Transcription activator-like effector nucleases (TALENs) are powerful tools for targeted genome editing in diverse cell types and organisms. However, the highly identical TALE repeat sequences make it challenging to assemble TALEs using conventional cloning approaches, and multiple repeats in one plasmid are easily catalyzed for homologous recombination in bacteria. Although the methods for TALE assembly are constantly improving, these methods are not convenient because of laborious assembly steps or large module libraries, limiting their broad utility. To overcome the barrier of multiple assembly steps, we report a one-step system for the con-venient and flexible assembly of a 180 TALE module library. This study is the first demonstration to ligate 9 mono-/dimer modules and one circular TALEN backbone vector in a one step process, generating 9.5 to 18.5 repeat sequences with an overall assembly rate higher than 50%. This system makes TALEN assembly much simpler than the conventional cloning of two DNA fragments because this strategy combines digestion and ligation into one step using circular vectors and different modules to avoid gel extraction. Therefore, this system provides a convenient tool for the application of TALEN-mediated genome editing in scientific studies and clinical trials.

An optimized space vector pulse width modulation (SVPWM) method with common mode voltage elimination and neutral point potential balancing is proposed for an open-end winding induction motor. The motor is fed from both of the ends with two neutral point clamped (NPC) three-level inverters. In order to eliminate the common mode voltage of the motor ends and balance the neutral point potential of the DC link, only zero common mode voltage vectors are used and a balancing control factor is gained from calculation in the strategy. In order to improve the harmonic characteristics of the output voltages and currents, the balancing control factor is regulated properly and the theoretical analysis is provided. Simulation and experimental results show that by adopting the proposed method, the common mode voltage can be completely eliminated, the neutral point potential can be accurately balanced and the harmonic performance for the output voltages and currents can be effectively improved.

This paper has completed the hardware design of STM32F103 system circuit, infrared code learning module, human body infrared sensor module, indoor temperature and humidity sensor module, completes the software design of learning air conditioning remote control code, indoor temperature and humidity sensor module, also completes the energy-saving controller hardware and software debugging. Finally, this paper takes split type air conditioning units in a conference room as the control object (including indoor lighting system) to complete the energy-saving controller in air conditioner electrical system design, installation and debugging, completes the air conditioning start-stop and infrared temperature control code learning test. The test results show that energy-saving controller has realized infrared automatic perception of human activity, machine start-stop code self-learning and temperature settings and other functions.
Study of the Optimal Film Structure by Using Conventional Elipsometry

The optimal film structure affected by the space effect has been studied for SiO2 film samples deposited onto Si wafers by using the e-beam evaporation method. The ellipsometric data were measured at different incidence angles and in the 300-800-nm wavelength range. In the range where the phase delay δ is close to π, the film thickness deduced from the conventional ellipsometry model varied with the phase delay. The optimal SiO2 film-thickness was achieved with a modified ellipsometric model, including the space effect. The results given in this work can generally be applied to other optical measurement methods used to study film structures.

We have investigated the probing beam transmitance due to the photon-induced anisotropy effect in the bacteriorhodopsin/polymer composite film(bR/PC film) by using the probing-pumping beam method under different laser wavelengths. By using a theoretical simulation, we give the characteristics of the birefringence Δn and the dichroism ΔD for various wavelengths of the probing and the pumping beams.

Creep Fracture Mechanism of a Single Crystal Nickel Base Alloy Under High Temperature and Low Stress

This paper reports the creep behavior of a nickel-based single crystal alloy and the creep fracture mechanism under hightemperature and low stress was discussed. The creep curves were analyzed with the Kelvin model. The retardation spectrasuggest that the involved atoms during creep need more relaxation time to achieve the viscous flow, nevertheless, the creepunder the stress of 120 MPa may be caused by the mismatch of dislocation motion and visco-plastic deformation. The fracturemorphologies of crept alloys indicate that the nickel base single crystal alloy presents micro-pore aggregation fracturemechanism under the condition of high temperature and low stress creep.

Background Keratin-associated protein (KAP), the structural protein molecule of hair fibers, plays a key role in determiningthe physical properties of hair. Studies of Krtap11-1 have focused only on its localization. Functional studies of Krtap11-1in hair follicle development have so far not been reported. Objective This study aimed to provide evidence for the role of Krtap11-1 in skin and hair development. Methods Full-length cloning and analysis of Krtap11-1 were conducted to ascertain its function. Overexpression vectors andinterference sequences were constructed and transfected into RAB-9 cells. Quantitative real-time polymerase chain reaction(qRT-PCR) was used to investigate the hair follicle developmental stage of Krtap11-1, the expression of different tissues,and the effects on other hair follicle development-related genes. Results The full length of cloned Krtap11-1 was 947 bp. Krtap11-1 was confirmed to be a hydrophilic protein localizedmostly in mitochondria. The greatest mRNA expression was observed in skin. Using a follicle synchronization model, itwas found that Krtap11-1 mRNA expression levels first increased then decreased over the passage of time, principally duringhair follicle catagen and telogen. Following the overexpression of Krtap11-1, mRNA expression levels of the WNT-2,KRT17, BMP-2, and TGF-β-1 genes increased, and LEF-1 decreased (P < 0.05), the converse after the corresponding useof si-RNA interference. Conclusions Krtap11-1 exerts a promoting effect. The results provide novel insight into the relationship between hair follicledevelopment and Krtap11-1 gene expression.
