      • Rhodococcus sp. 을 이용한 생물처리 시스템에 의한 hexane 제거 특성

        In this study, we investigated the characteristics of hexane removal in biological processes such as bofilter and biotrickling filter inoculated with Rhodococcus sp. EH831 or Rhodococcus sp. EC1. Hexane was removed in the biofilter inoculated with Rhodococcus sp. EH831 after lag period of 3 days, and then hexane elimination capacity and removal efficiency increased to 30 g•m-3•h-1 and more than 70%, respectively. However, the biofilter inoculated with Rhodococcus sp. EC1 showed lower hexane removal efficiency of 20~30% during initial 10 days, and it was up to 50% with large fluctuation. In the biotrickling filter inoculated with strain EH831, average hexane removal efficiency was 50%. These results indicate that biofilter and biotrickling filter system couldn’t offer the optimal system for hexane removal, although the systems have been widely used for the removal of other volatile organic compounds such as benzene, toluene, xylene, and styrene. Therefore, a novel biological system should be developed for the treatment of waste gas containing hexane. 본 연구에서는 Rhodococcus sp. EH831과 EC1을 사용하여 생물처리 시스템인biofilter와 biotrickling filter의 hexane 제거 특성을 조사하였다. Rhocococcus sp. EH831이 접종된 biofilter는 3일의 적응기후 hexane을 분해하기 시작하였고, hexane 제거능은 30 g•m-3•h-1, 제거효율은 70% 이상으로 향상되었다. 그러나, Rhodococcus sp. EC1이 접종된 biofilter는 초기 10일 까지는 hexane 제거율이 약 20~30%로 비교적 낮은 값을 보였고, 그 후 제거율이 50% 까지 향상되었으나 변동이 크게 나타났다. Rhodococcus sp. strain EH831이 접종된 biotrickling filter는 평균적으로 50%의 제거율을 보였다. 이러한 결과는 비록 biofilter와 biotrickling filter가 benzene, toluene, xylene, 그리고 styrene과 같은 VOCs 처리에 효과적으로 사용되었음에도 불구하고 hexane 제거에는 최적 조건을 제공하지 못한다는 것을 의미한다. 그러므로 hexane 폐가스 처리를 위해서는 새로운 개념의 생물학적 처리공정의 개발이 필요하다.

        Lipid Extraction from Tetraselmis sp. Microalgae for Biodiesel Production Using Hexane-based Solvent Mixtures

        Lipid extraction is a critical step in the downstream processing of biodiesel production from microalgae. Solvent extraction using mixtures of non-polar and polar solvents is one of the most well-known processes for this purpose. Hexane is the most common solvent of choice for large-scale lipid extractions due to its technical and economic advantages, especially its high selectivity toward lipids and low cost. In this study, extractions using mixtures of hexane and polar solvents were evaluated for their performance in order to develop a more efficient method for large-scale lipid extraction from microalgae. The combination of hexane and methanol resulted in the highest fatty acid methyl ester (FAME) yield for lipids from Tetraselmis sp. The effects of extraction conditions, including proportions of methanol to hexane, ratios of total solvent volume to dry biomass, and extraction time, on extraction yields were evaluated to determine optimum conditions providing higher lipid and FAME yields. The optimal conditions were as follows: proportion of hexane to methanol of 1:1, ratio of total solvent volume to dry biomass of 10 mL/g, and extraction time of 120 min. Finally, the selected solvent mixture and optimal conditions were applied to larger scale extraction experiments with scale-up factors of 10, 50, and 100. FAME yields of large-scale extractions were almost completely consistent with increasing scale-up factors. The results of this study suggest that a hexane and methanol mixture is a promising solvent for large-scale lipid extraction from microalgae.

        加味蔘笭白朮散 hexane세분획의 抗轉移 및 혈관형성억제작용에 대한 연구

        This experimental study was carried out to develop a new prescription to exert the antitumor and antimetastatic activity. The hexane fraction of Kamisamryungbaekchul-san(加味蔘笭白朮散: KSRBS) was most effective than other fraction from fractionation-guided screening. Thus, we studied the antimetastatic and antiangiogenic effects of hexane subfraction of KSRBS experimentally. Hexane fraction of KSRBS was devided into five groups(KS-H-S: S1-S5) to perform cytotoxity effect, cell adhesion assay, metalloproteinase expression and angiogenesis-related assays. On cytotoxicity, the subfractions of KSRBS inhibiting over 50% of cell growth were S3, S4 and S5 against HT1080 and S3, S5 were effective against the other cancer cell lines. Fraction inhibiting over 50% of cell adhesion was S1 against laminin, S4 and S5 against collagen Ⅳ. KRSBS-H-S inhibited MMP-2 mRNA expression in HT1080 cell up to 30% at 500㎍/㎖, up to 40% at 120㎍/㎖, while it didn't affect TIMP expression. S4 inhibited tube formation of ECV 304 to 90% at 250㎍/㎖, while it inhibited BCE proliferation of BCE cells treated with bFGF up to 40%.

        Microwave Plasma Process에 의한 N-Hexane으로부터 다이아몬드 박막제작 및 특성

        In this paper, the best conditions for the deposition of the high quality diamond thin-film from N-hexane as a carbon source in the microwave plasma process was carried out. Major parameters are the deposition time, flow rates of oxygen and hexane. The deposition time for the steady state thin-film was required more than 4[h], and the suitable flow rates of hexane and oxygen for the high-quality thin-film are 0.4[sccm] and 0.1~0.2[sccm], respectively. In addition, amorphous carbons such as DLC and graphite were grown by increasing the flow rate of hexane, and it decreased by increasing the flow rate of oxygen. Specifically, the growth rate is about 1.5[㎛h-1] under no addition of oxygen and it decreased about 60[%] as ca. 1.0[㎛h-1] with oxygen.

        노루발풀(Pyrola japonica) 추출물의 Bacillus subtilis에 대한 항균활성

        우리나라 전국 산지에서 쉽게 구할 수 있는 노루발풀을 건조시켜 80% ethanol 추출물과 여러 가지 용매 분획물의 미생물 증식 억제 효과에 대해서 검색하였다. 노루발풀의 80% ethanol 추출물은 그람 양성균 B. subtilis에서 농도 의존적으로 항균활성을 나타내는 반면 그람 음성균 E. coli에서는 활성이 나타나지 않았다. 노루발풀의 80% ethanol 추출물을 용매 분획하여 얻은 분획물 중 hexane층에서 항균활성이 나타나는 것을 확인하였다. 노루발풀의 항균활성 물질을 검색하기 위한 목적으로 hexane 분획물을 silica gel column chromatography, prep. TLC 및 HPLC를 실시하였으며 그 결과, peak I의 단일 물질이 노루발풀의 B. subtilis에 대한 항균활성을 나타내는 물질임을 확인할 수 있었다. The antimicrobial substance from Pyrola japonica were extracted and isolated. Eighty percent ethanol extract of dried Pyrola japonica was fractionated to hexane, diethyl ether, ethyl acetate and aqueous layer. The hexane-soluble fraction showed the highest inhibitory activity against Bacillus subtilis. Moreover the hexane layer was fractionated into 5 groups by silica gel column chromatography. From the results, group No. 2 ($18{\sim}40$ fractions) showed the highest antimicrobial activity. The group was re-separated to 10 fractions by preparative thin layer chromatography and the peak I as active fraction was isolated by HPLC.

        유통 올리브유의 잔류 헥산에 대한 연구

        The presence and content of residual hexane in the olive oils were studied. Total 41 olive oils of imported and domestic brands, which were labeled as extra virgin and refined (mixed), were collected from the market. For analysis, electronic nose and headspace SPME-GC/MS were used. Electronic nose equipped with 12 metal oxide sensors was used for the discrimination of odor pattern of olive oils against the different concentrations of hexane. From the results, it is assumed that the contents of residual hexane in the collected olive oils were below 5 ppm. For qualitative and quantitative analysis of hexane, polydimethylsiloxane (PDMS) fiber was employed for SPME-GC/MS. In the results, the peak of residual hexane was detected in 8 samples from 41 olive oils. But the detected level was no more than 1 ppm that is under the regulation limit (5 ppm) by Korea Food Additive Code.

      • Selective removal of polar VOCs by novel photocatalytic activity of metals co-doped TiO<sub>2</sub>/PU under visible light

        In this study, Ag and V co-doped TiO<SUB>2</SUB> deposited on polyurethane (Ag♀-TiO<SUB>2</SUB>/PU) was for novel photocatalytic removal of gaseous volatile organic compounds (VOCs). The combination of V doping, which enhanced internal electron transfer in the TiO<SUB>2</SUB> lattice, and Ag doping, which exhibited high internal electron transfer in the Ag<SUB>2</SUB>O and enhanced exterior electron transfer among Ag particles, Ag<SUB>2</SUB>O and TiO<SUB>2</SUB>, increased the photocatalytic activity of Ag♀-TiO<SUB>2</SUB>/PU for the removal of VOCs in aerosol. The surface area of this co-doped photocatalyst was also higher than that of the undoped and single-dopant photocatalysts. The optimal combination of weight ratios of Ag/TiO<SUB>2</SUB> and V/TiO<SUB>2</SUB> for maximizing the surface area and photocatalytic activity of TiO<SUB>2</SUB> was 4 and 2%, respectively. Under visible light, the removal efficiency of individual hexane and butyl acetate gas by 4Ag½V-TiO<SUB>2</SUB>/PU was 93.7 and 95.5%, respectively. The removed hexane and butyl acetate were mineralized into CO<SUB>2</SUB> with ratios of 93.2 and 96.2%, respectively. The individual removal of hexane and butyl acetate gas by Ag♀-TiO<SUB>2</SUB>/PU was similar; however, in the mixture stream of both VOCs, much more butyl acetate than hexane was photocatalytically removed. This was associated with the much higher polarity of butyl acetate than that of hexane, so that the photocatalyst surface, where most of the oxy radicals were generated to oxidize the VOCs, was more occupied or covered by butyl acetate than by hexane.

        바이오매스 유래 파클리탁셀의 경제적인 회수를 위한 물과 헥산의 순차적 세척 방법

        In this study, a tandem water and hexane washing process was developed to improve the recovery efficiency of paclitaxel derived from Taxus chinensis. The polar impurities contained in the sample were effectively removed by washing with water at a sample/water ratio of 1:40 (w/v) for 10 min. In addition, the non-polar impurities were effectively removed by washing with hexane at a sample/hexane ratio of 1:160 (w/v) for 20 min. A high purity of paclitaxel (>30.0%) was obtained in a short operating time (~30 min) by sequential washing with water and hexane.

      • LC-MS/MS를 활용한 갈색거저리 유충 중 농약 353성분 동시 분석법 확립 및 유효성 검증

        식용곤충 중 밀웜으로 잘 알려진 갈색거저리 유충(Tenebrio molitor larva)은 단백질 및 지방이 풍부하여 식용곤충 내 농약 다성분 분석법을 확립하기 위한 대표 시료로 적합하다. 본 연구에서는 갈색거저리 유충 중 농약 353성분의 동시 분석을 위해 acetonitrile-hexane 액액분배가 추가된 새로운 QuEChERS 전처리법 및 LC-MS/MS (liquid chromatography-tandem mass spectrometry)를 활용한 다성분 분석법을 검증하였다. 다성분 동시 분석을 위해 농약 353성분에 대한 LC-MS/MS (AB SCIEX Triple Quad<sup>TM</sup> 5500 및 Exion LC<sup>TM</sup>)의 MRM (multiple reaction monitoring) 조건을 확립하였고, 각 성분은 Halo C18 column (2.1 mm I.D. × 150 mm, 2.7 μm)을 사용하여 20분 이내로 분리하였다. 최종 확립된 전처리법에서는 갈색거저리 유충을 마쇄하여 균질화한 후 이중 5 g을 acetonitrile 12.5 mL로 추출하고 EN 15662 salt를 가한 후 원심분리하여 분배하였다. Acetonitrile-hexane 액액분배를 위해 상등액 6 mL에 hexane 4 mL을 가해 분배하였으며, 잔여 hexane은 새로운 acetonitrile (hexane 포화) 6 mL로 2회 액액분배하여 총 18 mL acetonitrile 추출액을 모았다. 추출액은 C18 sobent이 함유된 dSPE로 정제하고 원심분리한 다음 matrix-matching한 후, 이 중 2 μL를 LC-MS/MS에 주입하여 분석하였다. 분석법 검증 결과, 농약 353성분 모두 분석법상 정량한계(method limit of quantitation; MLOQ) ≤ 10 μg/kg으로 positive list system (PLS)에서 요구하는 기준(10 μg/kg)을 만족하였다. 검량선의 범위는 LOQ∼200 μg/kg이며, 모든 성분에 대해 r<sup>2</sup> ≥ 0.990으로 우수한 직선성을 보여주었다. 회수율 검증 결과 농약 320성분(전체 중 90.7%)이 10 및 50 μg/kg 두 수준 모두에서 회수율 70∼120% (RSD 20% 이하)였다. 검증된 분석법을 바탕으로 실제 갈색거저리 유충 시료 11점에 대해 농약 다성분 모니터링을 실시하여 분석법의 유효성을 입증하였다.

FAME yields of large-scale extractions were almost consistent with increasing scale-up factors. The results of this study suggest that a mixture of hexane and methanol is a promising solvent for a large-scale lipid extraction from microalgae.
