Precirrhotic Primary Biliary Cholangitis with Portal Hypertension: Bile Duct Injury Correlate

Background/Aims: The histological characteristics and natural history of precirrhotic primary biliary cholangitis (PBC) with portal hypertension (PH) are unclear. Our aim was to clarify the prevalence, risk factors, and histological characteristics of precirrhotic PBC patients with PH. Methods: This retrospective study compared the clinical features, histological characteristics, and response to ursodeoxycholic acid (UDCA) between the PH and non-PH groups of precirrhotic PBC patients. Results: Out of 165 precirrhotic PBC patients, 40 (24.2%) also had PH. According to histological stage 1, 2 and 3 disease, 5.3% (1/19), 17.3% (17/98), and 45.8% (22/48) of patients also had PH, respectively. Precirrhotic PBC with PH was significantly positively correlated with bile duct loss, degree of cytokeratin 7 positivity, and degree of fibrosis in the portal area, but significantly negatively correlated with lymphoid follicular aggregation. Compared to the non-PH group, patients in the PH group showed a higher prevalence of obliterative portal venopathy, incomplete septal fibrosis, portal tract abnormalities and non-zonal sinusoidal dilatation (p<0.05). In addition, patients with PH were more likely to present with symptoms of jaundice, ascites, epigastric discomfort, a poorer response to UDCA, and more decompensation events (p<0.05). High alkaline phosphatase levels, low white blood cell counts, high Mayo scores, and high FIB-4 index values were risk factors for precirrhotic PBC with PH. Conclusions: Approximately 24.2% of precirrhotic PBC patients have PH, which is histologically related to the injury of bile ducts. High alkaline phosphatase levels, low white blood cell counts, high Mayo scores, and high FIB-4 index values are associated with increased risk of precirrhotic PBC with PH.
Tumor Necrosis Factor-α Gene Polymorphisms and Risk of Oral Cancer: Evidence from a Meta-analysis

Numerous studies have been conducted regarding association between TNF-${\alpha}$ and oral cancer risk, but the results remain controversial. The present meta-analysis is performed to acquire a more precise estimation of relationships. Databases of Pubmed, the Cochrane library and the China National Knowledge Internet (CNKI) were retrieved until August 10, 2013. Pooled odds ratios (ORs) and 95% confidence intervals (95% CIs) were calculated with fixed- or random-effect models. The heterogeneity assumption was assessed by I-squared test. Among the eight included case-control studies, all were focused on TNF-${\alpha}$-308G>A and four also concerned the TNF-${\alpha}$-238G>A polymorphism. It was found that oral cancer risk were significant decreased with the TNF-${\alpha}$-308G>A polymorphism in the additive genetic model (GG vs. AA, OR=0.19, 95% CI: [0.04, 1.00], P=0.05, I2=68.9%) and the dominant genetic model (GG+GA vs. AA, OR=0.22, 95% CI: [0.06, 0.82], P=0.03, I2=52.4%); however, no significant association was observed in allele contrast (G vs. A, OR=0.70, 95% CI: [0.23, 2.16], P=0.54, I2=95.9%) and recessive genetic models (GG vs. GA+AA, OR=0.72, 95% CI: [0.33, 1.57], P=0.41, I2=93.1%). For the TNF-${\alpha}$-238G>A polymorphism, significant associations with oral cancer risk were found in the allele contrast (G vs. A, OR=2.75, 95% CI: [1.25, 6.04], P=0.01, I2=0.0%) and recessive genetic models (GG vs. GA+AA, OR=2.23, 95%CI: [1.18, 4.23], P=0.01, I2=0.0%). Conclusively, this meta-analysis indicates that TNF-${\alpha}$ polymorphisms may contribute to the risk of oral cancer. Allele G and the GG+GA genotype of TNF-${\alpha}$-308G>A may decrease the risk of oral cancer, while allele G and the GG genotype of TNF-${\alpha}$-238G>A may cause an increase.
Effect of Metal (Mn, Ti) Doping on NCA Cathode Materials for Lithium Ion Batteries

<P>NCA (LiNi0.85Co0.10Al0.05-<I>x</I> M<I>x</I>O2, M=Mn or Ti, <TEX>$ x$</TEX> < 0.01) cathode materials are prepared by a hydrothermal reaction at 170°C and doped with Mn and Ti to improve their electrochemical properties. The crystalline phases and morphologies of various NCA cathode materials are characterized by XRD, FE-SEM, and particle size distribution analysis. The CV, EIS, and galvanostatic charge/discharge test are employed to determine the electrochemical properties of the cathode materials. Mn and Ti doping resulted in cell volume expansion. This larger volume also improved the electrochemical properties of the cathode materials because Mn<SUP>4+</SUP> and Ti<SUP>4+</SUP> were introduced into the octahedral lattice space occupied by the Li-ions to expand the Li layer spacing and, thereby, improved the lithium diffusion kinetics. As a result, the NCA-Ti electrode exhibited superior performance with a high discharge capacity of 179.6 mAh g<SUP>−1</SUP> after the first cycle, almost 23 mAh g<SUP>−1</SUP> higher than that obtained with the undoped NCA electrode, and 166.7 mAh g<SUP>−1</SUP> after 30 cycles. A good coulombic efficiency of 88.6% for the NCA-Ti electrode is observed based on calculations in the first charge and discharge capacities. In addition, the NCA-Ti cathode material exhibited the best cycling stability of 93% up to 30 cycles.</P>
An investigation of local structures and EPR spectra for Cu2+ and VO2+ in biochar

Electron paramagnetic resonance (EPR) technology had been adopted to analyze the structures and properties of Cu2+ and VO2+ in Biochar (BG1 and BG2), but the local structural properties have not been clarified up to now. In present work, three possible structures (orthorhombically, rhombically and tetragonally elongated octahedra) are proposed for Cu2+ in BG1 and BG2 samples, and the former seems most reasonable in view of the best agreement between the theoretical EPR parameters based on the perturbation formulae and the experimental data. As for the VO2+ center, unlike the previous assignment of the tetragonally elongated [VO5]6− cluster based on the simple formulae of EPR parameters, presently proposed tetragonally compressed [VO6]8− cluster and the high order perturbation formulae of the EPR parameters yields good agreement with the observed values. This point is also supported by the d-d optical absorption 2B1g →2B2g for tetragonally compressed octahedral 3 d1 systems.

The stability of Bacillus licheniformis alpha-amylase (BLA) under acid condition was enhanced through direct evolution using the error-prone polymerase chain reaction. One beneficial mutation site, H281I, was obtained in BLA. The specific activity of H281I was 161/352 U/mg, which was 62.6/27.5% higher than that of the wild-type (WT) (99/276 U/mg) at pH 4.5/6.5 and 95°C. The pH optimum for H281I was decreased about 1 unit, whereas no significant changes of optimum temperature and thermostability were observed compared with the wild type (WT). The kcat/Km value of H281I was 1.7-/1.4-fold higher at pH 4.5/6.5, respectively, than that of WT. The structure model analysis indicated that the H281I mutation altered the predicted interaction between the amino acid residues at 281 and 273, thus creating a conducive local environment for substrate binding, as reflected by its decreased Km, and consequently increased the specific activity.
