        Induction of Defense Response Against Rhizoctonia solani in Cucumber Plants by Endophytic Bacterium Bacillus thuringiensis GS1

        An endophytic bacterium, Bacillus thuringiensis GS1, was isolated from bracken (Pteridium aquilinum) and found to have maximal production of chitinase (4.3 units/ml) at 5 days after culture. This study investigated the ability of B. thuringiensis GS1 to induce resistance to Rhizoctonia solani KACC 40111 (RS) in cucumber plants. Chitinase activity was greatest in RS-treated plants at 4 days. β-1,3- Glucanase activity was highest in GS1-treated plants at 5 days. Guaiacol peroxidase (GPOD) activity increased continuously in all treated plants for 5 days. Ascorbate peroxidase (APX) activity in RS-treated plants was increased 1.5-fold compared with the control at 4 days. Polyphenol oxidase (PPO) activity in RS-treated plants was increased 1.5-fold compared with the control at 3 days. At 5 days after treatment, activity staining revealed three bands with chitinase activity (Ch1, Ch2, and Ch3) on SDSPAGE of cucumber plants treated with GS1+RS, whereas only one band was observed for RS-treated plants (Ch2). One GPOD isozyme (Gp1) was also observed in response to treatment with RS and GS1+RS at 4 days. One APX band (Ap2) was present on the native-PAGE gel of the control, and GS1- and GS1+RS-treated plants at 1 day. PPO bands (Po1 and Po2) from RS- and GS1+RS-treated plants were stronger than in the control and GS1-treated plants upon native-PAGE at 5 days. Taken together, these results indicate that the induction of PR proteins and defense-related enzymes by B. thuringiensis GS1 might have suppressed the damping-off caused by R. solani KACC 40111 in cucumber plants.

        Ginsenoside Re Enriched Fraction (GS-F3K1) from Ginseng Berries Ameliorates Ethanol-Induced Erectile Dysfunction via Nitric Oxide-cGMP Pathway

        Erectile dysfunction (ED) is a highly prevalent disorder that affects millions of men and considered to be an early symptom of atherosclerosis and a precursor of various systemic vascular disorders. The aim of the present study was to prepare ginsenoside Re enriched fraction (GS-F3K1, ginsenoside Re 10%, w/w) from ginseng berries flesh and to investigate the enhanced activities of GS-F3K1 on alcohol-induced ED. GS-F3K1 was prepared by the continuous liquid and solid separating centrifugation and circulatory ultrafiltration from ginseng berries flesh. GS-F3K1 was administered for 5 weeks in ethanol-induced ED rat by oral administration of 20% ethanol. To investigate the effects of GS-F3K1 on ED model, the levels of nitrite expression, cyclic guanosine monophosphate (cGMP) and erectile response of the penile corpus cavernosum of rat were measured. The erectile response of the corpus cavernosum was restored after GS-F3K1 administration, to a level similar to the normal group. The level of nitrite and cGMP expression in the corpus cavernosum of GS-F3K1-administered male rats was increased significantly compared to positive control group. GS-F3K1 from ginseng berries should effectively restore ethanol-induced ED in male rats and could be developed as a new functional food for the elderly men.

        Isolation and molecular characterization of two novel HMW-GS genes from Chinese wheat (Triticum aestivum L.) landrace Banjiemang

        High molecular weight glutenin subunits(HMW-GS) play a significant role in determining breadmakingqualities of common wheat flour. The study presentedin the paper identified two novel HMW-GS (designatedas 1Bx14* and 1By15*) at the Glu-B1 locus inChinese wheat (Triticum aestivum L.) landrace Banjiemangby SDS-PAGE. The open reading frames (ORFs) of thegenes were amplified and cloned with designed specificprimers. Both of them had sequences highly similar tothose of other HMW-GS genes, but displayed minormodification. The ORF of 1Bx14* was 2367 bp long andencoded 789 aa with 4 cysteines, which included 25nanopeptides, 65 hexapeptides and 7 tripeptides motifs. The ORF of 1By15* was 2106 bp long and encoded 702 aawith 7 cysteines, which included 23 nanopeptides and 53hexapeptides motifs. The predicted molecular weights ofthe mature proteins encoded by 1Bx14* and 1By15* geneswere 83.1 and 73.2 k Da, respectively. The presence andauthenticity of the two genes in Banjiemang were confirmedby bacterial expression and liquid chromatographyelectrosprayionization mass spectrometry (LC–ESI–MS). By phylogenetic analysis, the 1Bx14* was clustered withx-type subunits but had a high divergence with other 1Bxsubunits, whereas the 1By15* was clustered with y-typesubunits and closely related to subunit 1By15. The studyconcluded that 1Bx14* and 1By15* from Banjiemang werenovel allelic variations of HMW-GS at Glu-B1 locus,which were probably exploitable as new resources forwheat quality improvement.

        ‘참동진/영호진미’ 재조합집단을 이용한 수량 관련 형질 양적형질 유전자좌 분석

        ‘Chamdongjin’ is a near-isogenic line with enhanced disease resistance compared to ‘Sindongjin,’ a mega-cultivar in Korea. ‘Younghojinmi’ is the rice cultivar with the highest taste quality among the premium cultivars. This study conducted quantitative trait loci (QTL) analysis on yield-related traits using the recombinant inbred lines derived from a cross between ‘Chamdongjin’ and ‘Younghojinmi’ to elucidate genetic interaction. We utilized KASP markers and a target-capture sequencing SNP genotyping platform to address the insufficient genetic diversity of the population using Korean japonica cultivars. A QTL associated with heading date (HD), qHd8, was detected, and the candidate gene Hd18 was identified. The hd18 allele delayed HD by 3 days and reduced the brown/rough rice ratio (BRR) by 0.3%. QTLs associated with culm length (CL), panicle length (PL), and yield (g/plant), namely qCL1, qPL1_2023, and qYield1_2023, respectively, were detected, and the candidate gene SD1 was identified. The sd1 allele shortened CL and PL by 11 and 0.8 cm, respectively, and reduced the yield by 2.3 g. It also reduced the number of spikelets per panicle (NS) by 10 and decreased the ratio of ripened grains (RRG) by 2.6 %. QTLs associated with 1,000-grain weight (TGW), NS, and BRR, namely qTGW3, qNS3, and qBRR3_ 2023, respectively, were detected, and the candidate gene GS3 was identified. The gs3 allele increased TGW and BRR by 3.8 g and 0.4%, respectively, and reduced NS by 12. It also reduced RRG content by 1.9% and increased the yield by 1.7 g. The QTL associated with the number of panicles per hill (PN), qTN3, and the candidate gene, OsTB1, were identified. The OsTB1TGTG allele reduced PN by 1.3 and affected the increase in NS and RRG by 10 and 1.4%, respectively. The allele combinations of SD1, GS3, and OsTB1 were analyzed to assess their effects on yield-related traits. NS and RRG were influenced by trait variations, depending on the combination background of other alleles during allele substitution. ‘Jeonju 697,’ an elite breeding line exhibiting improved lodging resistance with shortened CL, was selected by substituting SD1 with sd1 while maintaining the Hd18-gs3-OsTB1TGTG allele combination of ‘Chamdongjin.’ This is expected to be a practical alternative for addressing the lodging problem in ‘Chamdongjin.’

        Allelic Variation and Composition of HMW-GS in Advanced Lines Derived from D-genome Synthetic Hexaploid / Bread Wheat (Triticum aestivum L.)

        The objective of this study was to identify allelic variations at Glu-1 loci of wheat (Triticum aestivum L.) advanced lines derived from hybridization of bread wheat and synthetic hexaploid wheats (2n = 6x = 42; AABBDD). Locally adapted wheat genotypes were crossed with synthetic hexaploid wheats. From the 134 different cross combinations made, 202 F 8 advanced lines were selected and their HMW-GS composition was studied using SDS-PAGE. In total, 24 allelic variants and 68 HMW-GS combinations were observed at Glu-A1, Glu-B1, and Glu-D1 loci. In bread wheat, the Glu-D1 locus is usually characterized by subunits 1Dx2+1Dy12and 1Dx5+1Dy10 with the latter having a stronger effect on bread-making quality. The subunit 1Dx5+1Dy10 was predominantly observed in these advanced lines. The inferior subunit 1Dx2+1Dy12, predominant in adapted wheat germplasm showed a compara-tive low frequency in the derived advanced breeding lines. Its successful replacement is due to the other better allelic variants at the Glu-D1 locus inherited in these synthetic hexaploid wheats from Aegilops tauschii (2n = 2x = 14; DD).

        GS1을 활용한 빅데이터 분석 플랫폼 기반의 스마트 소화기구 모니터링 시스템

        This paper presents a smart firefighting appliances monitoring system based on big data analytics platform using GS1 for Smart City. Typical firefighting appliances are fire hydrant, fire extinguisher, fire alarm, sprinkler, fire engine, etc. for the fire of classes A/B/C/D/E. Among them, the dry chemical fire extinguisher have been widely supplied and 6 millions ones were replaced for the aging ones over 10 years in the past year. However, only 5% of them have been collected for recycling of chemical materials included the heavy metals of environment pollution. Therefore, we considered the trace of firefighting appliances from production to disposal for the public open service. In the paper, we suggest 1) a smart firefighting appliances system using GS1, 2) a big data analytics platform and 3) a public open service and visualization with the analyzed information, for fire extinguishers from production to disposal. It can give the information and the visualized diagrams with the analyzed data through the public open service and the free Apps

        Pseudohypoparathyroidism Type 1A with Normocalcaemia, due to the Novel C.389A>G Variant of Exon 5 of the Guanine Nucleotide-Binding Protein, α-Stimulating Gene

        Pseudohypoparathyroidism type 1A (PHP1A) is a rare disease caused by molecular defects in the maternally-inherited allele of the guanine nucleotide-binding protein, α-stimulating (GNAS) gene. The GNAS gene encodes the stimulatory G-protein α-subunit that regulates production of the second messenger cyclic adenosine monophosphate. Heterozygous inactivating mutations in these specific loci are responsible for a spectrum of phenotypic characteristics of the disease, including clinical features of the Albright’s hereditary osteodystrophy, due to resistance to parathyroid hormone (PTH). We report a case of PHP1A and explore the underlying novel point mutation of the GNAS gene that leads to an atypical PHP1A phenotype. A male patient with a round face, short stature, and brachydactyly accompanied by normocalcaemia and mild PTH resistance consulted at our center. The GNAS encoding region from the patient and both of his parents were amplified and sequenced directly in a sample of peripheral blood leukocytes. A novel c.389A>G point mutation in exon 5 of the GNAS gene, resulting in a p.Tyr130Cys peptidic chain change of the Gsα protein, detected in the proband, in heterozygous state. Sequencing of the GNAS gene from his parents did not reveal the c.389A>G mutation, confirming a de novo proband genotype. The maternal origin of the affected GNAS allele, along with mild PTH resistance, confirmed the PHP1A diagnosis. PHP1A, caused by inactivating GNAS mutations, presents a range of complex clinical phenotypes. The novel c.389A>G GNAS mutation presented in this case expands the spectrum of known PHP1A molecular defects and describes the associated phenotype.

      • Traceability System for Fire Fighting Equipment using GS1 and EPCIS: Production, Inspection, and Recycling

        This paper presents a traceability system for fire-fighting equipment using GS1 and EPCIS standards for production, distribution, installation, inspection, fire-fighting, collection for disused ones, disposal, and recycling of material. There have been lot kinds of fire-fighting equipment: fire engine, fire extinguisher, hydrant, sprinkler, fire detector, automatic prompt reporter, rescue and descent device, fire wall, etc. Among them, because it makes with chemical substances for fire-fighting, it needs to collect the disused/aged ones for disposal/recycling. Now a day, there is an issue for Extended Producer Responsibility (EPR) that has responsibility from production to disposal in whole process for the products. Typically, the dry chemical fire extinguisher over 4 million have been disused in the past year. Therefore, we considered the traceability of the fire-fighting equipment included harmful chemical substances from production to disposal/recycling. In the paper, we suggest 1) a unique device identifier of GS1 for the equipment, 2) an EPCIS for the traceability based on the producer responsibility. It can get/service the information from production to recycling not only the local products but also the imports/exports.

        고해상 광학센서의 스펙트럼 응답에 따른 영상융합 기법 비교분석

        본 연구는 서로 다른 센서 특성을 지닌 KOMPSAT-2, QuickBird 및 WorldView-2 고해상도 위성영상에 영상융합기법을 적용하여 그 결과를 비교평가 하는 것이다. 사용된 기법은 대표적인 CS 기반 융합기법인 GIHS, GIHSA, GS1 및 Adaptive IHS를 사용하였다. 영상융합 기법의 품질평가는 시각적 분석과 정량적 분석을 수행하였으며, 정량적 분석에는 SAM, Spectral ERGAS 및 Q4을 사용하였다. KOMPSAT-2 영상은 GHISA 기법의 경우 상대적으로 우수한 성능을 나타내는 반면, QuickBird와 WorldView-2영상은 GS1기법의 경우에 우수한 성능을 나타낸다. This study aims to evaluate performance of various image fusion methods based on the spectral responses of high-resolution optical satellite sensors such as KOMPSAT-2, QuickBird and WorldView-2. The image fusion methods used in this study are GIHS, GIHSA, GS1 and AIHS. A quality evaluation of each image fusion method was performed with both quantitative and visual analysis. The quantitative analysis was carried out using spectral angle mapper index (SAM), relative global dimensional error (spectral ERGAS) and image quality index (Q4). The results indicates that the GIHSA method is slightly better than other methods for KOMPSAT-2 images. On the other hand, the GS1 method is suitable for Quickbird and WorldView-2 images.
