        Analysis of the Genome Sequence of Strain GiC-126 of Gloeostereum incarnatum with Genetic Linkage Map

        Gloeostereum incarnatum has edible and medicinal value and was first cultivated and domesticated in China. We sequenced the G. incarnatum monokaryotic strain GiC-126 on an Illumina HiSeq X Ten system and obtained a 34.52-Mb genome assembly sequence that encoded 16,895 predicted genes. We combined the GiC-126 genome with the published genome of G. incarnatum strain CCMJ2665 to construct a genetic linkage map (GiC-126 genome) that had 10 linkage groups (LGs), and the 15 assembly sequences of CCMJ2665 were integrated into 8 LGs. We identified 1912 simple sequence repeat (SSR) loci and detected 700 genes containing 768 SSRs in the genome; 65 and 100 of them were annotated with gene ontology (GO) terms and KEGG pathways, respectively. Carbohydrate-active enzymes (CAZymes) were identified in 20 fungal genomes and annotated; among them, 144 CAZymes were annotated in the GiC-126 genome. The A mating-type locus (MAT-A) of G. incarnatum was located on scaffold885 at 38.9 cM of LG1 and was flanked by two homeodomain (HD1) genes, mip and beta-fg. Fourteen segregation distortion markers were detected in the genetic linkage map, all of which were skewed toward the parent GiC-126. They formed three segregation distortion regions (SDR1-SDR3), and 22 predictive genes were found in scaffold1920 where three segregation distortion markers were located in SDR1. In this study, we corrected and updated the genomic information of G. incarnatum. Our results will provide a theoretical basis for fine gene mapping, functional gene cloning, and genetic breeding the follow-up of G. incarnatum.

        Aging impairs insulin-stimulated glucose uptake in rat skeletalmuscle via suppressing AMPKα

        Insufficient intracellular fat oxidation is an important contributor to aging-related insulin resistance, while the precise mechanism underlying is unclear. AMP-activated protein kinase (AMPK) is an important regulator of intracellular fat oxidation and was evidenced to play a key role in high-glucose and high-fat induced glucose intolerance. In the present study, we investigated whether altered AMPK expression or activity was also involved in aging-related insulin resistance. Insulin sensitivity of rats’ skeletal muscles was evaluated using in-vitro glucose uptake assay. Activity of α subunit of AMPK (AMPKα ) was evaluated by measuring the phosphorylation of both AMPKα (P- AMPKα ) and acetyl-CoA carboxylase (P-ACC), while expression of AMPKα was assessed by determining the mRNA levels of AMPKα 1 and AMPKα 2, and protein contents of AMPKα . Compared with 4-month old rats, 24-month old rats exhibited obviously impaired insulin sensitivity. At the same time, AMPKα activity significantly decreased, while AMPKα expression did not alter during aging. Glucose transporter 4 expression also decreased in old rats. Compared with 24-month old rats, administration of the specific activator of AMPK, 5-aminoimidazole-4-carboxamide riboside (AICAR), significantly elevated AMPKα activity and GluT4 expression. Also, aging-related insulin resistance was significantly ameliorated by AICAR treatment. In conclusion, aging-related insulin resistance is associated with impaired AMPKα activity and could be ameliorated by AICAR, thus indicating a possible role of AMPK in aging-induced insulin resistance.

      • PKCδ-dependent Activation of the Ubiquitin Proteasome System is Responsible for High Glucose-induced Human Breast Cancer MCF-7 Cell Proliferation, Migration and Invasion

        Type 2 diabetes mellitus (T2DM) has contributed to advanced breast cancer development over the past decades. However, the mechanism underlying this contribution is poorly understood. In this study, we determined that high glucose enhanced proteasome activity was accompanied by enhanced proliferation, migration and invasion, as well as suppressed apoptosis, in human breast cancer MCF-7 cells. Proteasome inhibitor bortezomib (BZM) pretreatment mitigated high glucose-induced MCF-7 cell growth and invasion. Furthermore, high glucose increased protein kinase C delta ($PKC{\delta}$)-phosphorylation. Administration of the specific $PKC{\delta}$ inhibitor rottlerin attenuated high glucose-stimulated cancer cell growth and invasion. In addition, $PKC{\delta}$ inhibition by both rottlerin and $PKC{\delta}$ shRNA significantly suppressed high glucose-induced proteasome activity. Our results suggest that $PKC{\delta}$-dependent ubiquitin proteasome system activation plays an important role in high glucose-induced breast cancer cell growth and metastasis.

      • Effect of Diallyl Trisulfide on Human Ovarian Cancer SKOV-3/DDP Cell Apoptosis

        Aim: To investigate the effects of diallyl trisulfide (DT) on apoptosis of cisplatin (DDP)-resistant human epithelial ovarian cancer SKOV-3 cells (SKOV-3/DDP), and the role of p53 upregulated modulator of apoptosis (PUMA). Methods: SKOV-3/DDP cells were randomly divided into control, DT, DPP and DPP+DT groups, which were treated with DT or combined DT and DDP. All cells were incubated for 48 h. and apoptosis rates were assessed by flow cytometry. mRNA and protein expression of PUMA, Bax and Bcl-2 was determined by RT-PCR and Western blot assays, respectively. Results: Compared with control group, the apoptosis rates of SKOV-3/DDP cells in DT groups were obviously increased, with dose-dependence (P < 0.05), the mRNA and protein expressions of PUMA, Bax also being up-regulated (P < 0.05), while those of Bcl-2 were down-regulated (P < 0.05). Compared with DT groups, the apoptosis rate in the DDP+DT group was significantly increased (P < 0.05). After knockdown of PUMA with specific siRNA, the apoptosis rate of SKOV-3/DDP cells was obviously decreased (P < 0.05). Conclusion: DT can promote the apoptosis of SKOV-3/DDP cells with PUMA playing a critical role.

        Coalescence behavior of aqueous drops in water-in-oil emulsions under high-frequency pulsed AC fields

        High-frequency pulsed alternating currentfield (HFPACF) has attracted attentions owing to its excellentperformances of eliminating short circuit, saving energy and reducing environmental pollution. To studythe coalescence mechanism of aqueous drops in water-in-oil (W/O) emulsions under HFPACF, micro-behavior of dispersed drops for W/O emulsion was monitored by using a self-designed micro-visualmodel and coalescence mechanism was analyzed by discussing effects of pulse amplitude, pulsefrequency, pulse width ratio, demulsifier concentration, aqueous mass ratio of emulsion, and differentmodel oil emulsions. The results show that the dispersed drops coalesce under the action of electricfield,and the particle size of dispersed drops increases with the increase of pulse frequency and amplitude. When the pulse frequency or amplitude reach to a critical value, aqueous drops are driven to form waterstring or water channel between two electrodes which hinders further drops coalescence in other areas. However, with the increase of pulse width ratio, the size of dispersed drops increases slowly. Theoptimum pulse frequency and pulse amplitude are obtained at 3.9 kV and 4.21 kHz, respectively, theoptimal pulse width range is 54.9%-84.4%. When the concentration of demulsifier is 60 mg/L, thesynergistic demulsification effect achieves the highest performance. When the aqueous mass ratio ofemulsion reaches a high level (over 30%), which means the drops in emulsion have big size, thecoalescence performance under the same electricfield strength is great. Also, it is discovered that thedispersed drops in Yongping model oil emulsion have difficulty in forming water chains, since there aremuch asphaltenes and resins in the water-oil interface leading to high strength offilm. For Shuanghe oilemulsion drops are prevented from coalescing, because it contains a mass of paraffin, which can stabilizeemulsion at low temperature.

      • Preliminary Research on the Expression, Purification and Function of the Apoptotic Fusion Protein, Sival

        The objective of the present study was to investigate cloning, expression, and functions of the recombinant protein, Siva1. Siva1 gene was synthesized by RT-PCR from HCT116 cells. Plasmids were cleaved with the restriction endonuclease, BamH1/Sal1 and products were connected to pQE30, which underwent cleavage by BamH1/Sal1. The recombinant plasmid, pQE30-Siva1, was identified after digestion with restriction endonucleases followed by transformation into E. coli M15. Expression of Siva1 was induced by IPTG and identified by SDS-PAGE following purification with affinity chromatography. The results showed that size of Siva1 was 12 kDa, consistent with the molecular weight of the His-Siva1 fusion protein. Functional test demonstrated that Siva1 significantly inhibited the invasion and migration of HCT116 cells. It may thus find clinical application for control of cancers.

        Weighted sparsity-based denoising for extracting incipient fault in rolling bearing

        Given that the incipient fault is too weak for extraction, a novel approach that is based on sparse optimization is proposed for incipient fault diagnosis. The proposed optimization method consists of three steps: First, autocorrelation analysis is utilized to filter broadband random noise. Then, the weighted sparsity-based denoising method is proposed to extract periodic impulses. The prior knowledge that periodic impulses are sparse is used to constitute a penalty term; thus a novel weighted sparse optimization model is established. The majorization-minimization method is used to solve the optimization model. The high-pass filter in quadratic fidelity term is constructed by a Butterworth filter based on banded matrices, thus effectively improving computational efficiency. Lastly, the interval of periodic impulses, which corresponds to the fault frequency of rolling bearing, is obtained. Moreover, simulation and experimental results show that the proposed approach can successfully extract fault features from the signals of low signal to noise ratio.

        Influence of Nb on the hot corrosion behaviors of a Fe-Cr-Ni heat-resistant steel weld metal in the Na2SO4-NaCl molten salt at 700 ℃

        A multiple gas tungsten arc welding was performed on the fabrication of the Fe–Cr–Ni-based weld metal. Microstructuresand hot corrosion behaviors of the non-equilibrium weld metals with different Nb contents in 75 wt%Na2SO4–25 wt%NaClmolten salt at 700 °C have been investigated. A bilayer oxide film composed of an inner Cr2O3layer and an outer Fe2O3layer formed on the surface of the weld metals. The mass gain rate and the thickness of the surface oxide film were enlargedwith Nb addition, which could be attributed to the rapid inward diffusion of O through the cracks resulting from the Nboxide. Oxidation and sulfidation occur in the matrix, resulting in a large internal oxidation zone (IOZ) in the weld metal. Nb seemly promoted the sulfidation reaction due to a short-circuit diffusion of S through the interface of the Nb(C, N)and matrix and made the internal corrosion products to be distributed along the interdendritic zone of the weld metal. Anapparent Cr-dealloyed layer appears in the IOZ of the weld metal. Dealloying was the detrimental form of hot corrosion ofFe–Cr–Ni-based weld metal exposed to molten salts containing chloride salts. Alloying with 0.8 wt% of Nb further acceleratedthe dealloying and resulted in a profound attack.

        Synthesis and characterization of epoxy resin-based ion-imprinted polymer for selective removal of copper ions

        With the continuous development of the concept of green chemistry, more attention is being paid to the selective capture of copper (II) from wastewater and its recovery and reuse. Ion imprinting technology is widely used to remove heavy metals from aqueous solutions due to its high selectivity. In this work, Cu(II) imprinted polymer (Cu(II)-IIP) was prepared by stepwise polymerization using Cu(II) ions as the template ions, PEG-600 as a pore-forming agent, epoxy resin as the carrier, and polyethylene polyamine as curing agent and complexing agent. The influences of solution pH, reaction temperature, reaction time and initial solubility of metal ions on its adsorption ability were systematically examined. The studies showed that the maximum adsorption quantity was 91.58 mg/g when pH=5 and T=318 K. The adsorption for Cu(II) of the Cu(II)-IIP was considered to be the monolayer chemisorption and spontaneous endothermic process. In the presence of multiple coexisting ions, Cu(II)-IIP had high selectivity for Cu(II). After five adsorption-desorption cycles, high removal rate could still be obtained, which means that the material provides a feasible method for treating wastewater containing Cu(II) ions.

        Downregulation of cellular prion protein inhibited the proliferation and invasion and induced apoptosis of Marek’s disease virus-transformed avian T cells

        Cellular prion protein (PrPC) is ubiquitously expressed in the cytomembrane of a considerable number of eukaryotic cells. Although several studies have investigated the functions of PrPC in cell proliferation, cell apoptosis, and tumorigenesis of mammals, the correlated functions of chicken PrPC (chPrPC) remain unknown. In this study, stable chPrPC-downregulated Marek’s disease (MD) virus-transformed avian T cells (MSB1-SiRNA-3) were established by introducing short interfering RNA (SiRNA) targeting chicken prion protein genes. We found that downregulation of chPrPC inhibits proliferation, invasion, and migration, and induces G1 cell cycle phase arrest and apoptosis of MSB1-SiRNA-3 cells compared with Marek’s disease virus-transformed avian T cells (MSB1) and negative control cells. To the best of our knowledge, the present study provides the first evidence supporting the positive correlation between the expression level of chPrPC and the proliferation, migration, and invasion ability of MSB1 cells, but appears to protect MSB1 cells from apoptosis, which suggests it functions in the formation and development of MD tumors. This evidence may contribute to future research into the specific molecular mechanisms of chPrPC in the formation and development of MD tumors.
