        쌀 배아와 흑미 미강 색소 첨가 배아젤리가 고콜레스테롤 식이 흰쥐의 지질대사와 항산화효소 활성에 미치는 영향

        We investigated the effect of rice embryo and embryo jelly with black rice bran pigment on lipid metabolism and antioxidant activity. Thirty 4-week-old male Sprague-Dawley rats were fed high cholesterol diets supplemented with 15% rice embryo and 25% embryo jelly added black rice bran pigment, respectively, for 6 weeks. Plasma and hepatic lipid profile, lipid peroxidation, and the activity of antioxidant scavenger enzymes in liver were examined. Supplementation with rice embryo and embryo jelly had no effect on food intakes in high cholesterol-fed rats. The plasma triglyceride concentration was not significantly different among the groups. Supplementation with rice embryo and embryo jelly resulted in lower plasma and hepatic total cholesterol (TC) concentration and high-density lipoprotein-cholesterol (HDL-C)/TC ratio and atherogenic index compared to the control group, while the plasma HDL-C concentration tended to elevated. Rice embryo and embryo jelly tended to lower plasma and hepatic levels of thiobarbituric acid reactive substances than the control group. Moreover, hepatic antioxidant enzyme activities, including superoxide dismutase and glutathione peroxidase, were significantly higher in the rice embryo and embryo jelly groups. In conclusion, rice embryo and embryo jelly was very effective in improving the lipid metabolism and reducing oxidative stress by up-regulating the hepatic antioxidant enzymes in high cholesterol-fed rats.         식물유래 천연색소의 항산화 활성

        The objective of this study was to determine the effect of antioxidant enzyme activity and radical scavenging activities of thirteen kinds of natural plant pigments. The analytic method of antioxidant activities were measured by estimating DPPH free radical scavenging and nitrite scavenging ability activity. The free radical scavenging activity by method using stable free radical DPPH was the highest in the red cabbaged pigment. Addition of ethanol extract 1㎎/㎖ from onion peel pigment displayed remarkable effect on nitrite scavenging ability about 91.9%. Antioxidative enzyme activity was evaluated in terms of superoxide dismutase(SOD), catalase(CAT), and ascorbate peroxidase(APX) activity. The bitter melon pigment had the highest SOD activity of 87.3%. The activities of CAT and APX were higher in the mulberry leave pigment compared with other natural plant pigments. In contrast, CAT activity of plant pigment samples were unaffected. These results suggest that natural plant pigment had the potent biological activities such as antioxidant enzyme activities, and that their activities exhibited differently depending on each kind of pigments. 선발된 13종의 천연색소 추출물에 대하여 free radical 소거활성 및 아질산염 소거활성, 그리고 항산화효소 활성에 대하여 조사하였다. 천연색소 종류별 free radical 소거활성을 보면 적양배추(red cabbage), 양파껍질(onion peel), 레드비트(redbeet), 흑미(black rice), 오디(mulberry) 색소 등에서 상대적으로 높게 나타났고, 청치자(blue gardenia) 및 황치자(yellow gardenia), 완숙여주(mature bitter melon) 색소 등에서는 비교적 낮은 활성을 보였다 또한 아질산염에 대한 소거능을 조사한 결과, 반응용액 pH 1.2의 경우 양파껍질, 레드비트, 치자황, 지황(chinese foxglove), 치자청 색소에서 높은 소거 활성을 나타냈으며, 적양배추와 오디 색소에서는 상대적으로 낮은 소거활성을 보였다. 그리고 반응용액 pH 6.0에서는 대부분의 색소 추출물에서 아질산염 소거활성을 나타내지 않았다. 그리고 항산화효소 활성 결과를 보면, SOD 활성의 경우 여주, 흑미, 포도과피 등에서 높은 활성을 나타냈고, 자색고구마, 지황에서도 비교적 높은 활성을 보였다. 또한 APX 활성은 뽕잎, 오디, 지황의 순으로 높은 활성을 보였으며, CAT 활성도 APX 결과와 비슷한 양상으로 나타났다. 이는 천연색소에 있어 항산화 효소의 종류에 따라 그 활성에 차이가 있음을 알 수 있다. 본 연구결과를 통해 다양한 생리활성을 지니고 있는 천연식물색소에 대해 높은 항산화 기능성을 확인함으로서 향후 고부가가치의 향장소재 및 식품소재로서의 개발가능성이 매우 높음을 시사해 주고 있다.

        솔순 열수 추출물의 생리활성

        본 연구에서는 솔순의 기능성을 규명하기 위해 열수 추출물을 대상으로 여러 생리활성과 아질산염 소거작용에 대하여 분석하였다. 우선, 솔순의 항산화 활성을 측정하기 위하여 DPPH, SOD 유사활성, XO 저해활성을 측정하였다. 그 결과 DPPH법을 통해 측정한 솔순의 항산화력은 2 ㎎/mL에서 71.4%의 radical 소거능을 나타내었으며, SOD 유사활성은 2 ㎎/mL 농도에서 85.4%로 비교적 높은 활성을 나타내었다. Xanthine oxidase 저해활성 측정 실험에서는 2 ㎎/mL에서 70.9%의 높은 저해활성을 나타내었다. 솔순 열수추출물의 아질산염 소거능 측정 실험에서는 pH 1.2와 3.0에서 53~59%, pH 6.0에서 40%의 소거능을 나타내었다. 항고혈압 측정실험에서는 시판되는 항고혈압제 captopril과 비교분석 하였는데, 0.1 ㎎/mL에서 93.0%였고 같은 농도의 솔순 열수 추출물은 14.6%의 낮은 저해율을 나타냈다. 하지만, 1 ㎎/mL에서 44.7%의 저해율을 나타내어 솔순 열수 추출물의 비교적 양호한 저해활성을 보였다. 혈당강하능의 지표로 α-glucosidase 활성 억제능을 분석한 결과 솔순 열수 추출물 3 ㎎/mL의 농도에서 43.3%의 저해활성을 나타냈으며, 주름 예방 효과의 지표로 elastase 활성 억제능을 측정한 결과 10 ㎎/mL의 농도에서 59.6%의 저해능을 보였다. 이상의 결과들은 솔순 열수 추출물의 우수한 생리활성을 증명하고 있는 것으로 천연의 건강 및 미용 소재로써 그 활용도가 높을 것으로 판단된다. This research was performed to determine the antioxidant activity, nitrite scavenging activity, and its inhibitory activity on angiotensin converting enzyme (ACE), xanthine oxidase, α-glucosidase, and elastase of hot water extract from pine bud (WPB). Antioxidant activity of WPB was measured by 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity and superoxide dismutase (SOD)-like activity. DPPH radical scavenging activity and SOD-like activity of WPB were remarkably increased in a dose-dependent manner, and were about 71.4 and 85.4% at 2 ㎎/mL, respectively. The xanthine oxidase and ACE inhibitory activities were about 70.9 and 51.9% at 2 ㎎/mL of WPB, respectively. Nitrite scavenging activity of WPB was about 59.1, 53.8, and 39.5% on pH 1.2, 3.0, and 6.0 at 2 ㎎/mL, respectively. The WPB also showed elastase and α-glucosidase inhibitory effects. These results revealed that pine bud have strong antioxidant activity and positive effects on the inhibition of xanthine oxidase, ACE, and elastase.

        Antioxidant Enzyme Activity, Iron Content and Lipid Oxidation of Raw and Cooked Meat of Korean Native Chickens and Other Poultry

        This study was conducted to observe antioxidant enzyme activity, iron content and lipid oxidation of Korean native chickens and other poultry. The breast and thigh meat of three Korean native chicken breeds including Woorimatdak, Hyunin black and Yeonsan ogye, and three commercial poultry breeds including the broiler, White Leghorn and Pekin duck (Anasplatyrhyncos domesticus) were studied. The analyses of the antioxidant enzymes activity, iron content and lipid oxidation were performed in raw and cooked samples. The activity of catalase (CAT) in the thigh meat was higher than that of the breast meat of three Korean native chickens and the broiler, respectively. The activity of glutathione peroxidase (GPx) in the uncooked thigh meat of three Korean native chickens was higher than that of the breasts. The breast meat of Woorimatdak and Pekin duck had higher superoxide dismutase (SOD) activity than the others, while only the thigh meat of Pekin duck had the highest activity. Cooking inactivated CAT and decreased the activity of GPx and SOD. The thigh meat of Woorimatdak, White Leghorn, Yeonsan ogye and Hyunin black contained more total iron than the breast meat of those breeds. The heme-iron lost during cooking ranged from 3.2% to 14.8%. It is noted that the thigh meat had higher thiobarbituric acid reactive substances values than the breast in all chicken breeds. Though Woorimatdak showed higher antioxidant enzyme activity and lower released-iron percentage among Korean native chickens, no differences were found on lipid oxidation. We confirm that the dark meat of poultry exhibited higher antioxidant enzyme activity and contained more iron than the white meat.

        밀 배아의 항산화활성에 대한 효소처리 효과

        Purpose: Enzyme as Celluclast 1.5L, which degrades cell wall materials of wheat germ, was applied to stabilized wheat germ to increase the antioxidant activity. Methods: Total phenolic compounds (TPC), total flavonoid contents (TFC) and α-tocopherol contents were measured from the enzyme-treated wheat germ and antioxidant activity was confirmed by the ABTS and DPPH radical scavenging capability. Results: TPC and TFC increased sharply at 6 h after enzyme treatment but did not change significantly from 12 to 24 h. After 30 h, TPC and TFC increased by 4.3 and 3.0 times, respectively, relative to the untreated sample. When the antioxidant activity was compared with ABTS and DPPH radical scavenging capacity, it decreased at the beginning of enzyme treatment, but increased after 30 h of enzyme treatment and 30 h of treatment with Celluclast 1.5L to improve antioxidant activity. Conclusion: From the above results, it is suggested that TPC and TFC should be increased by enzyme treatment of wheat germ, but antioxidant activity should be improved after 30 h-treatment.

        Nitrite Scavenging and Alcohol Metabolizing Activities of Hot Water Extract from Makgeoly and Its Angiotensin Converting Enzyme Inhibitory Effect

        전통발효주 막걸리의 기능성을 증명하기 위하여 S사막걸리로부터 구입한 막걸리 침전물의 여러 가지 생리활성에 대하여 조사하였다. 우선 막걸리 침전물 열수 추출물의 항산화 효과를 측정하기 위해 DPPH radical 소거능과 SOD 유사활성을 측정하였다. 그 결과 DPPH법을 통해 측정한 막걸리 침전물 열수 추출물의 radical 소거능은 10 ㎎/㎖에서 48.0%으로 나타났으며, 농도가 증가함에 따라 유의적으로 증가하는 경향을 나타내었다. 또한 SOD 유사활성은 10 ㎎/㎖ 농도에서 98.7%로 비교적 높은 SOD 유사활성을 보였다. 항고혈압 활성 측정 실험에서는 현재 시판되고 있는 항고혈압제인 captopril은 0.1 ㎎/㎖에서 93.4%의 ACE 억제효과가 나타났고, 막걸리 침전물 열수 추출물 10 ㎎/㎖에서는 74.0%의 높은 저해 활성을 나타내었다. 따라서, 막걸리 침전물 열수 추출물은 인체에 부작용이 적은 천연 항고혈압소재로서 이용가능성이 높은 것으로 사료된다. 아질산염 소거능 측정 실험에서는 positive control인 Vit. C 1 ㎎/㎖의 경우 pH 1.2와 3.0에서는 74~64%, pH 6.0에서는 45%의 소거능을 보인반면, 막걸리 침전물 열수 추출물의 경우 pH 1.2와 3.0에서는 51~42%, pH 6.0에서는 28%의 소거능을 나타내었다. 막걸리 침전물 열수 추출물의 숙취해소 효능은 ADH와 ALDH 활성증진에 막걸리 침전물 열수 추출물이 미치는 영향을 조사함으로써 증명하고자 하였다. 그 결과, 알콜과 acetaldehyde 분해능은 높게 나타났다. 이상의 결과들은 막걸리 침전물의 우수한 기능성으로서의 이용 가능성에 대한 기초자료로 그 가치가 기대된다. In this study, we investigated the antioxidant activities, alcohol metabolizing activities, nitrite scavenging ability, angiotensin converting enzyme (ACE), and elastase inhibitory effects of hot water extract from Makgeoly (HWM). Antioxidant activities were measured by using 2,2 diphenyl 1 picryl hydrazyl (DPPH) free radical scavenging activity and SOD (superoxide dismutase) like activity. The DPPH radical scavenging activity and SOD like activity of HWM were remarkably increased in a dose dependent manner and were 48.0% and 98.7% at 10 ㎎/㎖, respectively. To determine the influence of HWM on alcohol metabolizing activity, the generating activities of reduced nicotinamide adenine dinucleotide (NADH) by alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) were measured. The facilitating rates of ADH and ALDH activity by HWM were remarkably increased in a dose dependent manner and were 70.2% and 64.1% at 10 ㎎/㎖, respectively. The inhibitory activity against angiotensin converting enzyme (ACE) of HWM was increased in a dose dependent manner and was 74.2% at 10 ㎎/㎖. The nitrite scavenging ability of HWM showed the most remarkable effect at pH 1.2 and 2 ㎎/㎖. These results indicated that HWM may have valuable biological properties owing to their antioxidant activities, ADH and ALDH activity, nitrite scavenging ability, and ACE inhibitory activity.

        효소 가수분해 백삼, 홍삼을 첨가한 양갱의 품질특성 및 항산화 활성

        본 연구에서는 백삼 및 홍삼 추출물과 그 효소 가수 분해물을 제조하고 이를 첨가하여 제조된 양갱의 항산화 활성 및 품질 특성을 조사하였다. 백삼과 홍삼 추출물의 효소 가수분해를 위해서는 Rapidase C80 max, Pyr-flo, Ultimase MFC를 선정하였다. Rapidase C80 max, Pyr-flo, Ultimase MFC로 가수분해 한 백삼과 홍삼에서는 효소반응 전에는 검출되지 않았던 ginsenoside F2와 Compound K (CK)가 검출되었으며 특히 홍삼의 2차 효소 반응군에서 CK의 함량이 가장 높았다. 효소 가수분해 전 또는 후의 백삼 및 홍삼을 함유한 양갱을 제조한 후 폴리페놀 함량, 항산화 능력을 분석한 결과 백삼 및 홍삼을 함유하지 않은 양갱(대조군)에 비해 총 폴리페놀 함량, DPPH 라디칼 소거능, ABTS 라디칼 소거능, FRAP 분석 결과 모두 우수하였으며, 첨가한 백삼 및 홍삼에 효소반응을 진행할수록 유의적으로 항산화 활성이 증가하였다(P<0.05). 밝기(L<sup>*</sup>)는 무첨가 양갱(대조군)이, 적색도(a<sup>*</sup>)는 홍삼양갱(RG)이, 황색도(b<sup>*</sup>)는 백삼양갱(WG)이 높게 나타났고, 조직감은 홍삼 첨가 후 2차 가수분해까지 진행한 양갱(RG-T2)이 강도(hardness), 탄력성(springiness), 씹힘성(chewiness), 응집성(cohesiveness), 검성(gumminess) 모두 유의하게 높은 결과를 나타내었다(P<0.05). 결론적으로, 백삼 또는 홍삼 추출물에 Rapidase C80 max, Pyr-flo, Ultimase MFC를 처리하면 진세노사이드 탈당화에 매우 유용하여 생리활성이 우수한 CK를 생산할 수 있으며, 효소로 가수분해 된 백삼 및 홍삼을 첨가한 양갱은 대조군(인삼 무첨가 양갱)에 비해 총 폴리페놀과 항산화 활성을 유의하게 증가시킨다는 것을 알 수 있었다. 이러한 결과는 향후 기능성 양갱 제조에 우수한 기초자료로 활용될 수 있을 것으로 보인다. This paper investigates the antioxidant activity and quality characteristics of yanggaeng containing white ginseng and red ginseng extracts and their enzyme hydrolysates that were produced for the purpose of the study. White and red ginseng extracts were hydrolyzed using Rapidase C80 max, Pyr-flo, and Ultimase MFC. Ginsenoside F2 and compound K (CK) were not detected in white and red ginseng before enzymic reaction but were detected in white and red ginseng hydrolyzed through Rapidase C80 max, Pyr-flo, and Ultimase MFC, and the content of CK was the highest in the second enzymic reaction group of red ginseng. Upon preparing yanggaeng containing white and red ginseng before or after enzymatic hydrolysis, the polyphenol content and antioxidant abilities were analyzed. The yanggaeng containing enzyme-hydrolyzed white ginseng and red ginseng showed greater total polyphenol content, superior DPPH radical scavenging activity, superior ABTS radical scavenging activity, and superior FRAP analysis results compared to the yanggaeng that doesn't contain white or red ginseng. As the enzymic reaction was performed in the added white and red ginseng, the antioxidant activity increased significantly (P<0.05). In brightness(L<sup>*</sup>), non-additive yanggaeng (control group) was the highest, red ginseng yanggaeng (RG) showed the highest redness(a<sup>*</sup>), and the white ginseng yanggaeng (WG) showed the highest yellowness(b<sup>*</sup>). In terms of texture, the yanggaeng containing red ginseng with second hydrolysis (RG-T2) showed significantly high results in hardness, springiness, chewiness, cohesiveness, and gumminess (P<0.05). In conclusion, treating white and red ginseng with Rapidase C80 max, Pyr-flo, and Ultimase MFC is very useful in ginsenoside deglycosylation and will produce CK with excellent biological activity. It can also be seen that yanggaeng containing white and red ginseng hydrolyzed with enzymes significantly increase total polyphenol and antioxidant activity compared to the control group (yanggaeng with no added ginseng). These results will be useful as excellent foundational data for the production of functional yanggaeng in the future.

        효소처리에 의한 초당옥수수 추출물의 이화학적 및 항산화 특성의 변화

        To improve usability of super sweet corn, extracts were prepared with hydrolytic enzyme and changes in physicochemical and antioxidant properties were analyzed. Soluble solids and reducing sugars contents were higher in all enzyme treatment groups than in the control. When enzyme treatment time increased, contents of soluble solids and reducing sugars were also increased. There was no significant difference in lightness between treatment groups, with redness showing the highest value in the control and yellowness showing the highest value in the invertase treatment group. Free sugar content in the control was the lowest. However free sugar content in the enzyme combination treatment group was increased by more than four times compared to that in the control. Contents of ascorbic acid, flavonoids and polyphenols were higher in the enzyme treatment group than in the control. In particular, the enzyme combination treatment group showed the highest content. DPPH and ABTS radical scavenging abilities were significantly higher in all enzyme treatment groups than in the control. Radical scavenging abilities of cellulase treatment group and enzyme combination treatment group showed high activity. The activity increased when enzyme treatment time increased. The combined enzyme treatment method for super sweet corn was suitable for food processing.

        재래종 대파(Allium fistulosum L.)의 생리 활성 평가

        Purpose: This study was conducted to evaluate physiological activities, such as antioxidant activity and enzyme activity, of hot water extract of the four parts (flower, leaf and pseudostem, stem, and root) of traditional Welsh onion (Allium fistulosum L.) and then to provide basic data on traditional Welsh onion to support future research. Methods: Four parts of traditional Welsh onion plant were extracted with water heated at 80℃. Antioxidative and enzymatic activities, such as total polyphenol content, total flavonoid content, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, 2,2'-azino-bis-3-ethylbenzothiazoline–6-sulfonic acid (ABTS) radical scavenging activity, and the inhibitory activity on α-glucosidase, tyrosinase, and elastase enzymes, were measured. Results: Out of the hot water extract of the different parts, Allium fistulosum flower (AFF) extract had the highest content of antioxidants. In the Allium fistulosum leaf and pseudostem (AFLP), Allium fistulosum stem (AFS), and Allium fistulosum root (AFR) extracts, the total polyphenol content and DPPH radical scavenging activity of the root were high, and AFS had high total flavonoid content and ABTS radical scavenging activity. Regarding enzyme activity, α-glucosidase and elastase inhibitory activities were the highest in AFR, and tyrosinase inhibitory activity was the highest in AFS. Conclusion: We confirmed the physiological activity of the hot water extract of four parts of traditional Welsh onion. Our findings provide basic data for research into traditional Welsh onion, and can facilitate its future development as a functional material.

        UV-B 반복노출에 따른 항산화효소의 변성 및 활성저하와 RGP렌즈의 차단효과

        Purpose: The present study was conducted to reveal the correlation of structural denaturation and decrease of enzyme activity when the antioxidative enzymes, superoxide dismutase (SOD) and catalase (CAT) were repeatedly exposed to UV-B, and further investigate whether the denaturation and inactivation of those enzymes can be effectively blocked by using UV-inhibitory RGP lens. Methods: Each enzyme solution was prepared from the standardized SOD and CAT, and repeatedly exposed to UV-B of 312 nm for 30 minutes, 1 hour and 2 hours a day over 1, 2, 3, 4 and 5 days. Structural denaturation of SOD and CAT induced by repeat UV-B irradiation was confirmed by the electrophoretic analysis, and their enzyme activity was determined by the colorimetric assay using the proper assay kit. At that time, the change in structure and activity of the antioxidant enzymes directly exposed to UV-B was compared to the case that UV-B was blocked by UV-inhibitory RGP lens. Results: SOD exposed repeatedly to UV-B showed the polymerization pattern in the electrophoretic analysis when it repeatedly exposed for 30 min a day, however, the change of its activity was less than 10%. On the other hand, CAT repeatedly exposed to UV-B reduced size and density of the electrophoretic band which indicated a structure denaturation, and its activity was significantly decreased. In the case that the repeat exposure time was longer, CAT activity was completely lost even though some enzyme band occurred in the electrphoretic analysis. In addition, the degeneration of CAT due to UV-B irradiation was inhibited to some extent by using RGP lens with a UV-B blocking of 63.7%, however, it was not completely inhibited. Conclusions: From these results, it was revealed that the structural denaturation of antioxidative enzymes was not perfectly correlated with the reduction in enzyme activity according to the type of enzyme. It is recommended to minimize the exposure time to UV when wearing contact lens, or wear the contact lenses having UV blocking rate of the FDA Class I blocker or the sunglasses having equivalent UV-blocking rate for reducing the damage of antioxidative enzymes induced by UV. 목적: 본 연구는 안구에 존재하는 항산화효소인 superoxide dismutase(SOD)와 catalase(CAT)가 UV-B에 반복적으로 노출되었을 때 유발되는 구조변성 및 활성저하의 상관관계를 밝히며, 자외선 차단 RGP렌즈의 사용으로 효소의 변성과활성저하가 효율적으로 차단되는가를 알아보기 위하여 수행되었다. 방법: SOD와 CAT의 표준품으로 각각의 효소용액을 제조하고 하루 30분, 1시간 및 2시간씩 312 nm의 UV-B에 1, 2, 3, 4 및 5일 동안 반복적으로 노출시켰다. 이 때 UVB에직접 노출시킨 항산화효소의 구조 및 활성변화를 자외선 차단기능이 있는 RGP렌즈로 UV-B를 차단시킨 경우와비교하였다. UV-B 반복노출에 따른 SOD와 CAT의 구조변성은 전기영동분석으로 확인하였으며, 이들 효소활성은 분석키트를 이용하여 비색분석법으로 측정하였다. 결과: UV-B에 반복노출된 SOD는 일일 30분 조사조건으로 반복노출되었을 때에도 전기영동분석에서 효소다중화(polymerization)가 관찰되었으나 활성의 변화는 10% 이내로 나타났다. 반면UV-B에 반복노출된 CAT은 전기영동 시 효소밴드크기나 진하기가 감소하여 구조변성이 나타났음을 확인할 수 있었으며, 반복노출시간이 긴 경우 CAT은 전기영동분석에서 효소밴드를 보임에도 불구하고 그 활성은 완전히 소실되었다. 또한 UV-B 조사로 인한 CAT의 변성은 63.7%의 UV-B 차단기능을 가진 RGP렌즈의 사용으로 어느 정도 억제되었으나완전히 억제되는 것은 아니었다. 결론: 이상의 결과로 UV-B 반복노출에 따른 항산화효소의 구조변성은 그 종류에 따라 효소활성의 감소정도와 반드시 일치하는 것은 아님을 알 수 있었다. 따라서 자외선으로 인한 항산화효소의 손상을막기 위하여서는 콘택트렌즈를 착용한 상태에서 자외선 노출시간을 최소화하거나, FDA Class I 차단제에 해당하는 UV 차단율을 가지는 콘택트렌즈를 착용 또는 이에 상응하는 UV차단율을 가지는 선글라스를 덧착용할 것을 권장한다.
