        Molecular Cloning and mRNA Expression a Glutathione S-Transferase cDNA from the Spider, Araneus ventricosus

        A fat body-specific glutathione S-transferase cDNA was cloned from the spider, Araneus ventricosus. The cDNA encoding A. ventricosus glutathione Stransferase (AvGST) is 645 base pairs long with an open reading frame of 215 amino acid residues with a calculated molecular weight of approximately 24 kDa. Northern blot analysis showed the tissue-specifically expression of AvGST in the A. ventricosus fat body.

        Molecular Cloning of a cDNA Encoding a Ferritin Subunit from the Spider, Araneus ventricosus

        We report for the first time the cDNA sequence encoding a ferritin subunit from the spiders Araneus ventricosus. The complete cDNA sequence of A. ventricosus ferritin subunit comprised 516 bp with 172 amino acid residues. The A. ventricosus ferritin subunit cDNA contained a conserved iron responsive element sequence in the 5 untranslated region. An alignment of the deduced protein sequence of the A. ventricosus ferritin subunit gene to that of other heavy chain ferritin molecules showed that A. ventricosus ferritin subunit is most similar to the great pond snail, Lymnaea stagnalis, ferritin with 70.2% of protein sequence identity.

        Molecular cloning and characterization of the partial major ampullate silk protein gene from the spider Araneus ventricosus

        Spider silks have great potential as biomaterials with extraordinary properties. Here, we report the cloning and characterization of the major ampullate silk protein gene from the spider Araneus ventricosus. A cDNA encoding the partial major ampullate silk protein (AvMaSp) was cloned from A. ventricosus. An analysis of the cDNA sequence shows that AvMaSp consists of a 240 amino acid repetitive region and a 99 amino acid C-terminal non-repetitive domain. The peptide motifs that were found in the spider major ampullate silk proteins, (A)n, (GA)n, and (GGX)n, were conserved in the repetitive region of AvMaSp. Phylogenetic analysis further confirmed that AvMaSp belongs to the spider major ampullate spidroin family of proteins. The AvMaSp-R cDNA, which encodes the 240 amino acid repetitive domain, was expressed as a soluble 22 kDa polypeptide in baculovirus-infected insect cells. Recombinant AvMaSp-R was degraded abruptly by trypsin. However, AvMaSp-R was stable at $100^{\circ}C$ for at least 30 min. Additionally, the AvMaSp-R was stable at pH values from 2 to 12 for at least 1 h. Taken together, our findings describe the molecular structure and biochemical properties of the A. ventricosus major ampullate silk protein and demonstrate its potential as a biomaterial.

        Molecular cloning and characterization of two peptide toxins from the spider Araneus ventricosus

        Spider toxins have great potential in the development of biopesticides. Here, we report the molecular cloning and characterization of two peptide toxins from the spider Araneus ventricosus. Two cDNAs encoding peptide toxins were cloned from A. ventricosus. Analysis of the cDNA sequence shows that the mature peptides of AvT-39 and AvT-48 consist of 39-amino acid residues and 48-amino acid residues, respectively. Both of the mature peptides include six conserved cysteine residues and a principal structural motif typical of spider toxins. The AvT-39 and AvT-48 cDNAs, which encode the mature peptide, were expressed in baculovirusinfected insect cells. AvT-39 and AvT-48 expression in insect cells significantly decreased cell viability. Additionally,the median lethal time (LT50) of Spodoptera exigua larvae inoculated with recombinant AcNPV expressing AvT-48 was approximately 1 day shorter than that of larvae expressing wild-type AcNPV, demonstrating that the recombinant virus reduced LT50 by approximately 25%. Taken together, our findings describe the molecular characterization of two peptide toxins from A. ventricosus and demonstrate the potential for these toxins to be used as biopesticides.

Sequence analysis of randomly selected cDNA clones was performed to obtain genetic information on the spider A. ventricosus, of which genetic information is currently not available. We have partially sequenced randomly selected 385 clones of the cDNA library constructed from A. ventricosus. This expressed sequence tags (EST) analysis revealed 383 genes had high homologies to known genes in GenBank. In this report, 241 independent genes were analyzed in detail.

        Microstructural Organization of the Central Nervous System in the Orb-Web Spider Araneus ventricosus (Araneae: Araneidae)

        Although the geometrical difference in body position between web-building and wandering spiders could affect the organization of their central nervous system (CNS), however most of our informations about spider's CNS are dependent on those revealed from the wandering spiders. Therefore, this paper describes microstructural organizations of the CNS in the geometric orb-web spider Araneus ventricosus. Similarly to other wandering spiders, the CNS of A. ventricosus is also consisted of a dorsal supraesophageal ganglion and a ventral subesophageal mass. The supraesophageal ganglia are fused together and made up of a large sized nerve cell clusters, whereas the subesophageal ganglia are made up of the foremost part of the ventral nerve cord. It has been revealed that the only nerve arising from the supraesophageal mass was the optic nerve which connected with four pairs of eyes, whereas a pair of pedipalpal and four pairs of appendage nerves including abdominal nerve pairs were arisen from the subesophageal nerve mass. Fibrous masses are highly organized into longitudinal and transverse tracts, and are only consisted of processes of neurons and the terminal ramnifications of peripheral sensory neurons. In addition, central fibrous mass of both the brain and the subesophageal mass are totally devoid of nerve cell bodies.

        탈피에 따른 산왕거미(Araneus ventricosus) 과립혈구의 미세구조 변화

        탈피에 따른 거미 과립혈구의 미세구조 변화를 고배율의 투과 전자현미경으로 관찰하였다. 산왕거미(Araneus ventricosus)의 과립혈구는 산호성 과립혈구(EGs)와 염기호성 과립혈구(BGs), 그리고 혈색소혈구 (cyanocytes) 등 세 종류가 구분되었다. EGs와 BGs는 공통적으로 전자밀도가 높은 구형 과립을 함유하고 있었지만, 과립의 크기와 미세구조의 차이에 따라 두 종류가 확연히 구분되었다. 과립의 크기가 작은 EGs는 탈피기간중 세포소기관의 다양한 형태적 변화가 나타나며, 왕성한 식작용을 통해 많은 식포를 형성하는 것으로 관찰되었다. 반면, 과립의 크기가 큰 BGs의 세포질에서는 식포가 관찰되지 않았으나, 탈피시기 동안 과립의 전자밀도가 낮아지는 특성이 관찰되었다. 가장 대형의 과립혈구인 혈색소세포의 세포질에는 많은 헤모시아닌 결정체가 동심원상으로 배열되어 있었으며, 탈피기간중 혈구의 일부는 불규칙한 구조로 변형되었다. 고배율의 투과전자현미경으로 헤모시아닌 결정체의 격자구조를 관찰한 결과, 미세소관(microtubule)과 매우 유사한 구조를 가지고 있었으며, 각각의 미세한 관상 구조는 다시 약 20여 가닥의 극히 미세한 필라멘트로 이루어져 있음이 확인되었다. The fine structural modification of the granulocytes between the molt and intermolt period were investigated by the transmission electron microscopy. The granular hemocytes of the spider Araneus ventricosus were composed of three subtypes: eosinophilic granulocytes (EGs), basophilic granulocytes (BGs) and cyanocytes. Both of the EGs and BGs have electron dense granules within their cytoplasms, however the granules of BGs are larger than those of EGs. During the molt period, some of the EGs have fine structural modification in their cell organelles including formation of phagosomes as a result of active phagocytosis. However, the BGs have no phagosomes, but electron densities of the granules are changed to lower states than the intermolt period. The cyanocyte is the biggest hemocyte among the granulocytes. They contain numerous hemocyanin crystals in the cytoplasm with some electron-lucent vacuoles. During the molt period, some of the cyanocytes are changed to irregular shapes. High magnification electron micrographs reveal that the lattice sub-structure of the hemocyanin crystals are very similar to those of microtubules, and each tubule is composed of approximately 20 filaments with fine fibrillar structure.
