This study is focused on clarifying the functional aspects of the extracts from Gleditsiae spina, which has been used as one of the favorite chinese medicinal herbs for a long time. To detect the possible effects of Gleditsiae spina extracts on the immune system of mice, macrophage lysosomal enzyme activity and anti-complementary activity were calculated in semi-vivo. Among different solvent extraction, hot water extract of Gleditsiae spina showed higher macrophage lysosomal enzyme activity and anti-complementary activity than others.<BR> During mass extraction, low molecular weight fraction was removed by using MtOH extraction of Gleditsiae spina. High molecular fweight raction was obtained by hot water extraction and the EtOH precipitation. Fraction GS-1, showing 212% macrophage lysosomal enzyme activity and 81.5% anti-complementary activity, consisted of 38.8% carbohydrate, 21.1% protein and 10.1% uronic acid. Besides no lethal toxicity was observed in GS-1.

Objectives: This research was investigated the anticoagulant effect of the Gleditsiae spina extract. Methods: We researched prothrombin time (PT) assay, activated partial thromboplastin time (APTT) assay in vitro and in vivo using arteriovenous (A-V) shunt rat model and shortening Rat tail bleeding time (BT). A-V shunt and BT were treated with extract of Gleditsiae spina (GS) 400 ㎎/kg for a week. Results: Bleeding time of Gleditsiae spina extract in vivo had a significant increase as about 1.2 times and thrombus weight of Gleditsiae spina extract had a significant reduction of thrombus weight as 26%. Gleditsiae spina extract represented an effect of anticoagulation by operating on extrinsic pathway factor II, V, VII, X and intrinsic pathway factor Ⅷ, Ⅸ, Ⅹ, XI, XII in the coagulation system. Conclusions: Considering the above mentioned results, it is judged that a Gleditsiae Spina extract has a control effect of thrombus creation.

Objectives : Gleditsiae Spina has the effects of expelling toxins, draining pus, invigorating blood and resolving abscesses. Some clinicians apply the herb for patients suffering from cancer. However, its anti-cancer activities are not well understood. In the present study, anti-tumor agents from Gleditsiae Spina were purified. Methods : The viability of the PC-3 cell line was determined using MTT assay, and the induction of apoptosis by Gleditsiae Spina extract in PC-3 cells was measured by Annexin-V/propidium iodide double staining assay detected by flow cytometry. TLC and HPLC analysis were used to separate and identify the anti-cancer agents. Results : Treatment of the extract resulted in significant decreased cell viability of PC-3 cells in a dose- and time-dependent manner. Dose-dependent apoptotic cell death was also measured by flow cytometry analysis. The anti-cancer agents were successfully separated and identified by using TLC and HPLC analysis and the most potential agent among them was separated from EtOAC fraction. Conclusions : These results might be applied in developing new drugs from natural resources like Korean traditional medicine, and also support the clinical usefulness of herbal medicine.

The anti-inflammatory activity of the aqueous extract from Gleditsiae spina was investigated utilizing carrageenin-induced edema, granuloma pouch and adjuvant arthritis in rats. The effects of this agent on vascular permeability and acetic acid-induced writhing in mice were also examined. Its anti-inflammatory activity on carrageenin edema was observed with oral administration. The aqueous extract from Gleditsiae spina(400mg/kg, 7 days) showed significant inhibitory effects on granuloma and exudate formation in rats. In the method of adjuvant arthritis, the aqueous extract(400mg/kg), orally administered for 21 days, inhibited the development of hind paw edema in rats. The aqueous extract was also inhibited the increase in vascular permeability and the number of writhings induced by acetic acid in mice. In the present study, the aqueous extract of Gleditsiae spina was indicated to have the anti-inflammatory action.
조角刺추출물의 Nrf2 활성화를 통한 간세포 보호 효과

Objectives: Oxidative stress is one of the most causes of hepatocyte injury. Gleditsia spina, the thorns of Gleditsia sinensis Lam., has been known for its anti-cancer and anti-inflammatory effects in Korean medicine. The present study investigated hepatoprotective effect of Gleditsia spina water extract (GSE) against oxidative stress induced by arachidonic acid (AA) + iron in HepG2 cells. Methods: To investigate cytoprotective effect of GSE, cells were pretreated with GSE and then subsequently exposed to 10 μM AA for 12 h, followed by 5 μM iron. Cell viability was monitored by MTT assay, and expression of apoptosis-related proteins was examined by immunoblot analysis. To identify responsible molecular mechanisms, reactive oxygen species (ROS) production, GSH contents, and mitochondrial membrane potential were measured. In addition, effect of GSE on nuclear factor erythroid 2-related factor 2 (Nrf2) activation was determined by immunoblot and antioxidant response element (ARE)-driven reporter gene assays. Results: GSE pretreatment prevented AA + iron-mediated cytotoxicity in concentration dependent manner. In addition, ROS production, glutathione depletion, and mitochondrial impairment by AA + iron were significantly inhibited by GSE. Furthermore, GSE promoted translocation of Nrf2 to nucleus, which acts as essential transcription factor for induction of antioxidant genes. Increased nuclear Nrf2 that caused by GSE treatment promoted transcriptional activity of ARE. Finally, GSE up-regulated sestrin-2 which was widely recognized as target gene of Nrf2. Conclusions: This study demonstrates that GSE protects hepatocytes from oxidative stress via activation of Nrf2 signaling pathway.

Glenditsia spina (GS) can resolve carbuncle, relive swelling, dispel wind and destroy parasites. For these reasons, GS has been widely used as dermatologic agent clinically. In this study, the specific pathways of anti-proliferative effect of GS on human derived melanoma cells were identified. The molecular profile was measured using microarray technique to identify up- or down-regulated genes in SK-MEL-2 cell line. Pathway analysis was done by listing percentage of pathway involvement, and the represented pathways were obtained from KEGG. The transcription factor binding sequences were obtained by Transfac database. By the promoter analysis, up-regulated genes by GS were mainly associated with MAPK, Regulation of actin cytoskeleton, Wnt, Focal adhesion and Long term potentiation pathway. Down-regulated genes by GS were mainly associated with MAPK and Antigen processing and presentation pathway. And some of the transcription factors like Sp1 and NF-Y in up-regulated genes and Oct-1 in down-regulated genes by GS also identified.

Objectives: This study was designed to investigate the effects of Gleditsiae spina(GS) on changes in cerebral blood flow in rats. Methods: The present study was investigated regional cerebral blood flow(rCBF) and mean arterial blod pressure(MABP) in rats. In addtion, the present study also investigated action mechanisms of GS on changes in rCBF and MABP by Pre-treatment with indomethacin(Idm) and methylene blue(MTB), an inhibitor of a vasodepressor material. Results: Treatment with GS elevated rCBF in dose-dependent manner, but MABP levels were not affected by treatment with GS. Pre-treatment with indomethacin(IDM), an inhibitor of cyclooxygenase, inhibited increase of rCBF induced by GS too, In addition, Pre-treatment with MTB inhibited increase of MABP too. But, Pre-treatment with IDM did not affect MABP levels. Conclusions: These results suggest that GS is effective to treat patient with disease related to cerebral ischemia, because GS can increase rCBF. In addition, the mechanisms are thought to be related to both of cyclooxygenase and guanylate cyclase pathways.

Objective : This study was designed to investigated effects of Gleditsia spina (GS) on human derived melanoma cells Methods : The genetic profile for the effect of medicine on human derived melanoma cells of SK-MEL-2, was measured by using microarray technique, and the functional analysis on these genes was conducted. The network of total protein interactions was measured by using cytoscape program. Results : Total 253 genes were up-regulated and 439 genes down-regulated in cells treated with GS. Genes induced or suppressed by GS were all mainly concerned with metabolic process, regulation of biological process and protein binding. Conclusion : Suggest the possibility of GS as anti-cancer drug and cosmetic agent, and also suggest that related mechanisms are involved in regulation of intra-cellular metabolism in melanoma cells.

This case was designed to evaluate the improving effect of hepatitis on drug metabolism. Gleditsiae Spina(?甬刺) was included Taeumin' herb. but it maybe induced hepatitis on drug metabolism, if Taeumin take this herb long time. We use Taeyeumin Chungsimyeunjatang to the hepatitis on drug metabolism, and we improved it. so we report it.
