        로부스타(Coffea Canephora) 커피그린빈의 생리활성에 관한 연구

        본 연구에서는 로부스타(Coffea canephora) 커피그린빈을 활용하기 위해 항산화 및 세포 독성을 확인하여 천연 소재로서의 개발 가능성을 확인하였다. 로부스타 커피그린빈을 열수 및 70% 에탄올 추출물에 대해 항산화 활성을 측정하였다. 총 폴리페놀 함량을 측정한 결과 열수 및 70% 에탄올추출물은 106.74±0.39mg/100g, 136.24±0.1mg/100g을 나타내었다 전자공여능 및 ABTS라디칼 소거능을 측정한 결과, 열수 및 70% 에탄올 추출물은 농도 의존적으로 활성을 나타내었으며, 열수 추출물보다 70% 에탄올 추출물이 우수한 항산화 효과가 있는 것을 확인하였다. MTT assay를 이용하여 세포 독성을 측정한 결과, 100㎍/㎖ 농도 이하에서 독성을 보이지 않는 것을 확인하였다. 이를 통해 로부스타 커피그린빈은 천연 소재로서의 개발 가능성을 가진 것을 시사한다. In this study, antioxidant and cytotoxicity were measured to use Coffea canephora green coffee bean to confirm the potential for development as a natural material. The antioxidant activity of Coffea canephora green coffee bean was measured with hot water and 70% ethanol extract. The total polyphenol content was measured with 106.74 ± 0.39 mg/100 g and 136.24 ± 0.1 mg/100 g of hot water and 70% ethanol extract. Measurements of electron donating ability and ABTS Radical scavenging effect in hot water and 70% ethanol extract showed concentration-dependent increases. It was found that 70% ethanol extract has a better antioxidant effect than hot water. Subsequently, the cytotoxicity was measured using MTT assay and found that it did not show toxicity below 100 ㎍/㎖ concentrations. This suggests that Coffea canephora green coffee bean has potential for development as a natural material.

        원산지별 그린커피빈에 대한 항산화 및 항염증 비교 연구

        In this study, the extracts of green Coffea arabica bean by country of origin were tested as a potential source for natural ingredients in cosmetic products. Antioxidative activities were measured by total polyphenol, 1,1-diphenyl-2-picrylhydrazyl radical (DPPH), and 2,2 -azino-bis (3- ethylbenzothiazoline-6-sulphonic acid) (ABTS) assay. Anti-inflammatory activities were checked by nitric oxide (NO) assay. The total polyphenol content was found to be higher in ethanol extract than in water extracts, and in particular Ethiopia ethanol extracts had 852.39 mg/100 g, making them the highest in content. The antioxidant measurement was measured with DPPH and ABTS, indicating that antioxidant strength increased dependent on the concentration of all extracts to be superior. The cell survival rate of the extracts of green Coffea arabica beans by country of origin was confirmed by MTT assay and was close to 100 ㎍/㎖ to 100%. Anti-inflammatory activity by NO assay showed lipopolysacchride (LPS)-induced NO was significantly inhibited following treatment with extract of green Coffea arabica beans by country of origin and decreased in a concentration-dependent manner. This study confirmed the efficacy of green Coffea arabica beans by country of origin through research related to antioxidants and anti-inflammatory, and confirmed its availability as a natural material for cosmetics.
