        봉독이 피부 노화에 미치는 영향

        Background and Objective : Increasing interest in anti-aging and anti-wrinkling agents for the skin has triggered the recent outflow of researches and studies in this field. This study was designed to investigate the effects of bee venom on skin wrinkling and skin aging by testing the skin wrinkling, skin elasticity, trans-epidermal water loss (TEWL), free radical level, anti-oxidative agent level, and skin tissue after infusion of bee venom on hairless mouse. Materials and Methods : Fifteen hairless mice aged between 36~40 weeks were divided randomly into 3 Group; the Bee Venom Syringe Group, the Bee Venom Needle Group, and the control group. The Bee Venom Syringe Group were injected subcutaneously with bee venom (0.1cc in total) using an insulin syringe on three spots in the lumbar spine (one spot on the center and two spots 1~2cm to the side bilaterally). The Bee Venom Needle Group were pricked with bee venom-smeared acupuncture needles on three longitudinal spots in the lumbar spine each 1cm apart, after which the needles were removed 10 minutes later. The Control Group did not receive any form of intervention. All procedures took place thrice a week for four weeks, during which the mice were allowed free access to water and fodder. The mice were measured and compared in the weight, skin wrinkling scale, skin elasticity, and TEWL before and after the experiment. After the experiment, blood samples were taken to measure the free radical and anti-oxidative agent level, and the skin tissue was sliced for examination. Data was analyzed using the SPSS program (ver 12.0). The ANOVA analysis was used to compare and contrast the three groups, and t-test for paired samples was used to evaluate skin-wrinkling before and after experiment. The cut-off p-value of significance was set at p<0.05. Results : 1. Administration of bee venom did not cause serious weight loss or gain. 2. Compared to the control group, the Bee Venom Syringe Group and the Bee Venom Needle Group both showed a decrease in skin wrinkling scale after intervention. Especially, the Bee Venom Syringe Group showed a significant decrease (p<0.05). 3. Compared to the control group, the Bee Venom Syringe Group and the Bee Venom Needle Group both showed an increase in skin elasticity. Especially, the Bee Venom Syringe Group showed a significant increase (p<0.05). 4. No significant change in TEWL was found in the mice in all the three groups before and after experiment. 5. Free radical level was normal in all 15 mice in all the three groups, and anti-oxidative agent was not significantly different across the three groups. 6. The Bee Venom Syringe Group, the Bee Venom Needle Group, and the control group did not show any significant difference in the thickness of epidermis and dermis, infiltration of inflammatory cells, and skin wrinkling. The epidermis layer was relatively better preserved in the Bee Venom Syringe Group as compared to the Bee Venom Needle Group and the control group. Conclusion : Direct injection of bee venom on the hairless mouse using a syringe was found to improve wrinkling of the skin and increase skin elasticity but did not show effectiveness on skin dryness due to water loss. The bee venom appears to have suppressive effects on skin wrinkling, one of the symptoms of skin aging, through a process independent of suppression of free radicals or increase of anti-oxidative agent.

        봉독약침(蜂毒藥鍼)이 척수내(脊髓內) 통증관련(痛症關聯) 신경세포(神經細胞)의 활성(活性)에 미치는 영향(影響)

        Introduction : In spite of the use of Bee Venom aqua-acupuncture in the clinics, the scientific evaluation on effects is not enough. Bee Venom aqua-acupuncture is used according to the stimulation of acupuncture point and the chemical effects of Bee Venom. The aims of this study is to investigate the analgegic effects of the Bee Venom aqua-acupuncture, through the change of writhing reflex and the change of c-fos in secondary neurons in the spinal cord. Materials and Methods : Pain animal model was used acetic acid method. The changes of writhing reflex of the mice which were derived pain by injecting acetic acid into the abdomen, after stimulating Bee Venom aqua-acupuncture on Chungwan(CV12) were measured. We used Fos immunohistochemical technique to study the neuronal activity in the spinal cord. Results : 1. Expression of c-fos in superficial dorsal horn(SDH), nucleus proprius(NP) and neck of dorsal hom(N) on 6~9th thoracic spine decreased significantly at $2.5{\times}10-4$g/kg Bee Venom aqua-acupuncture, compared with saline-acetic acid group. 2. The numeral change of Fos-LI neurons on the NP, N, and ventral gray(V) on 6-9th thoracic spine, SDH on 9-11th thoracic spine, and SDH and V on 11~13th thoracic spine decreased significantly at Chungwan(CV12) Bee Venom aqua-acupuncture, compared with saline-acetic acid group. 3. The correlation between the numbers of writhing refleax and Fos-LI neurons in T6-13 segment was statistically statistically significant at Chungwan(CV12) Bee Venom aqua-acupuncture. Conclusion : This study shows that the Bee Venom aqua-acupuncture on Chungwan(CV12) decreases the numbers of Fos-LI neurons. As the analgegic effects of Bee Venom aqua-acupuncture is recognized. Bee Venom aqua-acupuncture treatment is expected for pain modulation. In order to use it in many ways, more researches are needed for the dose and stability of Bee Venom aqua-acupuncture.

        봉약침(Bee Venom)과 Sweet Bee Venom의 Allergy 반응에 대한 비교연구

        Objectives : Sweet bee venom is made by removing allergen from the bee venom through gel filtration chromatography and propionic acid/urea polyacrylamide gel electrophoresis. The aim of this study was to verify allergy inhibitory action in Sweet Bee Venom in which the allergy causing enzyme is removed. Methods : 95 healthy adult men and women were selected through a survey whom had never received the bee venom therapy in the past. The concentration of bee venom pharmacopuncture and Sweet BV pharmacopuncture was equally at 0.1mg/ml and the experiment was conducted as the double blind test. Experiment groups were classified into low dosage groups(0.1ml for both bee venom pharmacopuncture and Sweet BV) and high dosage groups where 0.4ml of respective administrations were rendered made observations for allergic responses. Results : Participants of the study was comprised of 71 men and 24 women with the average age of 29.0 years. According to results of the low dosage groups, Sweet BV group showed significant reduction in pain after 4 hours and 24 hours compared to the bee venom pharmacopuncture group. Other allergic responses were insignificant between the groups. For the high dosage groups, Sweet bee venom group showed reduction in pain after 30 minutes and 4 hours. Other allergic responses such as edema, itchiness, dizziness from hypersensitivity, and fatigue were significantly lower in the Sweet bee venom administered group after 30 minutes. Conclusions : As a result of removed allergen, Sweet bee venom significantly inhibits allergic responses both locally and throughout the body. This indicates wider and easier application of Sweet bee venom for the symptoms applicable to the bee venom pharmacopuncture. Further comparative studies should be conducted to yield more objective verification.

        Bee Venom 및 Melittin 약침액(藥鍼液)이 신경아세포종(神經芽細胞腫)의 세포사(細胞死)에 미치는 영향(影響)

        Objective : This study was designed to analyze the effects of bee venom and melittin on cell death in neuroblastoma cell line. Methods : MTT assay, morphologic method, DNA fragmentation, flow cytometry, immunocytochemistry analysis, RT-PCR and Western blot were performed. Results : The obtained results are summarized as follows: 1. The MTT assay demonstrated that neuroblastoma cell viability was significantly inhibitted dose-dependently by treatment with bee venom and melittin in comparison with control. 2. Cell culture demonstrated that control group proliferated highestly at he 5th day in comparison with the 4th day in bee venom and melittin group. And in bee venom and melitti group cell proliferation decreased 2.5 times than control group. 3. The morphologic study demonstrated that neuroblastoma cell showed apoptosis after treatment with bee venom and melittin for 6 hours using microscope. 4. The Flow cytometry demonstrated that apoptosis of neuroblastoma cell treated with bee venom and melittin was related with stop of cell cycle in stage of $G_0/G_1$. 5 .DNA fragmenation demonstrated that neuroblastoma cell treated with bee venom and melittin showed DNA ladder below 1 Kbp. 6. Immunocytochemistry assay demonstrated that Fos and MAPK which are related with cancer were down-regulated by treatment with bee venom and melittin. 7. RT-PCR analysis demonstrated that Fos and MAPK mRNA were transcripted. Fos was down-regulated form treatment with $5{\mu}g/ml$ bee venom and MAPK was down-regulated form $1{\mu}g/ml$ bee venom. 8. Western blot demonstrated that Fos was down-regulated from $1{\mu}g/ml$ bee venom whereas MAPK was expressed by $1{\mu}g/ml$ bee venom but down-regulated by $10{\mu}g/ml$ bee venom. Conclusions : We found that some cancer related genes ware down-regulated by treatment with bee venom and melittin. Further study is needed for investigating the anti-cancer effect of bee venom and melittin.

        봉독과 Sweet Bee Venom의 항균 및 항산화능 비교연구

        Objectives : This study was conducted to compare antibacterial activities and free radical scavenging activity between the Bee Venom and Sweet Bee Venom in which the allergy-causing enzyme is removed. Methods : To evaluate antibacterial activities of the test samples, gram negative E. coli and gram positive St. aureus were compared using the paper disc method. For comparison of the antioxidant effects, DPPH(1,1-diphenyl-2picrylhydrazyl) free radical scavenging assay and Thiobarbituric Acid Reactive Substances(TBARS) assay were conducted. Results : 1. Antibacterial activity against gram negative E. coli was greater in the Sweet Bee Venom group than the Bee Venom group. 2. Antibacterial activity against gram positive St. aureus was similar between the Bee Venom and Sweet Bee Venom groups. 3. DPPH free radical scavenging activity of the Bee Venom group showed 2.8 times stronger than that of the Sweet Bee Venom group. 4. Inhibition of lipid peroxidation of the Bee Venom group showed 782 times greater than that of the Sweet Bee Venom group. Conclusions : The Bee Venom group showed outstanding antibacterial activity against gram positive St. aureus, and allergen-removed Sweet Bee Venom group showed outstanding antibacterial activity against both gram negative E. coli and gram positive St. aureus. For antioxidant effects, the Bee Venom was superior over the Sweet Bee Venom and the superiority was far more apparent for lipid peroxidation.

        봉독(蜂毒)의 연구 경향에 대한 분석 -국내 학위논문 중 봉독에 관한 연구를 중심으로-

        Objectives : To research the trend of the study related to bee venom and to establish the hereafter direction for the study on bee venom therapy. Methods : I referred to national assembly library and national library of Korea, with `bee venom`, `apitoxin`, `apitherapy`, `bee sting`. After collecting all the theses about bee venom published by domestic university from 1992 to 2005, I figured out number and percentage of theses according to university, year, major field of study, subject. In case of subject, I classified and analyzed it with anti-inflammation effect;travail effect;immunity relation;safe treatment & prevention of side effect of bee venom. Results : The following results were obtained in this study. 1. In the arrangement by the year, the theses started to publish after 1992, and the number of theses has been increasing rapidly since 2000. And it has been decreasing since gaining the summit at 2003. 2. Classified by university, Kyung-Hee University published the most theses, and other theses were published by Kyung-Won University, Dong-Guk University, Kyoung-San University, Seoul National University order. 3. Classified by the major field of study, number of theses was the most by 69 in oriental medicine, next, it was 8, 2 in veterinary science and medicine. 4. Analyzed by the subject, numbers of theses that is related to anti-inflammation effect are 39(50%), numbers of theses related to travail effect are 15(18%), numbers of theses related to immunity are 19(23%), numbers of theses related to safty are 7(8%). 5. Theses that is related to anti-inflammation effect ; travail effect of bee venom shaded to mechanism study inclination after 2002 in primitive effect proof study inclination. Theses that is related to immunity of bee venom shaded to anticancer-related research inclination after 2000 in primitive immunity promotion effect study inclination. 6. In the study for safe treatment & prevention of side effect of bee venom, there were reports bee venom secures when it is injected in wide region, by low concentration, in 3 days cycle. 7. Theses related to bee venom are for the most part experiment theses by 76. Conclusions : As see in above result, safety of bee venom was proved, I think that the more clinical access by using bee venom should be achieved because objective, scientific basis was readied for effect of anti-inflammation, travail, immunity promotion, anticancer etc.

        요각통 환자에 대한 Bee Venom과 Sweet Bee Venom의 효능 비교 연구

        Objective The aim of this study is to investigate if Sweet Bee Venom therapy has the equal effect in comparison with Bee Venom Therapy on Low back pain with Radiation pain. Methods Clinical studies were done 24 patients who were treated low back pain with radiation pain to Dept. of Acupuncture & Moxibusition, of Oriental Medicine Se-Myung University from April 1, 2007 to September 30, 2007. Subjects were randomly divided into two groups ; Bee Venom treated group(Group A, n=10), Sweet Bee Venom treatred group(Group B, n=14). In Bee Venom treated group(Group A), we treated patients with dry needle acupuncture and Bee Venom therapy. In Sweet Bee Venom treatred group(Group B), we treated patients with dry needle acupuncture and Sweet Bee Venom therapy. All process of treatment were performed by double blinding method. To estimate the efficacy of controlling pain. we checked Visual Analog Scale(VAS). For evaluating functional change of patients, Straight Leg Raising Test(S.L.R.T) was measured. Results 1. In controlling pain, Sweet Bee Venom treatred group(Group B) had similar ability in comparison with Bee Venom treated group(Group A). 2. In promoting function, Sweet Bee Venom treatred group(Group B) had similar ability in comparison with Bee Venom treated group(Group A). Conclusions It may be equal effects as compared with using Bee Venom to treat low back pain with radiation pain using Sweet Bee Venom. We can try to treat other disease known to have effect with Bee Venom.

        Bee Venom과 Sweet Bee Venom이 지방세포 대사에 미치는 영향

        Objectives The purpose of this study was to investigate the effects of Bee Venom and Sweet Bee Venom to the primary cultured preadipocyte, adipocytes, and localized fat tissue. Methods Decreased preadipocyte proliferation and decreased lipogenesis are mechanisms to reduce obesity. So, preadipocytes and adipocytes were performed on cell cultures using Sprague-Dawley Rats and treated with 0.01-1mg/ml Bee Venom and Sweet Bee Venom. And porcine skin including fat tissue after treated Bee Venom and Sweet Bee Venom according to the dosage dependent variation are investigated the histologic changes after injection of these Pharmacopuncture. Result Following results were obtained from the preadipocyte proliferation and lipolysis of adipocyte and histologic investigation of fat tissue. 1. Bee Venom and Sweet Bee Venom showed the effect of decreased preadipocyte proliferation depend on concentration. 2. Bee Venom and Sweet Bee Venom showed the effect of decreased the activity of glycerol-3-phosphate dehydrogenase(GPDH) significantly. 3. Bee Venom was not showed the effect of lipolysis, but Sweet Bee Venom was increased in low dosage and decreased in high dosage. 4. Investigated the histologic changes in porcine fat tissue after treated Bee Venom and Sweet Bee Venom, we knew that these Pharmacopuncture was activated nonspecific lysis of cell membranes depend on concentration. Conclusion These results suggest that Bee Venom and Sweet Bee Venom efficiently induces decreased proliferation of preadipocyte and lipolysis in adipose tissue.

        봉독약침(蜂毒藥鍼)이 Writhing reflex 및 척수내(脊髓內) 통증관련(痛症關聯) 신경세포(神經細胞)의 활성(活性)에 미치는 영향(影響)

It showed that the writhing reflex were appeared on the groups which injected aceticacid only, and saline-acetic acid group(sample I), but not on the group bee venom-saline group(sample II). 2. The change of writhing reflex by Chungwan($CV_{12}$) Bee Venom aqua-acupuncture showed significant decrease in the order of Chungwan($CV_{12}$) Bee Venom aqua-acupuncture group III($2.5{\times}10^{-3}$g/kg), II($2.5{\times}10^{-4}$g/kg), and I($2.5{\times}10^{-5}$g/kg), compared with control group. There were significant decrease of number of writhing reflex in $5{\sim}10,\;10{\sim}15\;and\;15{\sim}20$ minutes intervals of Chung wan($CV_{12}$) Bee Venom aqua-acupuncture group I, and in $0{\sim}5,\;5{\sim}10,\;10{\sim}15\;and\;15{\sim}20$ minutes intervals of II and III, compared with control group. 3. The change of writhing reflex by non acupuncture point Bee Venom aqua-acupuncture showed significant decrease in the $0{\sim}5\;and\;5{\sim}10$ minutes intervals and the total number of writhing reflex in $2.5{\times}10^{-4}$g/kg group, compared with control group 4. The effects of writhing reflex of Chungwan($CV_{12}$) Bee Venom aqua-acupuncture group showed significant decrease, compared with non acupuncture point Bee Venom aqua-acupuncture group. Conclusion : This study shows that the Bee Venom aqua-acupuncture on Chungwan($CV_{12}$) decreases the numbers of writhing reflex. As the analgegic effects of Bee Venom aqua-acupuncture is recognized. Bee Venom aqua-acupuncture treatment is expected for pain modulation.         급성기 족관절 염좌 환자의 봉약침 시술 효과에 대한 임상적 연구

        Objectives : This study was to evaluate the clinical effectiveness of Bee Venom acupuncture therapy for acute ankle sprain. Methods : From September 1st, 2005 to December 31st, 2005, the 60 patients who had visited Conmaul oriental medical hospital with acute ankle sprain were sampled into 2 groups for this study; one group for the bee-venom acupuncture therapy combined with needle acupuncture therapy and the other group for needle acupuncture therapy only at least 3 times respectively. Both group had been treated with same additional infrared lamp, physical therapy and ankle supporter during the whole treatment period. Among 60 patients, 20 participants satisfied the whole inclusion criteria. To estimate the efficacy of treatments, Numerical Rating System(NRS) and Ankle-Hindfoot Scale(AHS) were applied before 1st and 4th treatment. Each score results were analysed and compared by Mann-Whitney test and Wilcoxon signed rank test with the level of 5% significance. Results : The NRS score in bee-venom acupuncture therapy group was increased significantly and the AHS score in bee-venom therapy group was decreased significantly compared to each of single acupuncture therapy group(P<0.01). The score change comparisons between the two groups had no significance before and after the treatment. Conclusion : Both bee-venom acupuncture therapy and single acupuncture therapy were effective to treat the acute ankle sprain. but there were no significant data to prove that bee-venom acupuncture therapy is more effective than single acupuncture therapy.
