        Physiological Effects of Hot Water Extract and Solvent Fractions of Carthamus tinctorius L.

        In this study, we investigated the biological activities of Carthamus tinctorius including antioxidative, fibrinolytic, thrombin inhibitory, α-glucosidase inhibitory and collagenase inhibitory activities. Carthamus tinctorius, hot water extract was fractionated into hexane, chloroform, ethyl acetate, butanol and water fraction. Each of these was assayed individually. The hot water extract showed high antioxidative activity and thrombin inhibitory activity at 90.17% and 97.10% respectively. In the fraction activity tests, chloroform fraction showed the highest antioxidative activity at 81.85%. The fibrinolytic activity was strong only in the butanol fraction at 0.70 plasmin units/ml. The thrombin inhibitory activities of hexane, ethyl acetate and butanol fractions were 97.35%, 86.74% and 93.18% respectively. In collagenase inhibitory activity test, hexane fraction showed the highest activity at 87.78%. In conclusion, the hot water extract and solvent fractions of Carthamus tinctorius L can be used as a material for the development of biofunctional tea and foods respectively.

        홍어류(Dipturus chilensis ) 껍질로부터 분리 정제된 collagenase-1 저해 펩타이드의 특성

        We attempted to isolate a collagenase-1 inhibitory peptide from skate Dipturus chilensis skin protein. The protein from skate skin was digested by various enzymes (alcalase, α-chymotrypsin, neutrase, papain, pepsin, and trypsin) to produce a collagenase-1 inhibitory peptide. The collagenase-1 inhibitory activity of the peptides obtained was measured by gelatin digestion assay. Among the six hydrolysates, pepsin hydrolysate exhibited the highest collagenase-1 inhibitory activity. The peptide showing strong collagenase-1 inhibitory activity was purified by Sephadex G-25 gel chromatography and HPLC using an octadecylsilyls (ODS) column. The amino acid sequence of purified collagenase-1 inhibitory peptide was identified to be Asn-Leu-Asp-Val -Leu-Glu-Val-Phe (961 Da) by quadrupole time of flight (Q-TOF) and electrospray ionization mass spectrometry (ESI-MS) mass spectroscopy. The IC50 value of purified peptide was 87.0 μM. Moreover, the peptide did not exhibit cytotoxic effects on human dermal fibroblast cell lines.

        시판 쑥차 제품의 여드름 균에 대한 생육억제 효과 및 tyrosinase와 collagenase 저해 활성

        본 연구에서는 시판되고 있는 쑥차 제품의 항여드름균 활성과 피부 노화와 관련 있는 항산화 효능, collagenase 저해 활성 등에 대한 연구를 통하여 이 추출물들 을 여드름 치료제로서의 이용가능성을 확인하고자 하였다. 시판 쑥차 10종의 제품의 여드름 원인균에 대한 항균활성을 검정한 결과, C. albicans에 대한 생육억제효과를 나타내는 쑥차 제품은 8제품에서, P. acnes는 8제품, S. aureus는 8제품 그리고 S. epidermidis는 7제품에서 생육억제효과를 나타냈다. Tyrosinase 저해활성을 분석한 결과, 제품간의 활성은 22.6~67.6%의 범위를 나타냈다. 거문도산 쑥차(67.0 ± 9.6%) 및 티백차(64.6 ± 12.2%) 그리고 산청산 쑥차(66.5 ± 8.3%) 제품에서 tyrosinase 저해 활성이 상대적으로 높게 나타났다. 쑥차 제품의 collagenase 저해활성은 제품 간에 20.4~50.7%의 범위를 나타냈다. Collagenase 저해활성은 진도산, 신안산 그리고 강화도산 쑥차 제품에서 상대적으로 높게 나타났지만, tyrosinase 활성이 비교적 높았던 거문도 쑥차 및 티백 그리고 산청산 쑥차 제품의 collagenase 저해활성은 상대적으로 낮게 나타났다. 본 연구에서는 총 페놀성 화합물이 tyrosinase, collagenase 저해효과 및 여드름 균의 생육억제 효과에 있어서 상관관계를 형성하지 않았고 쑥차의 제품에 대해 각각 특이적인 생리활성을 나타내었다. Mugwort is an important food and medicinal resource in Korea. To confirm the biological activities of mugwort tea, we investigated its antimicrobial activity and the inhibitory activity of tyrosinase and collagenase. The antibacterial effects showed that most mugwort tea products exhibit potency against acne-inducing bacteria such as Candida albicans, Propionibacterium acnes, Staphylococcus aureus, and Staphylococcus epidermidis. Furthermore, the antimicrobial activities against acne-inducing bacteria of GT, GB, MC, MD, and MH were higher than that of any other mugwort tea products. The tyrosinase inhibitory activity of GT (67.0 ± 9.6%), GB (64.6 ± 12.2%), and MF (66.5 ± 8.3%) showed remarkably high activity. Still, the collagenase inhibitory activity showed that the MA, MB, and MC exhibit higher activity than that of the other mugwort tea products.

        Anti-wrinkle and anti-inflammation effects of Camellia sinensis seed extracts

        The aim of this study was to investigate toxicity, elastase inhibitory activity, collagenase inhibitory activity, procollagen biosynthesis, hyaluronidase inhibitory activity and nitric oxide production inhibitory activity for improving anti-wrinkle and anti-inflammation using different extracts obtained from Camellia sinensis seeds. All extracts showed no cytotoxicity to Hs68 cell within 500 μg/mL. Among various extracts, elastase and collagenase inhibitory activities showed the highest inhibitory activity of 58.6% and 69.1% in 500 μg/mL of ethanol extract, respectivey. Procollagen biosynthesis showed high activity at 158.8% and 157.6% at 500 μg/mL of ethylacetate extract and ethanol extract, respectively. Hyaluronidase inhibitory activity showed the highest inhibitory activity at 31.2% in 500 μg/mL of ethylacetate extract, and nitric oxide production inhibitory activity showed high inhibitory activity at 40.4% in 500μg/mL of ethylacetate extract. The hyaluronidase and nitric oxide production was suppressed in ethylacetate extract, and it is considered that it can be applied to the treatment of inflammatory diseases. Taken together, the ethanol and ethylacetate extracts of C. sinensis seeds used in this study can be expected for skin protection and anti-inflammatory effects.

        잔가시 모자반 추출물의 주름 개선 및 미백 효과

        잔가시 모자반 추출물의 피부 노화 억제 효과를 확인하기 위하여 항산화 활성, tyrosinase 저해 활성, collagenase 저해 활성을 측정하였다. 잔가시 모자반 에탄올 및 물 추출물의 폴리페놀 함량은 11.20 mg/g 및 11.70 mg/g으로 차이가 나지 않았다. DPPH radical 소거능에서는 잔가시 모자반 에탄올 및 물 추출물 모두 0.5 mg/ml 농도에서 83.68% 및 79%로 활성이 높게 나타났으며, 금속봉쇄력에서는 에탄올 추출물이 물 추출물보다 높게 나왔으나 대조구인 EDTA보다 활성이 낮게 나타났다. Tyrosinase 및 collagenase 저해 활성은 에탄올 추출물이 물 추출물보다 더 높은 것으로 나타났고, collagenase 저해 활성은 에탄올 추출물이 1 mg/ml 농도에서 80% 이상으로 높은 효소 저해능을 보였다. 따라서, 잔가시 모자반은 에탄올 추출물에서 항산화능, tyrosinase 및 collagenase 저해 활성이 더 높게 나타나 피부 노화 억제를 위한 소재로서 더 효과적으로 이용될 수 있을 것으로 사료된다. Sargassum micracanthum (SM) is a member of the family Sargassaceae and commonly occurs along the coast of Korea. Extracts from SM were evaluated for their antioxidant and collagenase and tyrosinase inhibitory activities based on their total phenolic concentration (TPC), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging effect, and metal-chelating effect. The TPC of SM ethanol and water extracts used in the study was 11.20 and 11.70 mg/g of dry sample, respectively. Both SM ethanol and water extracts had high DPPH radical-scavenging effect. The metal-chelating effect of SM ethanol extract (40.47% at 0.5 mg/ml) was higher than that of water extract (23.28% at 0.5 mg/ml). With regard to the antiwrinkling effect, SM ethanol extract showed collagenase inhibitory activity with an IC50 value of 488.20 μg/ ml. Lastly, regarding the anti-melanogenesis effect, SM ethanol extract showed higher tyrosinase inhibitory activity (45.08% at 5 mg/ml) than that shown by the water extract (21.29% at 5 mg/ml). These results suggest that SM has the potential to be a resource with natural anti-melanogenesis, anti-wrinkle, and antioxidant effects.

        증숙된 더덕 추출물의 피부기능 개선 효과

        In this study, hot water and ethanol, acetone, and hexane extracts of the steamed Deodeok were investigated for use as raw materials for natural cosmetics. Hot water and ethanol, acetone and hexane extracts of steamed Deodeok did not indicate cell toxicity up to 300 ㎍/mL concentration in the MTT assay. The tyrosinase inhibition effects of the ethanol extract were higher than the ascorbic acid. The efficacy of the steamed Deodeok's ethanol extract on the elastase inhibition was similar to that of quercetin at higher concentrations. In conclusion, water, ethanol, acetone and hexane extracts of the steamed Deodeok indicated strong possibility as a whitening and anti-wrinkle functional cosmetic ingredient. Among them, the steamed Deodeok's ethanol extract, which showed excellent activity, is highly available because it has high safety compared to other organic solvent extraction methods used in cosmetics, and has minimal irritation to the skin.
