbial effects.</P>

        한국산 겨우살이의 항염증 효과

        본 연구에서는 한국산 겨우살이 (Viscum album L. var. coloratum)이 아토피 피부염과 관련된 염증성 사이토카인에 영향을 미치는지 여부를 알아보았다. 실험에는 헥산, 부탄올, 에틸아세테이트, 메틸렌클로라이드, 총 4 가지 분획물을 사용하였으며, RAW264.7 마우스 대식세포와 RBL-2H3 렛트 호중구를 이용하여 염증성 마커를 연구하였다. 실험 결과 에틸아세테이트 분획이 tumor necrosis factor alpha (TNF-α), interleukin(IL)-6, IL-4의 mRNA 발현 및 단백질 분비량을 감소시켰으나, 헥산 분획은 뚜렷한 효능이 없었다. 또한 부탄올 분획은 IL-4, IL-6의 mRNA 발현을 감소시켰고, 메틸렌클로라이드 분획은 IL-4와 TNF-α의 mRNA 발현을 감소시켰다. 결과적으로 한국산 겨우살이(V. album var. coloratum)가 아토피 피부염과 관련된 사이토카인 분비를 억제시켜 항염증 효과를 나타낼 수 있으므로, 이와 관련된 기능성 화장품 개발이 가능할 것으로 사료된다. In this study, we investigated whether Korean mistletoe (Viscum album L. var. coloratum) has anti-inflammatory effects that play key roles in the regulation of pathological mechanism of atopic dermatitis (AD). Four kinds of fractions, hexane (HX), ethyl acetate (EA), butanol (BU), and methylene chloride (MC), were used and RAW264.7 mouse macrophages and RBL-2H3 rat basophils were used to measure various inflammatory markers. EA significantly decreased mRNA expression and protein secretion levels of tumor necrosis factor alpha (TNF-α), interleukin (IL)-6, and IL-4 but HX did not affect these markers. In addition, BU decreased mRNA expressions of IL-4 and IL-6 whereas MC decreased IL-6 and TNF-α mRNA expressions. As a result, Korean mistletoe can show anti-inflammatory effects by inhibiting the secretion of cytokines related to AD, so it is thought that it will be possible to develop functional cosmetics related to this.

        누에 나방에 있어서 한국산 겨우살이(Viscum album coloratum) 추출물의 생명연장 효과

        Viscum album coloratum (Korean mistletoe; KM) is a semi-parasitic plant growing on various trees. This plant hasbeen shown to possess a variety of biological functions such as immunomodulation, apoptosis-induction and antitumor activity. However, there is no information about how the extract of KM affects life span of the host. In this study, in order to examine the effect of KM on the longevity of the host, we investigated whether a Korean mistletoe extract (KME) was able to extend the life length in an experimental model using the spring silkworm moth of Bombyx mori (Lepidoptera bombycidae). We established a model test for survival in which all of the male and female moths died within 18 days after the beginning of the experiment. Mean survival time of female moths was longer than that of male moths. Both male and female moths fed with KME containing diet (200 or 400 μg/head/day) showed significantly higher mean survival times than those of the control moths. Studying the effect of KME on prolongation of mean survival time showed that male moths were more susceptible than female moths. The moths fed with KME-containing diet showed a slight decrease in body weight. Interestingly, however, no difference in food intake was observed between moths fed with KME-containing diet and those with control diet. In analysis for mRNA increase in the SIRT2 gene, a member of the Sir2 gene family playing important roles in regulation of cell death and prolongation of life span, moths fed with KME-containing diet showed a significant increase in SIRT2 gene expression. These results suggest that KME has a potential to extend the life span in Bombyx mori, and its effect is partly associated with increase in SIRT2 gene expression.

        한국산 겨우살이 추출물 M11C(렉틴 구성물질)가단구세포의 TNF-α 유전자 발현유도 및 분비에 비치는 효과

        It is well-known that Korean mistletoe (Viscum album) extract has an immune activity and anticancer effect. In this study, Korean mistletoe extract, M11C (non-lectin components), was used to examine whether this extract might activate human peripheral monocyte to produce tumor necrosis factor-α (TNF-α). To examine the effect of M11C on the production of TNF-α from monocyte, the monocyte were stimulated by the M11C, and then collected the supernatant (M11C stimulated monocyte-conditioned media; MCM). MCM was treated to the TNF-α sensitive L929 cells, and then L929 cytotoxicity was measured by means of MTT. MCM had cytotoxic effect on L929. And the cytotoxic effect of MCM on L929 was almost abolished by anti-TNF-α antibody. These data indicated that MCM contained TNF-α, suggesting the TNF-α generation from M11C-stimulated monocyte. This suggestion was confirmed from the data that TNF-α was highly detected in MCM by immunoblotting technique. M11C effect on TNF-α production from monocyte was in the dose and stimulating time dependent manners. Also the effect of M11C on the expression of TNF-α mRNA from monocyte was shown in the dose and stimulating time dependent manners. As a result, Korean mistletoe extract, M11C, could be used for an immunostimulator.

        Korean Mistletoe (Viscum album var. coloratum) Inhibits Amyloid b Protein (25-35)-induced Cultured Neuronal Cell Damage and Memory Impairment

        The present study aims to investigate the effect of methanol extract of Korean mistletoe (KM; Viscum album var. coloratum), on amyloid b protein (Ab) (25-35), a synthetic 25-35 amyloid peptide, -induced neurotoxicity in cultured rat cerebral cortical neurons and memory impairment in mice. Exposure of cultured neurons to 10 mM Ab (25-35) for 24 h induced a neuronal cell death, which was measured by a 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) assay and Hoechst 33342 staining. KM (10, 30 and 50 mg/ml) significantly inhibited the Ab (25-35)-induced apoptotic neuronal death. KM (50 mg/ml) inhibited 10 mM Ab (25-35)-induced elevation of intracellular calcium concentration ([Ca2+]i), which was measured by a fluorescent dye, Fluo-4 AM. Glutamate release into medium and generation of reactive oxygen species (ROS) induced by 10 mM Ab (25-35) were also inhibited by KM (10, 30 and 50 mg/ml). These results suggest that KM may mitigate the Ab (25-35)-induced neurotoxicity by interfering with the increase of [Ca2+]i and then inhibiting glutamate release and generation of ROS in cultured neurons. In addition, orally administered KM (25 and 50 mg/kg, 7 days) significantly prevented memory impairment induced by intracerebroventricular injection of Ab (25-35) (8 nmol). Taken together, it is suggested that anti-dementia effect of KM is due to its neuroprotective effect against Ab (25-35)-induced neurotoxicity and that KM may have therapeutic role in prevention of the progression of Alzheimer's disease.

        한국산 겨우살이의 항산화, 항노화 및 미백 효과

        In this study, we investigated the anti-oxidant, anti-aging, and skin whitening effects of Korean mistletoe (Viscum album var. coloratum). The mistletoe fraction was composed of four types: hexane (HX), ethyl acetate (EA), butanol (BU), and methylene chloride (MC). In total phenol content assay, HX showed the highest phenol content among four fractions. In addition, EA significantly increased 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and catalase-like activities, and MC significantly increased superoxide dismutase (SOD)-like activity. When we compared IC50 value in the hyaluronidase and elastase inhibition assay, MC had the lowest IC50. In addition, we also performed tyrosinase inhibition assay to demonstrate the possibility of Korean mistletoe as a cosmetic component. HX showed the highest tyrosinase inhibition rate among the fractions. 본 연구에서는 한국산 겨우살이(Viscum album var. coloratum)의 항산화, 항노화, 미백 효능을 알아보았다. 겨우살이 분획물은 hexane (HX), ethyl acetate (EA), butanol (BU), methylene chloride (MC) 총 4 가지를사용하였다. 실험 결과 HX 분획물이 4 가지 분획물 중 가장 높은 페놀 함량을 보였다. 또한 EA 분획물이 가장높은 1,1-diphenyl-2-picrylhydrazyl (DPPH)자유 라디칼 소거능 및 catalase-유사 활성을 나타내었으며, MC 분획물이 가장 높은 superoxide dismutase (SOD)-유사 활성을 나타냈다. MC 분획물은 또한 가장 우수한hyaluronidase와 elastase 억제능을 보였다. 미백 효능을 알아보기 위하여 tyrosinase 억제능을 알아보았는데, HX 분획물이 가장 우수한 억제능을 보여주었다. 본 연구 결과를 통하여 한국산 겨우살이는 항산화, 항노화및 미백 활성을 가지는 기능성 화장품 소재로 사용이 가능할 것으로 보인다.
